The head, which performs many biological functions, is the most complicated structure of an insect. Development, locomotor behavior, food intake, environmental sensing, and signal transduction are all controlled by the insect’s head. As a well-studied insect in Lepidoptera, the silkworm head has an additional function of spinning silk fibers. To understand which molecules are involved in these physiological activities, we performed a metabolomics analysis of silkworm heads. By integrating GC-MS and LC-MS/MS, 90 metabolites were identified in the larval heads of silkworms. These were classified into 13 categories, including amino acids, sugars, organic acids, nucleotides, alcohols, and fatty acids. Informatics analysis revealed that these metabolites are involved in cellular processes, environmental information processing, genetic information processing, human diseases, metabolism, organismal systems, and other pathways. The identified metabolites and pathways are involved in biological processes such as signal transduction, carbohydrate metabolism, endocrine activities, and sensory activities; reflecting the functions of various organs in silkworm heads. Thus, our findings provide references which elucidate the potential functions of the silkworm head and will be of great value for the metabolomics research of silkworms and other insects.

Silk fiber has become a research focus because of its comprehensive mechanical properties. Metal ions can influence the conformational transition of silk fibroin. Current research is mainly focused on the role of a single ion, rather than the whole metal ion environment. Here, we report the effects of the overall metal ion environment on the secondary structure and mechanical properties of silk fibers after direct injection and feeding of silkworms with EDTA. The metal composition of the hemolymph, silk gland, and silk fiber changed significantly post EDTA treatment. Synchrotron FTIR analysis indicated that the secondary structure of silk fiber after EDTA treatment changed dramatically; particularly, the β-sheets decreased and the β-turns increased. Post EDTA treatment, the silk fiber had significantly decreased strength, Young’s modulus, and toughness as compared with the control groups, while the strain exhibited no obvious change. These changes can be attributed to the change in the metal ion environment in the silk fibroin and sericin in the silk gland. Our investigation provides a new theoretical basis for the natural silk spinning process, and our findings could help develop a method to modify the mechanical properties of silk fiber using metal ions.

The silkworm (Bombyx mori) is an economically-important insect that can secrete silk. Carboxypeptidases have been found in various metazoan species and play important roles in physiological and biochemical reactions. Here, we analyzed the silkworm genome database and characterized 48 carboxypeptidases, including 34 metal carboxypeptidases (BmMCP1–BmMCP34) and 14 serine carboxypeptidases (BmSCP1–BmSCP14), to better understand their diverse functions. Compared to other insects, our results indicated that carboxypeptidases from silkworm have more family members. These silkworm carboxypeptidases could be divided into four families: Peptidase_M2 carboxypeptidases, Peptidase_M14 carboxypeptidases, Peptidase_S10 carboxypeptidases and Peptidase_S28 carboxypeptidases. Microarray analysis showed that the carboxypeptidases had distinct expression patterns, whereas quantitative real-time PCR demonstrated that the expression level of 13 carboxypeptidases significantly decreased after starvation and restored after re-feeding. Overall, our study provides new insights into the functional and evolutionary features of silkworm carboxypeptidases.

Apolipoprotein D (ApoD) is an essential protein for insect internal circulation because of its role in lipid transportation. In this study, we cloned and characterized ApoD and analyzed its expression characteristics in Bombyx mori (L.) (Lepidoptera: Bombycidae). The ApoD cDNA fragment, which has an open reading frame of 522 bp with a lipocalin domain (PF08212), encodes 174 amino acids. The deduced ApoD protein sequence has 28.2% identity to its ortholog in Mus musculus and 27.8% identity to its ortholog GLaz in Drosophila melanogaster . Several putative cis-acting elements related to development were also examined. Using quantitative real time polymerase chain reaction (RT-PCR), the ApoD gene expression was detected in the head and silk glands. Western blotting showed that ApoD protein was present in the head, hemolymph, fat body, gonad, middle silk gland, and posterior silk gland. ApoD expression was relatively higher in the hemolymph during days 1–3 of fifth-instar larvae and days 6–8 of the pupal stage. The highest mRNA levels were detected in the pupae on day 9 by using quantitative RT-PCR. After an oxidative stress treatment and a bacterial challenge, expression levels of the ApoD gene from B. mori were up-regulated at different time points compared with the control group. Simultaneously, immunohistochemical localization showed that ApoD is widely detected in the brain, eye, ovary, and testis on day 3 of the fifth-instar larval stage. The results indicated that ApoD is involved in insect development and reproduction, and plays an important role in the response to bacterial challenge and other stress signals.

Starvation imposes significant stress on animal survival and development, resulting in organ damage within the organism. The brain, being one of the most vital organs in animals, plays a crucial role in coordinating the physiological functions of other organs. However, performing brain experiments on the human body is challenging. In this work, we selected the silkworm, a model Lepidoptera organism, due to its favorable characteristics. A comprehensive transcriptome analysis was conducted on the brain of silkworm subjected to starvation treatment. The analysis of differentially expressed genes revealed significant alterations in 330 genes following the period of starvation. Through an enrichment analysis, we successfully identified pathways associated with metabolism, hormones, immunity, and diseases. Our findings highlight the transcriptional response of the brain to starvation, providing valuable insights for comprehending the impact of starvation stress in other animals.
