The potential of microalgae biofuel has not been realized because of low productivity and high costs associated with the current cultivation systems. In this paper, an attached cultivation method was introduced, in which microalgae cells grew on the surface of vertical artificial supporting material to form algal film. Multiple of the algal films were assembled in an array fashion to dilute solar irradiation to facilitate high photosynthetic efficiency. Results showed that a broad range of microalgae species can grow with this attached method. A biomass productivity of 50-80 g m(-2) d(-1) was obtained outdoors for Scenedesmus obliquus, corresponding to the photosynthetic efficiency of 5.2-8.3% (total solar radiation). This attached method also offers lots of possible advantages over traditional open ponds, such as on water saving, harvesting, contamination controlling and scale-up. The attached cultivation represents a promising technology for economically viable production of microalgae biofuels.

The ‘attached cultivation’ technique for microalgae production, combining the immobilized biofilm technology with proper light dilution strategies, has shown improved biomass production and photosynthetic efficiency over conventional open-pond suspended cultures. However, how light is transferred and distributed inside the biofilm has not been clearly defined yet. In this research, the growth, photosynthetic oxygen evolution, and specific growth rate for microalgal cells in both open-pond and attached cultivation were studied to determine the effective light penetration at different phases of the cultivation. As a result, the light conditions inside the culture broth as well as the biofilm were revealed for the first time. Results showed that outdoor, in a conventional 20-cm deep open pond, all of the algal cells were fully illuminated in the first 3 days of cultivation. As the biomass concentration increased from day 4 to day 10, the light could only effectively penetrate 45.5% of the open-pond depth, and then effective light penetration gradually decreased to 31.1% at day 31, when the biomass density reached a maximum value of 0.45 g L−1 or 90 g m−2. In the attached cultivation system, under nitrogen-replete condition, almost 100% of the immobilized algal cells inside the biofilm were effectively illuminated from day 0 through day 10 when the biomass density increased from 8.8 g m−2 to 107.6 g m−2. Higher light penetration efficiency might be the reason why, using attached cultivation, observed values for photosynthetic efficiency were higher than those recorded in conventional open-pond suspended cultures.

Recently, Tribonema sp., a kind of filamentous microalgae, has been studied for biofuel production due to its accumulation of triacylglycerols. However, the polysaccharides of Tribonema sp. and their biological activities have rarely been reported. In this paper, we extracted sulfated polysaccharides from Tribonema sp. (TSP), characterized their chemical composition and structure, and determined their immunostimulation and anticancer activities on RAW264.7 macrophage cells and HepG2 cells. The results showed that TSP is a sulfated polysaccharide with a Mw of 197 kDa. TSP is a heteropolysaccharide that is composed mainly of galactose. It showed significant immune-modulatory activity by stimulating macrophage cells, such as upregulating interleukin 6 (IL-6), interleukin 10 (IL-10), and tumor necrosis factor α (TNF-α). In addition, TSP also showed significant dose-dependent anticancer activity (with an inhibition rate of up to 66.8% at 250 µg/mL) on HepG2 cells as determined by the 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The cycle analysis indicated that the anticancer activity of TSP is mainly the result of induced cell apoptosis rather than affecting the cell cycle and mitosis of HepG2 cells. These findings suggest that TSP might have potential as an anticancer resource, but further research is needed, especially in vivo experiments, to explore the anticancer mechanism of TSP.

To improve cultivation efficiency for microalgae Spirulina platensis is related to increase its potential use as food source and as an effective alternative for CO2 fixation. The present work attempted to establish a technique, namely attached cultivation, for S. platensis. Laboratory experiments were made firstly to investigate optimal conditions on attached cultivation. The optimal conditions were found: 25 g m(-2) for initial inoculum density using electrostatic flocking cloth as substrata, light intensity lower than 200 μmol m(-2) s(-1), CO2 enriched air flow (0.5%) at a superficial aeration rate of 0.0056 m s(-1) in a NaHCO3-free Zarrouk medium. An outdoor attached cultivation bench-scale bioreactor was built and a 10d culture of S. platensis was carried out with daily harvesting. A high footprint areal biomass productivity of 60 g m(-2) d(-1) was obtained. The nutrition of S. platensis with attached cultivation is identical to that with conventional liquid cultivation.
