This report presents the outcome of the joint work of PhD students and senior researchers working with DNA-based biodiversity assessment approaches with the goal to facilitate others the access to definitions and explanations about novel DNA-based methods. The work was performed during a PhD course (SLU PNS0169) at the Swedish University of Agricultural Sciences (SLU) in Uppsala, Sweden. The course was co-organized by the EU COST research network DNAqua-Net and the SLU Research Schools Focus on Soils and Water (FoSW) and Ecology - basics and applications. DNAqua-Net (COST Action CA15219, 2016-2020) is a network connecting researchers, water managers, politicians and other stakeholders with the aim to develop new genetic tools for bioassessment of aquatic ecosystems in Europe and beyond. The PhD course offered a comprehensive overview of the paradigm shift from traditional morphology-based species identification to novel identification approaches based on molecular markers. We covered the use of molecular tools in both basic research and applied use with a focus on aquatic ecosystem assessment, from species collection to the use of diversity in environmental legislation. The focus of the course was on DNA (meta)barcoding and aquatic organisms. The knowledge gained was shared with the general public by creating Wikipedia pages and through this collaborative Open Access publication, co-authored by all course participants.

Effective identification of species using short DNA fragments (DNA barcoding and DNA metabarcoding) requires reliable sequence reference libraries of known taxa. Both taxonomically comprehensive coverage and content quality are important for sufficient accuracy. For aquatic ecosystems in Europe, reliable barcode reference libraries are particularly important if molecular identification tools are to be implemented in biomonitoring and reports in the context of the EU Water Framework Directive (WFD) and the Marine Strategy Framework Directive (MSFD). We analysed gaps in the two most important reference databases, Barcode of Life Data Systems (BOLD) and NCBI GenBank, with a focus on the taxa most frequently used in WFD and MSFD. Our analyses show that coverage varies strongly among taxonomic groups, and among geographic regions. In general, groups that were actively targeted in barcode projects (e.g. fish, true bugs, caddisflies and vascular plants) are well represented in the barcode libraries, while others have fewer records (e.g. marine molluscs, ascidians, and freshwater diatoms). We also found that species monitored in several countries often are represented by barcodes in reference libraries, while species monitored in a single country frequently lack sequence records. A large proportion of species (up to 50%) in several taxonomic groups are only represented by private data in BOLD. Our results have implications for the future strategy to fill existing gaps in barcode libraries, especially if DNA metabarcoding is to be used in the monitoring of European aquatic biota under the WFD and MSFD. For example, missing species relevant to monitoring in multiple countries should be prioritized for future collaborative programs. We also discuss why a strategy for quality control and quality assurance of barcode reference libraries is needed and recommend future steps to ensure full utilisation of metabarcoding in aquatic biomonitoring.

Most ribbon worms (phylum: Nemertea) are found in marine environments, where they act as predators and scavengers. They are characterized by an eversible proboscis that isused to hunt for prey and thick mucus covering their skin. Both proboscis and epidermal mucus mediate toxicity to predators and preys. Research into the chemical nature of the substances that render toxicity has not been extensive, but it has nevertheless led to the identification of several compounds of potential medicinal use or for application in biotechnology. This review provides a complete account of the current status of research into nemertean toxins.

Research Infrastructures (RIs) are facilities, resources and services used by the scientific community to conduct research and foster innovation. LifeWatch ERIC has developed various virtual research environments, which include many virtual laboratories (vLabs) offering high computational capacity and comprehensive collaborative platforms that supporting the needs of digital biodiversity science. Over its 250 years of history, the taxonomic research community has developed a system for describing, classifying and naming taxa across multiple levels. For the marine biota, taxonomic information is organized and made publicly available through the World Register of Marine Species (WoRMS) that records more than 250,000 described valid species. Although scientists tend to assign an equal status (in terms of contribution to overall diversity) to each taxon used in taxonomy, biogeography, ecology and biodiversity, the question “are all taxa equal?” has never been tested at a global scale. We present evidence that this question can be addressed by applying relatedness indices (Taxonomic Distinctness) over the entire WoRMS metazoan tree. The RvLab, developed by the LifeWatchGreece RI, operating on a high-performance computer cluster, has been used to meet the high computational demands required for such an analysis at a global scale.

Research Infrastructures (RIs) are facilities, resources and services used by the scientific community to conduct research and foster innovation. LifeWatch ERIC has developed various virtual research environments, which include many virtual laboratories (vLabs) offering high computational capacity and comprehensive collaborative platforms that supporting the needs of digital biodiversity science. Over its 250 years of history, the taxonomic research community has developed a system for describing, classifying and naming taxa across multiple levels. For the marine biota, taxonomic information is organized and made publicly available through the World Register of Marine Species (WoRMS) that records more than 250,000 described valid species. Although scientists tend to assign an equal status (in terms of contribution to overall diversity) to each taxon used in taxonomy, biogeography, ecology and biodiversity, the question “are all taxa equal?” has never been tested at a global scale. We present evidence that this question can be addressed by applying relatedness indices (Taxonomic Distinctness) over the entire WoRMS metazoan tree. The RvLab, developed by the LifeWatchGreece RI, operating on a high-performance computer cluster, has been used to meet the high computational demands required for such an analysis at a global scale.

We test previous claims that the bacteria Vibrio alginolyticus produces tetrodotoxin (TTX) when living in symbiosis with the nemertean Lineus longissimus by a setup with bacteria cultivation for TTX production. Toxicity experiments on the shore crab, Carcinus maenas, demonstrated the presence of a paralytic toxin, but evidence from LC-MS and electrophysiological measurements of voltage-gated sodium channel–dependent nerve conductance in male Wistar rat tissue showed conclusively that this effect did not originate from TTX. However, a compound of similar molecular weight was found, albeit apparently non-toxic, and with different LC retention time and MS/MS fragmentation pattern than those of TTX. We conclude that C. maenas paralysis and death likely emanate from a compound <5 kDa, and via a different mechanism of action than that of TTX. The similarity in mass between TTX and the Vibrio-produced low-molecular-weight, non-toxic compound invokes that thorough analysis is required when assessing TTX production. Based on our findings, we suggest that re-examination of some published claims of TTX production may be warranted.

In recent years the Norwegian Taxonomy Initiative started thorough investigations of poorly known organism groups. In this context, several marine inventories have rendered a number of marine invertebrate species new to science. Within the phylum Nemertea (ribbon worms) a characteristic hoplonemertean was encountered on two different occasions. We describe the new species Amphiporus rectangulus sp. nov. with a combination of histology and DNA data (COI). For the morphological description we use a previously proposed character matrix and, in a context given by the results, also provide a brief discussion on benefits and drawbacks with both methods. We argue that for small animals with soft bodies external characters can be more informative than hitherto expected.

Vulcanonemertes rangitotoensis gen. et sp. nov. (Hoplonemertea: Monostilifera) is described and illustrated. Major morphological features of the new taxon include an anteriorly divided body wall longitudinal musculature, no pre-cerebral septum, cephalic glands which reach far back behind the brain, and accessory lateral nerves which extend the full length of the body.
