The characterization of garlic germplasm improves its utility, despite the fact that garlic hasn't been used much in the past. Garlic has an untapped genetic pool of immense economic and medicinal value in India. Hence, using heuristic core collection approach, a core set of 46 accessions were selected from 625 Indian garlic accessions based on 13 quantitative and five qualitative traits. The statistical measures (CV per cent, CR per cent, VR per cent) were used to sort the core set using Shannon-Wiener diversity index and the Nei diversity index. In addition, the variation within the core set was tested for 18 agro-morphological and six biochemical characteristics (allicin, phenol content, pyruvic acid, protein, allyl methyl thiosulfinate (AMTHS), and methyl allyl thiosulfinate (MATHS)). Further study of the core set's molecular diversity was performed using sequence related amplified polymorphism (SRAP) markers, which revealed a wide range of diversity among the core set's accessions, with an average polymorphism efficiency (PE) of 80.59 percent, polymorphism information content (PIC) of 0.29, effective multiplex ratio (EMR) of 3.51, and marker index (MI) of 0.99. The findings of this study will be useful in identifying high-yielding, elite garlic germplasm lines with the trait of interest. Since this core set is indicative of total germplasm, these selected breeding lines will be used for genetic improvement of garlic in the future.
Crop production is directly associated with the improvement of germplasm, which is mainly reliant on genetic diversity. Diversity among various genotypes has been investigated employing a variety of statistical approaches. The most widely utilized of these methods for determining the genetic overlap of genotypes is multivariate. In the present investigation, a total of 27 onion genotypes/advanced lines/varieties comprising high and low total soluble solids (TSS) white onion lines along with red varieties were evaluated at the ICAR-Directorate of Onion and Garlic Research station. Data were recorded on seven biochemical parameters. In multivariate analysis, genotypes were clustered into three major groups: the first group comprised thirteen genotypes with high TSS; however, the second group (eight) comprised low TSS white onion genotypes, and in the third group (six), mostly red varieties were clubbed together. The analysis primarily focused on the trait TSS; it was significantly associated with the antioxidant assay 2, 2-diphenyl-1-picrylhydrazyl(DPPH) AE, while total sugar content was positively associated with the antioxidant assay 2, 2-azinobis (3-ethylbenzothiazoline- 6-sulfonic acid)ABTS. In principal component analysis (PCA), the first three principal components (PCs) with >1 Eigen value contributed 71.36% of the variability among genotypes. Characters with a maximum value at PC1 were total soluble solids (0.48), antioxidant capacity AE DPPH (0.46), and total sugar content (0.49). PC2 comprises total phenolic content (0.62) and total flavonoid content (0.62); for PC3, the major contributors were thiosulfinate content (0.82) and antioxidant capacity ABTS (0.32). From the findings of the present study, the best-performing high TSS lines can be used for advancement through strongly correlated traits using breeding strategies. These sorted high TSS lines (W-103, W-107, and W-123) (>22 °Brix.), high-sugar-content genotypes (W-108, W-111, and W-308), and W-361, which recorded high thiosulfinate content, can be advanced or used as parental material for the development of processing-suitable onion varieties.

Acquisition and germination of seeds are the most desired targets for the improvement of vegetatively propagated crops. In the present study, we developed a potential embryo germination protocol for the Red Globe grape cultivar having a low seed germination rate. Three grape berries at different developmental stages, viz. 50, 60 and 70 days after flowering (DAF), were selected for in-vitro embryo germination. Three growth media, namely Emershad and Ramming (ER), Nitsch and Nitsch (NN) and Murashige and Skoog (MS), and plant growth regulators (benzyl amino purine (BA), 0.5, 0.7 and 0.9 mg/l; indole butyric acid (IBA), 1.0, 1.5 and 2.0 mg/l; and gibberellic acid (GA), 0.1, 0.3 and 0.9 mg/l) were screened individually in different combinations with three amino acids, namely cysteine, glutamine and proline (2.0 μmol/l each). The maximum embryos germination percentage recorded at 70 DAF was 63.33, 47.78 and 45.56% in ER, NN and MS media, respectively, supplemented with 0.9 mg/l BA, 2.0 mg/l IBA, 0.9 mg/l GA and 2.0 μmol glutamine. Glutamine was found to have the most significant impact, and it improved the rescued embryos germination. The present study provides a potential recipe for a medium that can facilitate efficient germination of grape embryos.

Protocol for plantlets regeneration through callus induction using root meristem was developed in present investigation. Different mediums were standardized for each step vis-a-via callus induction, callus proliferation and shoot regeneration. In first stage, B5 medium added with 0.7mg/l 2, 4-D and 1mg/l BAP showed 100% callusing while reduction in both growth regulators up to level of 0.5 mg/l 2,4-D and 0.1mg/l BAP results in significant callus proliferation. In last step, shoot regeneration was found to good and healthier in B5 medium supplemented with 0.1 mg/l 2, 4-D only. After regeneration normal growth in plantlets observed under plain B-5 medium, however no abnormality has been noted in developed population. Present protocol will be efficiently useful for chemical mutagen induction studies through callus, observing somaclonal variations and transgenic studies.
