Rye is one of the most commonly used sources of elite genes in wheat improvement programs. Due to the high collinearity of the genomes of both cereal species, it is possible to obtain interspecific chromosomal translocations and substitution lines. Rye chromatin is used to transfer numerous genes for resistance to biotic and abiotic stresses into the wheat genome. Introgression has also resulted in improved agronomic traits. However, despite the numerous advantages, the transfer of large fragments or whole chromosomes has been quite often accompanied by a decrease in end-use quality. This paper presents an overview of the benefits and drawbacks of using rye as a source of variability in wheat breeding.

Efficient in vitro regeneration remains a major constraint in the genetic transformation, genome editing, and molecular breeding of wheat (Triticum aestivum L.), largely due to strong genotype-dependent recalcitrance and limited activation of developmental programs required for somatic embryogenesis. Plant regeneration relies on extensive transcriptional reprogramming and epigenetic remodeling orchestrated by morphogenic regulators that modulate meristem identity, as well as cellular pluri- and totipotency. In this review, we synthesize current molecular knowledge on key transcription factors (BBM, WUS/WUS2, GRF-GIF, WOX, LAX1, SERK, WIND1/ERF115) and signaling peptides (CLE/CLV-WUS module, phytosulfokine/PSK) that regulate embryogenic competence in monocot cereals, with emphasis on their orthologs and functional relevance in wheat. We highlight how controlled expression of these morphogenic genes, promoter engineering, and transient or excisable induction systems can significantly enhance regeneration capacity, reduce chimerism in CRISPR-Cas-edited plants, and facilitate genotype-independent transformation. We also discuss epigenetic and metabolic constraints underlying wheat recalcitrance and their potential modulation to improve culture responsiveness. By integrating evidence from wheat, rice, maize, and barley, we outline conserved gene-regulatory networks that reinitiate totipotency and propose strategies to accelerate doubled haploid production and speed-breeding pipelines. Collectively, morphogenic factors emerge as central molecular tools for overcoming regeneration bottlenecks and enabling next-generation wheat improvement. The objective of this review is to synthesize and critically evaluate current molecular knowledge on morphogenic regulators controlling in vitro regeneration in wheat (Triticum aestivum L.), with particular emphasis on their roles in genetic transformation and genome editing.

Sand oat (Avena strigosa Schreb.), one of the four cultivated species of the genus Avena, could be considered as another alternative crop. In gene banks 865 germplasm samples of this species have been preserved that have not been thoroughly investigated so far. The results of phenotyping (36 traits), isoenzymatic (12 systems) and genetic (8 pairs of Sequence-related amplified polymorphism markers) variation were used to obtain the complete description of 56 accessions diversity originated from different parts of world. Breeded and weedy forms represented similar pool of morphological traits that indicated a short-term and extensive breeding process, albeit all accessions which we classified as cultivated were characterized by better grain and green mass parameters compared to the weedy ones. Isoenzymes showed relationships with geographical origin, which was not possible to detect by SRAP markers. There was no similarity between morphological and biochemical results. The polymorphism level of SRAP markers was lower than indicated by the available literature data for other species, however it may result from the analysis of pooled samples of accessions with a high internal variability. The extensive type of breeding and its relatively short duration was also reflected in the population structure results. Joint analysis revealed that a secondary centre of diversity is being created in South America and that it has its genealogy from the Iberian Peninsula. Despite the relatively large representation of this species is in various gene banks, it is highly probable that the vast majority of stored worldwide accessions are duplicates, and the protected gene pool is relatively narrow. Sand oat meets all the requirements for an alternative crop species, but further studies are needed to identify the genotypes/populations with the most favourable distribution of utility and quality parameters.

The extensive genetic diversity of crop landraces deposited in gene banks is an important part of future breeding programs. However, in order to exploit this potential, it is necessary both to preserve germplasm and to collect and share a wide range of data on its characteristics and evaluation. The main objective of the presented paper was to evaluate the diversity of spring barley landraces originating from Poland. Historical data from standard field evaluation of agronomic traits, genetic data obtained from Inter Simple Sequence Repeat (ISSR) analysis and grain morphometric data were used. The analysis showed that groups of hulled and hulls barley accessions were substantially separate. Genetic and grain morphological variation did not show any relationship with eco-geographical conditions in the place of origin. There was also no clear division into forms with two- and six-row ears. The combination of the results of various types of analyzes describing this collection makes us believe that its chances of being used in research and breeding will increase.

This research is about the profiling of barley (Hordeum vulgare L.), wheat (Triticum aestivum L.), and rye (Secale cereale L.) FPG and OGG1 genes during grain germination. During seed germination, reactive oxygen species accumulate, which leads to DNA damage. In the base excision repair (BER) system, the enzymes formamidopyrimidine DNA glycosylase (FPG) and 8-oxoguanine DNA glycosylase (OGG1), among others, are responsible for repairing such damage. We decided to check how the expression of genes encoding these two enzymes changes in germinating grains. Spring varieties of barley, wheat, and rye from the previous growing season were used in the study. Expression level changes were checked using Real-Time PCR. After analyzing the obtained results, the maximum expression levels of FPG and OGG1 genes during germination were determined for barley, wheat, and rye. The results of the study show differences in expression levels specific to each species. The highest expression was observed at different time points for each of them. There were no differences in the highest expression for FPG and OGG1 within one species. In conclusion, the research provides information on how the level of FPG and OGG1 gene expression changes during the germination process in cereals. This is the first study looking at the expression levels of these two genes in cereals.

Plants have developed a number of mechanisms that are responsible for repairing the oxidative damage caused by reactive oxygen species. The base excision repair system is the main repair system for removing such changes. Herein, 8-oxoguanine DNA glycosylase and formamidopyrimidine DNA glycosylase are the two enzymes of importance for removing 8-oxoG. Researchers have confirmed that FPG and OGG1 levels increase during seed imbibition. Indeed, many researchers note the relationship between the accumulation of oxidative changes in the cell and the activity of OGG1 and FPG and the aging of seeds. This short review aims at presenting the published data on the operation of the base excision repair system in the process of repairing oxidative damage affecting seed aging.
