Hesperidin is a flavonoid glycoside with proven therapeutic activities for various diseases, including cancer. However, its poor solubility and bioavailability render it only slightly absorbed, requiring a delivery system to reach its therapeutic target. Hesperidin loaded on gold nanoparticles (Hsp-AuNPs) was prepared by a chemical synthesis method. Various characterization techniques such as UV-VIS spectroscopy, FTIR, XRD, FESEM, TEM and EDX, Zeta potential analysis, particle size analysis, were used to confirm the synthesis of Hsp-AuNPs. The cytotoxic effect of Hsp-AuNPs on human breast cancer cell line (MDA-MB-231) was assessed using MTT and crystal violet assays. The results revealed significant decrease in proliferation and inhibition of growth of the treated cells when compared with human normal breast epithelial cell line (HBL-100). Determination of apoptosis by fluorescence microscope was also performed using acridine orange-propidium iodide dual staining assay. The in vivo study was designed to evaluate the toxicity of Hsp-AuNPs in mice. The levels of hepatic and kidney functionality markers were assessed. No significant statistical differences were found for the tested indicators. Histological images of liver, spleen, lung and kidney showed no apparent damages and histopathological abnormalities after treatment with Hsp-AuNPs. Hsp-AuNPs ameliorated the functional activity of macrophages against Ehrlich ascites tumor cells-bearing mice. The production of the pro-inflammatory cytokines was also assessed in bone marrow–derived macrophage cells treated with Hsp-AuNPs. The results obviously demonstrated that Hsp-AuNPs treatment significantly inhibited the secretion of IL-1β, IL-6 and TNF.

Herbicides are natural or synthetic chemicals used to control unwanted plants (weeds). To avoid the harmful effects of synthetic herbicides, considerable effort has been devoted to finding alternative products derived from natural sources. Essential oils (EOs) from aromatic plants are auspicious source of bioherbicides. This review discusses phytotoxic EOs and their chemical compositions as reported from 1972 to 2020. Using chemometric analysis, we attempt to build a structure-activity relationship between phytotoxicity and EO chemical composition. Data analysis reveals that oxygenated terpenes, and mono- and sesquiterpenes, in particular, play principal roles in the phytotoxicity of EOs. Pinene, 1,8 cineole, linalool, and carvacrol are the most effective monoterpenes, with significant phytotoxicity evident in the EOs of many plants. Caryophyllene and its derivatives, including germacrene, spathulenol, and hexahydrofarnesyl acetone, are the most effective sesquiterpenes. EOs rich in iridoids (non-terpene compounds) also exhibit allelopathic activity. Further studies are recommended to evaluate the phytotoxic activity of these compounds in pure forms, determine their activity in the field, evaluate their safety, and assess their modes of action.

Green synthesis of silver nanoparticles (AgNPs) using different plant parts has shown a great potential in medicinal and industrial applications. In this study, AgNPs were in vitro green synthesized using A. graecorum, and its antifungal and antitumoractivities were investigated. Scanning electron microscopy (SEM) image result indicated spherical shape of AgNPs with a size range of 22‐36 nm indicated by using Image J program. The functional groups indicated by Fourier‐transform infrared spectroscopy (FTIR) represented the groups involved in the reduction of silver ion into nanoparticles. Alhagi graecorum AgNPs inhibited MCF‐7 breast cancer cell line growth in increased concentration depend manner, significant differences shown at 50, 100, and 150 μg/ml concentrations compared to the control. Strong antifungal activity against Candida species (C. albicans., C. glabrata, C. parapsilosis, C. tropicales, and C. krusei) was observed and the inhibition zone range from 14‐22 mm at a concentration of 0.01 mmol/ml and from 17‐27 mm at a concentration of 0.02 mmol/ml. Based on our findings, it is concluded that synthesized silver nanoparticles from A. graecorum can be used as a potential antitumor and antifungal agent for various therapeutical applications.

Annona muricata is one of the most important traditional medicinal plants which contains numerous chemicals that exhibit various pharmacological properties. In this study, silver nanoparticles were prepared using A. muricata peel extract as a reducing agent and the effect was enhanced through A. muricata like pharmaceutical activity. AgNPs formation was confirmed by color changes, UV-visible spectroscopy, SEM, DLS, and XRD. The anti-proliferative activity of AgNPs against THP-1, AMJ-13, and HBL cell lines was studied. Apoptotic markers were tested using AO/EtBr staining assay, cell cycle phases using flowcytometry, and the expression of P53. Autophagy takes an essential part in controlling inflammasome activation by primary bone marrow-derived macrophages (BMDMs). We report novel functions for AgNPs-affected autophagy, represented by the control of the release of IL-1β, caspase-1, adaptor protein apoptosis-associated speck-like protein containing a CARD (ASC), and NLRP3 in BMDMs following treatment with LPS+ATP. The current study revealed that the AgNPs inhibited THP-1 and AMJ-13 cell proliferation. Meanwhile, the AgNPs significantly increased autophagy and reduced IL-1b and NLRP3 levels in both in vivo and in vitro models. The secretion of IL-1β was reduced whereas the degradation of NLRP3 inflammasome was enhanced. These findings propose that AgNPs apply an anti-proliferative activity against THP-1 and AMJ-13 cells through the stimulation of apoptosis via mitochondrial damage and induction of p53 protein pathway. In addition, AgNP-induced autophagy reduced the levels of IL-1β and NLRP3 inflammasome activation. This indicated that the AgNPs augment autophagy controlled by the IL-1β pathway via two different novel mechanisms. The first one is regulating activation of the IL-1 β, caspase-1, and ASC, while the second is NLRP3 targeting for lysosomal degradation. Overall, this study suggests that AgNPs could be a potent therapy for various types of cancer and an alternative treatment for preventing inflammation via enhancing autophagy.
