Pak choi (Brassica rapa ssp. chinensis Makino) is a representative seed vernalization vegetable and premature bolting in spring can cause significant economic loss. Thus, it is critical to elucidate the mechanism of molecular regulation of vernalization and floral bud initiation to prevent premature bolting. Gibberellin (GA) is the key plant hormone involved in regulating plant development. To gain a better understanding of GA metabolism in pak choi, the content of GA in pak choi was measured at different stages of plant development using enzyme-linked immunosorbent assay. The results showed that the GA content increased significantly after low-temperature treatment (4°C) and then decreased rapidly with vegetative growth. During floral bud initiation, the GA content increased rapidly until it peaked upon floral bud differentiation. To elucidate these changes in GA content, the expression of homologous genes encoding enzymes directly involved in GA metabolism were analyzed. The results showed that the changes in the expression of four genes involved in GA synthesis (Bra035120 encoding ent-kaurene synthase, Bra009868 encoding ent-kaurene oxidase, Bra015394 encoding ent-kaurenoic acid oxidase, and Bra013890 encoding GA20-oxidase) were correlated with the changes in GA content. In addition, by comparing the expression of genes involved in GA metabolism at different growth stages, seven differentially expressed genes (Bra005596, Bra009285, Bra022565, Bra008362, Bra033324, Bra010802, and Bra030500) were identified. The differential expression of these genes were directly correlated with changes in GA content, suggesting that these genes were directly related to vernalization, floral bud initiation and development. These results contribute to the understanding of the molecular mechanism of changes in GA content during different developmental phases in pak choi.

Pak choi is a seed vernalization-type plant whose vernalization mechanism is currently unclear. Therefore, it is critical to discover genes related to vernalization and research its functions during vernalization in pak choi. Here, the gene expression profiles in the shoot apex were analyzed after low temperature treatment using high-throughput RNA sequencing technology. The results showed that there are 1,664 and 1,192 differentially expressed genes (DEGs) in pak choi in cold treatment ending and before flower bud differentiation, respectively, including 42 genes that exhibited similar expression trend at both stages. Detailed annotation revealed that the proteins encoded by the DEGs are located in the extracellular region, cell junction and extracellular matrix. These proteins exhibit activity such as antioxidant activity and binding protein/transcription factor activity, and they are involved in signal transduction and the immune system/biological processes. Among the DEGs, Bra014527 was up-regulated in low temperature treatment ending, Bra024097 was up-regulated before flower bud differentiation and Bra035940 was down-regulated at both stages in low temperature-treated shoot apices. Homologues of these genes in A. thaliana, AT3G59790, AT4G30200 and AT5G61150, are involved in flowering and vernalization, suggesting that they take part in the vernalization process in pak choi. Further pathway enrichment analysis revealed that most genes were enriched in the tryptophan metabolism and glucosinolate biosynthesis pathways. However, the functions of tryptophan and glucosinolate in vernalization are not yet clear and require further analysis.

The CO2 saturation point can reach as high as 1819 μmol· mol−1 in carrot (Daucus carota L.). In recent years, carrot has been cultivated in out-of-season greenhouses, but the molecular mechanism of CO2 enrichment has been ignored, and this is a missed opportunity to gain a comprehensive understanding of this important process. In this study, it was found that CO2 enrichment increased the aboveground and belowground biomasses and greatly increased the carotenoid contents. Twenty genes related to carotenoids were discovered in 482 differentially expressed genes (DEGs) through RNA sequencing (RNA-Seq.). These genes were involved in either carotenoid biosynthesis or the composition of the photosystem membrane proteins, most of which were upregulated. We suspected that these genes were directly related to quality improvement and increases in biomass under CO2 enrichment in carrot. As such, β-carotene hydroxylase activity in carotenoid metabolism and the expression levels of coded genes were determined and analysed, and the results were consistent with the observed change in carotenoid content. These results illustrate the molecular mechanism by which the increase in carotenoid content after CO2 enrichment leads to the improvement of quality and biological yield. Our findings have important theoretical and practical significance.

Strawberry (Fragaria ananassa Duch.) is an economically important crop. Its yield is highly dependent on photosynthesis, and carbon dioxide (CO2) is one of the most important constituents in photosynthesis. In this study, we investigated the physiological and molecular mechanisms of CO2 enrichment in increasing strawberry leaf growth, and ultimately fruit yield. Strawberry plants were grown under ambient CO2 (350–500 μmol/mol) and elevated CO2 (750–850 μmol/mol) in a controlled environment. The results showed that CO2 enrichment significantly increased the size, net photosynthetic rate and light saturation point of the leaves, but decreased the light compensation point of leaves. Using RNA sequencing (RNA-Seq), transcriptome profiling of leaf tissues under two CO2 concentrations was conducted. The analysis identified 150 differentially expressed genes in response to increased CO2 concentration; 14 of them were shown to be involved in photosynthesis. Gene Ontology (GO) enrichment and pathways analyses revealed that CO2 enrichment enhanced the biosysthesis of metabolism-promoting growth hormones; sped up the reduction step of CO2 assimilation, and increased the transport of photosynthetic products, which help explain the increased net photosynthetic rate and accelerated leaf growth under elevated CO2 level. These results provide a valuable reference for exploring the mechanism of CO2 application in strawberry production in greenhouses especially during offseason.

Carrot is an important vegetable with roots as the edible organ. A complex regulatory network controls root growth, in which auxin is one of the key players. To clarify the molecular mechanism on auxin regulating carrot root expansion, the growth process and the indole-3-acetic acid (IAA) content in the roots were measured in this experiment. It was found that the rapid expansion period of the root was from 34 to 41 days after sowing and the IAA content was the highest during this period. The root growth then slowed down and the IAA levels decreased. Using the transcriptome sequencing database, we analyzed the expression of IAA-metabolism-related genes and found that the expression of most of the IAA synthesis genes, catabolism genes, and genes related to signal transduction was consistent with the changes in IAA content during root expansion. Among them, a total of 31 differentially expressed genes (DEGs) were identified, including 10 IAA synthesis genes, 8 degradation genes, and 13 genes related to signal transduction. Analysis of the correlations between the DEGs and IAA levels showed that the following genes were closely related to root development: three synthesis genes, YUCCA10 (DCAR_012429), TAR2 (DCAR_026162), and AMI1 (DCAR_003244); two degradation genes, LPD1 (DCAR_023341) and AACT1 (DCAR_010070); and five genes related to signal transduction, IAA22 (DCAR_012516), IAA13 (DCAR_012591), IAA27 (DCAR_023070), IAA14 (DCAR_027269), and IAA7 (DCAR_030713). These results provide a reference for future studies on the mechanism of root expansion in carrots.
