A general chemo-enzymatic approach to synthesize both enantioenriched trans-3-alkoxyamino-4-oxy-2-piperidones, which are important scaffold for various naturally occurring alkaloids, is reported. To this end, a selective transition-metal-free dual C−H oxidation of piperidines mediated by the TEMPO oxoammonium cation (TEMPO+) was used, followed by enzymatic resolution of the corresponding alkoxyamino-2-piperidones with Candida antarctica lipase (CAL-B), to yield the title compounds in high enantiomeric excess (ee). The absolute configuration of both enantioenriched compounds was determined using chemical correlation and circular dichroism (CD) spectroscopy. The former method highlights the oxidative ring contraction of the trans-alkoxyamine-2-piperidone ring into its corresponding 2-pyrrolidinone.
In recent years, metallodrugs have been playing an important role, showing to be more efficient in the treatment of several diseases, such as cancer. Indeed, it is important to synthesize novel molecules to be used as more effective agents against cancer. In the present paper, the synthesis of two new molecules belonging to Casiopeínas® is reported. These compounds present a β2-aminoacidate derivative as the secondary ligand. The novel metal complexes were characterized by high-resolution mass spectrometry, FT-IR, UV-Vis, EPR, effective magnetic moment and cyclic voltammetry measurements, and single crystal X-ray diffraction analysis. Furthermore, these compounds were evaluated in vitro against the cancer lines MCF-7 (breast cancer) and A549 (lung cancer).
Singlet oxygen ene reactions produce 2-(tert-butyl)-4a-hydroperoxy-3-methyl-2,4a, 5,6,7,8-hexahydroquinazolin-4(3H)-one quantitatively during diffusion crystallization of 2-(tert-butyl)-3-methyl-2,3,5,6,7,8-hexahydroquinazolin-4(1H)-one in n-hexane/CH2Cl2 solvent mixture. To confirm this photo-oxidation, a 1H-NMR study in CDCl3 was performed with exposure to ambient conditions (light and oxygen), with neither additional reactants nor catalysts. A theoretical study at the B3LyP/6311++G** level using the QST2 method of locating transition states suggests a two-step mechanism where the intermediate, which unexpectedly did not come from the peroxide intermediate, has a low activation energy.
