We report on employing in vitro biosynthesized ZnO nanoparticles using L. pubescens shoot methanol extract (50 and 100 mg LP–ZnO NPs) to examine their antimicrobial efficacy against Pseudomonas aeruginosa (ATCC27853), Staphylococcus aureus (ATCC 29213), Aspergillus niger (ATCC 16404 NA), and Aspergillus terreus (TCC 10029). The formation and stability of the investigated ZnO nanoparticles were proven by transmission electron microscopy (TEM), Fourier-transform infrared spectroscopy (FT-IR), UV–vis spectroscopy, X-ray diffraction (XRD), and thermal gravimetric analysis (TGA). The ZnO nanoparticles were rod-shaped (width: 10.76–30.93 nm). The nanoparticles in dimethyl sulfoxide (DMSO) outperformed their water counterparts in terms of their zones of inhibition (ZIs) (marginal means of 12.5 and 8.19 mm, respectively) and minimum inhibition concentrations (MICs) (means of 4.40 and 8.54 mg/mL, respectively). The ZI means for S. aureus, P. aeruginosa, A. terreus, and A. niger were 10.50, 6.13, 12.5, and 11.5 mm, respectively. When treating S. aureus and P. aeruginosa, the ZI of the 50 mg LP–ZnO NPs in water was better (14 mm), with a lower MIC and lower minimum bactericidal/fungicide concentrations (MBC/MFC) (7.22 and 4.88 mg/mL, respectively) than the ZnO and control drugs. The SEM images showed cellular alterations in the surface shapes after the LP–ZnO-NP treatments. Biosynthesized LP–ZnO NPs could have beneficial antibacterial properties, which could allow for future contributions to the development of new antimicrobial drugs.

The present study aimed to synthesize iron oxide nanoparticles loaded with quinine and alkaloids-rich Cinchona officinalis (Peruvian bark) stem bark extract, and further evaluate their cytotoxic effect and apoptosis mechanisms in MCF-7 breast cancer cells. Nanoparticles were prepared by biological reduction of iron oxide with Cinchona officinalis extract, using the green synthesis method. The nanoparticles were characterized by XRD, FT-IR, and UV-vis spectroscopy and transmission electron microscopy (TEM). In vitro cytotoxicity analyses of Cinchona officinalis extract, ferrous oxide, and Cinchona officinalis extract-loaded iron oxide nanoparticles (CO-NPs) were carried out using the MTT test for 24 h and 48 h. We found that CO-NPs reduced the MCF-7 cell viability with IC50 values of 16.2 and 9 µg/mL in 24 h and 48 h, respectively. In addition, CO-NPs were tested with normal hMSCs to determine their toxicity, and we did not find noticeable cytotoxicity. Confocal fluorescent microscopy revealed that CO-NPs efficiently increased the nuclear condensation and chromatin damage in propidium iodide staining; meanwhile, there was decreased mitochondrial membrane potential in CO-NPs-treated MCF-7 cells. In addition, AO-EB staining confirmed the late apoptotic and apoptotic morphology of cancer cells. Further gene expression analysis confirmed that the upregulation of tumor suppressors, Cdkn1A, Prb, and p53 was significantly increased, and inflammatory traits such as TNF-α and Nf-κb were increased in cancer cells treated with CO-NPs. Apoptotic stimulators such as Bax and caspase-3 expression were highly significantly increased, while mdm-2 and Bcl-2 were significantly decreased. Overall, the enhanced cytotoxic potential of the Cinchona officianlis stem bark extract loaded CO-NPs versus free Cinchona officianlis extract might be due to the functional stabilization of bioactive compounds, such as alkaloids, quinine, flavonoids, phenolics, etc., into the iron oxide, providing bioavailability and internalization of cinchona metabolites intracellularly.

In this research, a simple, green approach was employed to synthesize silver nanoparticles with the aid of Ziziphus spina-christi (L.) methanol root extract, which can act as a reducing, capping agent to treat obesity and inflammation. Globally, Ziziphus spina-christi (Jujube) root is used in traditional therapy as a lipolysis promoter. GC-MS results confirmed the availability of kaempferol (flavonol), cannabinol and indole-3-carboxylic acid in Ziziphus spina-christi root methanol extract (ZSE). ZSE silver nanoparticles (ZS-Ag-NPs) were synthesized and their effect on mitochondrial fatty acid oxidation capacity and adipokine levels in maturing adipocytes were analyzed. Maturing adipocytes treated with 0.4 µg/dL of ZSE and ZS-Ag-NPs significantly reduced the lipid content in adipocytes by 64% and 82%, respectively. In addition, lipolysis-related genes such as LPL (1.9 fold), HSL (2.3 fold), PGC-1α (3 fold), UCP-1 (4.1 fold), PRDM16 (2 fold) and PPARα (2.7 fold) increased significantly in ZS-Ag-NPs treated maturing adipocytes. The ZS-Ag-NPs treatment significantly decreased insulin resistance and metabolic inflammation-related LTB4-R, TNF-α, IL-4 and STAT-6 mRNA levels. Mitochondrial thermogenesis stimulating capacity of ZS-Ag-NPs was further confirmed by the significantly enhanced CREB-1 and AMPK protein levels in adipocytes. Furthermore, ZS-Ag-NPs treated adipokines (condition media, CM) were treated with human umbilical vein endothelial cells (HUVECs) to determine cytotoxicity and pro-inflammatory stimulus capacity. We found that ZS-Ag-NPs treated adipocyte CM effectively increased mRNA expression levels of the vascular endothelial cell growth factor (VEGF), and down-regulated oxidative stress (LPO, eNOS, and HO) and vascular cell inflammation (ICAM, VCAM, TNF-α, IL-1β, and NF-κB). In conclusion, ZS-Ag-NPs displayed an action at the molecular level in mitochondrial fatty acid oxidation, decreased adipokine secretion in adipocytes, and enhanced vascular endothelial cell growth. This molecular mechanical action of ZS-Ag-NPs reduced effectively obesity progressions and metabolic inflammatory pathogenesis associated with aging.

The present study reports a cost-effective, environmentally friendly method to increase the bioavailability and bio-efficacy of B. rufescens stem bark extract in the biological system via functional modification as B. rufescens stem bark nanoparticles (BR-TO2-NPs). The biosynthesis of BR- -NPs was confirmed by UV-visible (UV-vis) and Fourier-transform infrared (FT-IR) spectroscopy, transmission electron microscopy (TEM), and X-ray diffraction analyses. The shifts in FT-IR stretching vibrations of carboxylic and nitro groups (1615 cm−1), the O–H of phenolics or carboxylic acids (3405 cm−1), alkanes, and alkyne groups (2925 and 2224 cm−1) of the plant extract and lattice (455) indicated successful biosynthesis of BR- -NPs. Compared with the stem bark extract, 40 ng/dL dose of BR- -NPs led to a reduction in adipogenesis and an increase in mitochondrial biogenesis-related gene expressions, adiponectin-R1, PPARγC1α, UCP-1, and PRDM16, in maturing-adipocytes. This confirmed the intracellular uptake, bioavailability, and bio-efficiency of BR-TiO2-NPs. The lipid-lowering capacity of BR-TiO2-NPs effectively inhibited the metabolic inflammation-related gene markers, IL-6, TNF-α, LTB4-R, and Nf-κb. Further, BR-TiO2-NPs stimulating mitochondrial thermogenesis capacity was proven by the significantly enhanced CREB-1 and AMPK protein levels in adipocytes. In conclusion, BR-TiO2-NPs effectively inhibited lipid accumulation and proinflammatory adipokine levels in maturing adipocytes; it may help to overcome obesity-associated comorbidities.

Polycystic ovary syndrome (PCOS) is a frequent disorder that affects reproductive-aged women and has reproductive, metabolic, and psychosocial effects. This research was intended to investigate the comparison between food intake and adipose tissue distribution in Saudi women suffering from PCOS and a control group. To determine the sociodemographic variables, a case–control study was performed with patients from King Fahad Medical City’s Reproductive Endocrine and Infertility Medicine Department (REIMD). The case–control study comprised 42 PCOS patients (PCOS-Ps) and 63 as a control group, all aged 20–45 years. Three-day records were collected from participants to estimate the nutrient intake of cases and controls. A body composition analyzer was used to measure body mass index (BMI), body fat (BF), and visceral fat (VF). Biochemical measurements were taken to determine the lipid profile, total testosterone, and serum vitamin D-25-OH. The women’s frequency distribution based on sociodemographic characteristics revealed significant differences within and between the groups. The variations in dietary intake between the PCOS-P and control groups were primarily in terms of total calories, carbohydrates, niacin, and folate, all of which were significantly higher in the PCOS-P group. Dietary fiber, unsaturated fat, vitamin A, vitamin B12, calcium, phosphorus, and selenium, on the other hand, were significantly higher in the control group. A majority of both groups had significantly higher BMI (overweight or obese) and higher BF, but normal VF. According to the findings, testosterone levels in PCOS-Ps were significantly higher than in the control group, but vitamin D-25-OH and high-density-lipoprotein cholesterol (HDL-C) were significantly lower. Age, monthly income, cholesterol, low-density-lipoprotein cholesterol (LDL-C), and testosterone were the fundamental causes impacting women’s anthropometric indices. In conclusion, although both groups were overweight or obese, and differences in calorie and nutrient intake, HDL-C, testosterone, and vitamin D-25-OH levels were observed. The study advises such population groups to limit their consumption of foods high in calories.

The study aimed to investigate the effects of mixed-mode autoclaving (MMA; sterilization of flour–water mixture; as control) and single-mode autoclaving (SMA; sterilization of flour and water separately) on the potential of the fermenting microorganism Lactiplantibacillus plantarum ATCC 8014 (L. plantarum), focusing on nutrient content, bioactive compounds, and antioxidant activity in millet flour. Pearl millet (Pennisetum glaucum (L.) R.Br.) flour mixed with water (1:4, w/v) was fermented with L. plantarum (108 colony-forming unit [CFU]/g) in a fermentor at 37°C for 12, 24, 36, 48, 60, and 72 h, with continuous stirring at 120 rpm, follow-ing sterilization in an autoclave (121.1°C; 15 psi; 15 min). SMA and MMA had varied effects on micronutrients and minerals, and fermentation affected them. Fermentation of L. plantarum results in substantially higher glucose production in SMA compared to MMA, whereas the opposite is true for fructose. SMA had higher glu-cose levels and lower fructose levels than MMA. The SMA L. plantarum and MMA-fermented samples showed similar trends in glucose and fructose changes. SMA samples contained higher total phenolic content (TPC) than MMA, while total flavonoid content (TFC) and total tannin content (TTC) remained unchanged. TPC and TFC increased gradually, while TTC decreased, after 72-h L. plantarum fermentation. The 2,2-Diphenyl-1-picrylhydrazyl (DPPH) scavenging rate in sterilized raw millet flour (0MMA) was lower than in raw millet flour, and it was also lower in SMA than in MMA. The DPPH scavenging rate of 0MMA increased significantly after 12 h of fermentation, reaching its peak at 36 h, with the MMA surpassing SMA in scavenging activity. High-performance liquid chromatography–mass spectrometry (HPLC-MS) analysis detected bioactive substances in SMA- and MMA-fermented millet, with some variations in their nature and levels. Overall, these results indicate that sterilization method and fermentation time are key factors in shaping the nutrient and bioactive compound content of millet flour, highlighting the need to optimize them to develop nutritious, health-promoting fermented millet products.
