It is estimated that 90% of hyperuricemia cases are attributed to the inability to excrete uric acid (UA). The two main organs in charge of excreting UA are the kidney (70%) and intestine (30%). Previous studies have reported that punicalagin (PU) could protect against kidney and intestinal damages, which makes it a potential candidate for alleviating hyperuricemia. However, the effects and deeper action mechanisms of PU for managing hyperuricemia are still unknown. To investigate the effect and action mechanisms of PU for ameliorating hyperuricemia. The effects and action mechanisms of PU on hyperuricemia were assessed using a hyperuricemia mice model. Phenotypic parameters, metabolomics analysis, and 16S rRNA sequencing were applied to explore the effect and fundamental action mechanisms inside the kidney and intestine of PU for improving hyperuricemia. PU administration significantly decreased elevated serum uric acid (SUA) levels in hyperuricemia mice, and effectively alleviated the kidney and intestinal damage caused by hyperuricemia. In the kidney, PU down-regulated the expression of UA resorption protein URAT1 and GLUT9, while up-regulating the expression of UA excretion protein ABCG2 and OAT1 as mediated via the activation of MAKP/NF-κB in hyperuricemia mice. Additionally, PU attenuated renal glycometabolism disorder, which contributed to improving kidney dysfunction and inflammation. Similarly, PU increased UA excretion protein expression via inhibiting MAKP/NF-κB activation in the intestine of hyperuricemia mice. Furthermore, PU restored gut microbiota dysbiosis in hyperuricemia mice. This research revealed the ameliorating impacts of PU on hyperuricemia by restoring kidney and intestine damage in hyperuricemia mice, and to be considered for the development of nutraceuticals used as UA-lowering agent.

Berberine is an effective antimicrobial and antidiabetic alkaloid, primarily extracted from divergent botanical lineages, specifically Coptis (Ranunculales, early-diverging eudicot) and Phellodendron (Sapindales, core eudicot). In comparison with its known pathway in Coptis species, its biosynthesis in Phellodendron species remains elusive. Using chromosome-level genome assembly, coexpression matrix, and biochemical assays, we identified six key steps in berberine biosynthesis from Phellodendron amurense , including methylation, hydroxylation, and berberine bridge formation. Notably, we discovered a specific class of O -methyltransferases (NOMT) responsible for N -methylation. Structural analysis and mutagenesis of PaNOMT9 revealed its unique substrate-binding conformation. In addition, unlike the classical FAD-dependent berberine bridge formation in Ranunculales, Phellodendron uses a NAD(P)H-dependent monooxygenase (PaCYP71BG29) for berberine bridge formation, originating from the neofunctionalization of tryptamine 5-hydroxylase. Together, these findings reveal the convergence of berberine biosynthesis between Coptis and Phellodendron and signify the role of the convergent evolution in plant specialized metabolisms.

Alkali and drought stresses are increasing severe environmental problems throughout the world, especially in the Songnen grassland of northern China. Leymus chinensis is the dominant grass species in the Songnen grassland of northern China and the most promising species for grassland restoration. Arbuscular mycorrhizal fungi (AMF) can colonize 80% of vascular plants, which can enhance the growth of host plants and provide extrinsic protection against abiotic stresses. However, little is known about the interaction effect of alkali and drought stresses on plant-AM symbionts. Here, seedlings of Leymus chinensis inoculated with or without mycorrhizae were cultivated in soil with 0, 100 or 200 mM NaHCO3 under 0, 5 or 10% (w/v) PEG treatment, and the changes in growth, osmotic adjustment substances and ions were measured. The results showed that the interaction of alkali and drought stresses caused greater seedling growth inhibition than either single alkali or drought stress due to ion toxicity and oxidative damage. Mycorrhizae could alleviate the growth inhibition of seedlings under alkali or drought stress. The interaction of alkali and drought stresses did not affect the alleviating effect of mycorrhizae on seedling growth but improved the osmotic regulation ability and ionic balance of the seedlings. Our results clearly show different effects of the interaction of alkali and drought stresses versus a single stress (alkali or drought) on plant development and provide new insights into the positive effect of arbuscular mycorrhizal fungi on host plants under such stress conditions.

The increasing impact of global climate change has resulted in adversity stresses, like salt and drought, gradually becoming the main factors that limit crop growth. Hemp, which contains numerous medicinal active components and multiple bioactive functions, is widely used in the agricultural, industrial, and medical fields, hence promoting the rapid development of related industries. Arbuscular mycorrhizal fungi (AMF) can establish a symbiotic relationship with 80% of vascular plants. This symbiosis promotes host plant growth, regulates plant physiology and biochemistry, facilitates secondary metabolite synthesis, and enhances resistance to abiotic stresses. However, the effects of salt stress, drought stress, and AMF interaction in hemp are not well understood. In this study, to investigate this, we performed a study where we cultured hemp that was either inoculated or uninoculated with Funneliformis mosseae and determined changes in effective colonization rate, growth, soluble substances, photosynthesis, fluorescence, ions, and secondary metabolites by cultivating hemp under different concentrations of NaCl (0 mM, 100 mM, and 200 mM) and different soil moisture content (45%, 25%, and 15%). The results showed that salt, drought stress, or salt–drought interaction stress all inhibited colonization rate after stress, plant growth, mainly due to ion toxicity and oxidative damage. Inoculation with F. mosseae effectively alleviated plant growth inhibition under 100 mM NaCl salt stress, drought stress, and salt–drought interaction stress conditions. It also improved osmoregulation, photosynthetic properties, fluorescence properties, and ion homeostasis, and promoted the accumulation of secondary metabolites. However, under 200 mM NaCl salt stress conditions, inoculation with F. mosseae negatively affected plant physiology, biochemistry, and secondary metabolite synthesis, although it did alleviate growth inhibition. The results demonstrate that there are different effects of salt–drought interaction stress versus single stress (salt or drought stress) on plant growth physiology. In addition, we provide new insights about the positive effects of AMF on host plants under such stress conditions and the effects of AMF on plants under high salt stress.

Compared with other crops, pennycress (Thlaspi arvense L.) is a niche emerging oil crop. In recent years, research on pennycress has been increasingly reflected in various directions. Pennycress belongs to the Brassicaceae family and was introduced from Eurasia to North America. It has been found worldwide as a cultivated plant and weed. In this paper, we review the advantages of pennycress as a supplementary model plant of Arabidopsis thaliana, oil and protein extraction technology, seed composition analysis based on metabolomics, germplasm resource development, growth, and ecological impact research, abiotic stress, fatty acid extraction optimization strategy, and other aspects of studies over recent years. The main research directions proposed for the future are as follows: (1) assemble the genome of pennycress to complete its entire genome data, (2) optimize the extraction process of pennycress as biodiesel, (3) analyze the molecular mechanism of the fatty acid synthesis pathway in pennycress, and (4) the functions of key genes corresponding to various adversity conditions of pennycress.

Cd stress can cause severe damage to the chloroplast structure and plant function. The chloroplast stay-green gene (SGR2) is senescence-inducible and has several important functions such as regulating senescence, responding to abiotic stress, and regulating seed and fruit ripening. The SlSGR2 gene was cloned from the tomato “Hongluocheng” (full-length of 819 bp encoding 272 amino acids). Subcellular localization prediction and laser confocal observation revealed that the SlSGR2 protein was localized to the chloroplast of A. thaliana protoplasts. A qRT-PCR analysis suggested that the SlSGR2 gene was upregulated by different Cd concentrations and that the gene expression was significantly upregulated in tomato roots and leaves under 50 μM CdCl2 treatment. The pCAMBIA2300-SlSGR2 overexpression constructs were introduced into wild-type Arabidopsis plants via Agrobacterium-mediated transformation. Homozygous mutant lines of sgr2 were identified using the three-primer method. The growth of SlSGR2-overexpressed lines under Cd stress was significantly better than that of WT and sgr2. The chlorophyll, SOD, POD, and CAT contents of OE4-1 and OE4-2 were significantly higher than those of WT and sgr2 while the MDA content was significantly lower than other lines under cadmium stress. This study showed that the SlSGR2 gene is involved in Cd-induced plant senescence and is a negative regulator of plant chlorophyll degradation.
