Rice plants contain high basal levels of salicylic acid (SA), but some of their functions remain elusive. To elucidate the importance of SA homeostasis in rice immunity, we characterized four rice SA hydroxylase genes (OsSAHs) and verified their roles in SA metabolism and disease resistance. Recombinant OsSAH proteins catalyzed SA in vitro, while OsSAH3 protein showed only SA 5-hydroxylase (SA5H) activity, which was remarkably higher than that of other OsSAHs that presented both SA3H and SA5H activities. Amino acid substitutions revealed that three amino acids in the binding pocket affected SAH enzyme activity and/or specificity. Knockout OsSAH2 and OsSAH3 (sahKO) genes conferred enhanced resistance to both hemibiotrophic and necrotrophic pathogens, whereas overexpression of each OsSAH gene increased susceptibility to the pathogens. sahKO mutants showed increased SA and jasmonate levels compared to those of the wild type and OsSAH-overexpressing plants. Analysis of the OsSAH3 promoter indicated that its induction was mainly restricted around Magnaporthe oryzae infection sites. Taken together, our findings indicate that SA plays a vital role in immune signaling. Moreover, fine-tuning SA homeostasis through suppression of SA metabolism is an effective approach in studying broad-spectrum disease resistance in rice.

Maize (Zea mays L.) is a globally vital crop for food, feed, and biofuel production, with plant height (PH) being a key agronomic trait that significantly influences yield, lodging resistance, and stress tolerance. This study identified a single-base mutation in the D1 (Dwarf 1) gene responsible for the dwarf phenotype in the maize mutant 16N125. Through genetic analysis and fine mapping, the candidate region was localized to chromosome 3, narrowing it down to an interval containing three genes. Sequencing revealed a non-synonymous mutation in D1, which encodes a gibberellin 3-beta-dioxygenase, leading to amino acid substitutions at positions 61 and 123. Genetic analysis of F2 populations confirmed that the mutation at position 61 was responsible for the dwarf trait. Furthermore, the mutation was detected in several Chinese inbred lines, indicating its potential role in dwarfing under specific conditions. These findings provide critical insights into the genetic mechanisms regulating maize plant height, offering valuable information for breeding programs focused on improving crop architecture and yield to address the challenges of global food security and climate change.

Salicylic acid (SA) serves as a pivotal plant hormone involved in regulating plant defense mechanisms against biotic stresses, but the extent of its biological significance in relation to peanut resistance is currently lacking. This study elucidated the involvement of salicylic acid (SA) in conferring broad-spectrum disease resistance in peanuts through the experimental approach of inoculating SA-treated leaves. In several other plants, the salicylate hydroxylase genes are the typical susceptible genes (S genes). Here, we characterized two SA hydroxylase genes (AhS5H1 and AhS5H2) as the first S genes in peanut. Recombinant AhS5H proteins catalyzed SA in vitro, and showed SA 5-ydroxylase (S5H) activity. Overexpression of AhS5H1 or AhS5H2 decreased SA content and increased 2,5-DHBA levels in Arabidopsis, suggesting that both enzymes had a similar role in planta. Moreover, overexpression of each AhS5H gene increased susceptibility to Pst DC3000. Analysis of the transcript levels of defense-related genes indicated that the expression of AhS5H genes, AhNPR1 and AhPR10 was simultaneously induced by chitin. Overexpression of each AhS5H in Arabidopsis abolished the induction of AtPR1 or AtPR2 upon chitin treatment. Eventually, AhS5H2 expression levels were highly correlated with SA content in different tissues of peanut. Hence, the expression of AhS5H1 and AhS5H2 was tissue-specific.

Maize production in Fuxin City, Liaoning Province, China, is threatened by northern corn leaf blight (NCLB) and Fusarium stalk rot, with straw return under conservation tillage exacerbating the NCLB severity by 20% in local fields. This study evaluated the efficacy of combining difenoconazole, a commonly used fungicide, with a Trichoderma bioagent for disease control in straw-incorporated soils. Field trials in Fuxin showed that applying 300 g/ha difenoconazole with 1.5 L/ha Trichoderma fermentate achieved superior results: a 72.4% reduction in the NCLB disease index and a stalk rot incidence of only 0.61%. These outcomes significantly outperformed single-component treatments like difenoconazole alone (56.2% NCLB suppression) or other fungicides (e.g., carbendazim, triadimefon). The combined treatment also outperformed the single treatments with biocontrol agent (67.1% NCLB inhibition). The results highlight the synergistic potential of integrating chemical and biological agents to manage residue-borne diseases, offering a practical strategy for sustainable disease control in conservation agriculture systems with straw return in Liaoning, China.

Breeding for dwarfing traits in sorghum is crucial. However, only three genes (Dw1–Dw3) that control plant height have been mapped. In this study, 634 sorghum cultivars were collected to investigate plant height and genotypes. Four were genotyped Dw1DW2Dw3 (wild type) but with different plant heights, and they were selected to construct two populations and map new dwarf genes. Bulked segregant analysis with whole-genome resequencing of the two populations identified the candidate gene in one same genomic region—on chromosome 7. Then, it was narrowed down to a region containing nine genes. Amino acid and DNA sequence analysis of the parent and offspring plants revealed that two novel allelic mutations in the Dw3 gene play a role in reducing the plant height—8R262 or 8R417, including 1 bp substitution and 2 bp deletions. Furthermore, we sequenced 19 cultivars that primarily exhibited a “one-dwarf” hybrid or wild-type and presumed another allelic mutation via the amino acid alignment of 8R019, 8R100, and 8R402, which was another one-base substitution. These results indicate that multiple types of allelic mutations in the Dw3 gene should be considered when identified or applied.

A comprehensive two-year investigation (2024–2025) was conducted across Northeast China’s crucial grain production base to assess the status of maize diseases. Field surveys spanning three provinces and Inner Mongolia revealed a significant shift in the regional disease profile, with diagnosis performed by experienced personnel based on characteristic field symptoms. The results demonstrated that maize white spot (MWS) has emerged as a severe new threat, recording remarkably high disease severity indices exceeding 80 at multiple locations (e.g., LDD25-1: 86.83). Concurrently, gray leaf spot (GLS) was confirmed as the most prevalent foliar disease, forming stable areas of high severity in the eastern mountainous regions where its disease indices consistently surpassed 60 (e.g., LFS25-1: 65.26), thereby exceeding the impact of northern corn leaf blight. In contrast, stalk rot (SR) maintained a low field incidence rate below 10%, while other diseases such as Curvularia leaf spot and maize eyespot were only observed locally or were absent during the 2025 survey period. These findings underscore the emergence of MWS as a critical threat and affirm the dominant status of GLS, offering a scientific foundation for prioritizing disease management strategies in the region.
