We prospectively assessed 49 coronavirus disease cases in Guangdong, China, to estimate the frequency and duration of detectable severe acute respiratory syndrome coronavirus 2 RNA in human body fluids. The prolonged persistence of virus RNA in various body fluids may guide the clinical diagnosis and prevention of onward virus transmission.

Background and aims: Understanding the age-related trend of risk in high blood pressure (BP) is important for preventing heart failure and cardiovascular diseases. But such a trend is still underexplored. This study aims to (a) depict the relationship of BP patterns with age, and (b) understand the trend of high BP prevalence over time in different age groups.Materials and methods: Health check-up data with an observational period of 8 years (January 1, 2011, to December 31, 2018) was used as the data source. A total of 71,468 participants aged over 18 years old with complete information on weight, height, age, gender, glucose, triglyceride, total cholesterol, systolic (SBP), and diastolic blood pressure (DBP) were included for analysis. Generalized additive models were adopted to explore the relationship between the risk of high BP and age. Variance analysis was conducted by testing the trend of high BP prevalence in age groups over time. Results: Risk of high SBP showed a continuous rise from age 35 to 79 years and a concurrent early increase in the risk of high DBP; after age 50–65 years, high DBP risk declined. The risk of SBP rises linearly with age for men, whereas increases non-linearly for women. In addition, a significant increasing trend of high SBP risk among middle-aged people was found during the past decade, men experienced a later but longer period of increase in high SBP than women. Conclusion: The high SBP risk progresses more rapidly in the early lifetime in women, compared to the lifetime thereafter. Thresholds of increasing trend of SBP suggest a possible need for hypertension screening in China after the age of 40.

Tumor-derived circulating cell-free DNA (cfDNA) provides critical clues for cancer early diagnosis, yet it often suffers from low sensitivity. Here, we present a cancer early diagnosis approach using tumor fractions deciphered from circulating cfDNA methylation signatures. We show that the estimated fractions of tumor-derived cfDNA from cancer patients increase significantly as cancer progresses in two independent datasets. Employing the predicted tumor fractions, we establish a Bayesian diagnostic model in which training samples are only derived from late-stage patients and healthy individuals. When validated on early-stage patients and healthy individuals, this model exhibits a sensitivity of 86.1% for cancer early detection and an average accuracy of 76.9% for tumor localization at a specificity of 94.7%. By highlighting the potential of tumor fractions on cancer early diagnosis, our approach can be further applied to cancer screening and tumor progression monitoring.

Neuroimaging studies have shown that heroin addiction is related to abnormalities in widespread local regions and in the functional connectivity of the brain. However, little is known about whether heroin addiction changes the topological organization of whole-brain functional networks. Seventeen heroin-dependent individuals (HDIs) and 15 age-, gender-matched normal controls (NCs) were enrolled, and the resting-state functional magnetic resonance images (RS-fMRI) were acquired from these subjects. We constructed the brain functional networks of HDIs and NCs, and compared the between-group differences in network topological properties using graph theory method. We found that the HDIs showed decreases in the normalized clustering coefficient and in small-worldness compared to the NCs. Furthermore, the HDIs exhibited significantly decreased nodal centralities primarily in regions of cognitive control network, including the bilateral middle cingulate gyrus, left middle frontal gyrus, and right precuneus, but significantly increased nodal centralities primarily in the left hippocampus. The between-group differences in nodal centralities were not corrected by multiple comparisons suggesting these should be considered as an exploratory analysis. Moreover, nodal centralities in the left hippocampus were positively correlated with the duration of heroin addiction. Overall, our results indicated that disruptions occur in the whole-brain functional networks of HDIs, findings which may be helpful in further understanding the mechanisms underlying heroin addiction.
