This study aimed to utilize enzymatic treatment and pressurized hot water extraction (PHWE) to recover soluble food-grade protein and collagen peptides from mechanically deboned chicken meat (MDCM), a side-stream from the meat industry. Food-grade enzyme blends Ermitase 1 and Ermitase 2 were used to fractionate the mechanically deboned meat into fat, soluble protein, and solids. Response surface methodology was utilized to optimize treatments to maximize the protein yield. At the optimum conditions (hydrolysis time 240 min, E:S 0.27%, and a hydromodule 1 L/kg), the enzymatic treatment produced high protein yield, approximately 90%. The protein hydrolysates showed a good solubility index, but weak gelling properties. The PHWE of the bones resulted in a high nitrogen yield, approximately 87%, at the optimum conditions of 190 °C and 83 min. Peptides in the bone extract were in the range of 0.5–13.7 kDa. Overall, our study highlights the importance of response surface methodology to optimize parameters for mechanically deboned chicken meat enzymatic and PHWE treatments to achieve high yields of protein for food applications and low-molecular-weight collagen peptides for cosmetic applications. The crucial role of protein and peptide prices was observed in preliminary profitability analysis.

During recent years there have been shortages of certain drugs due to problems in raw material supply. These are often related to active ingredients but could also affect excipients. Lactose is one of the most used excipients in tableting and comes in two anomeric and several solid-state forms. The aim of this study was to utilize lactose from a dairy side-stream and compare it against a commercial reference in direct compression. This would be a sustainable option and would secure domestic availability during crises. Two types of lactose, spray-dried and freeze-dried, were evaluated. Lactose was mixed with microcrystalline cellulose in different ratios together with lubricant and glidant, and flowability and tabletability of the formulations was characterized. The fully amorphous and small particle-sized spray-dried lactose flowed inadequately but exhibited good tabletability. The larger particle-sized, freeze-dried lactose exhibited sufficient flow and better tabletability than the commercial reference. However, disintegration and drug release were slower when using the investigational lactose formulations. This was most likely due to remaining milk proteins, especially caseins, in the lactose. Overall, the investigational lactose provides promise for the use of such a side-stream product during crisis situations but enhancing their properties and/or purity would be needed.

Drug shortages are a current issue and often originate from problems in raw material supply. Even though they are more related to active ingredients the reason could be the lack of excipient(s) due to a major disruption to normal supply chains. Lactose is one of the most used excipients in oral solid dosage forms such as tablets. The aim of this study was to utilize lactose from a dairy side-stream in wet granulation and tableting and evaluate its behaviour in a formulation in comparison to a commercial reference. The possibility to use this type of lactose would be a sustainable option to ensure domestic availability during crises. The study design included different lactose proportions of fillers as well as liquid to solid ratios to investigate whether the resulting granules show similar responses when using side-stream or commercial lactose. The responses to changes were rather similar regardless of the lactose grade but liquid to solid ratio needed to be decreased (0.6 to 0.8. vs. 0.4 to 0.6) when using side-stream lactose. The formulations using side-stream lactose exhibited adequate flowability (3rd best classification in Ph.Eur) which was slightly inferior to that with commercial but tabletability was better due to its amorphous nature. However, disintegration (about 20 min vs. 2 min) and drug release (60-80 % vs. 100 %) were slower when using the side-stream lactose. This was most likely due to remaining milk proteinsin the lactose. Overall, the side-stream lactose provided promise for the use of such a side-stream material during supply disruptions.

Hankkeessa on tuotettu verkko-oppimisympäristöön ammattikorkeakoulujen yhteisiä biotalousalan verkkokursseja.
