The clinical detection of carbohydrate antigen 19-9 (CA 19-9), a tumor marker in biological samples, improves and facilitates the rapid screening and diagnosis of pancreatic cancer. A simple, low cost, fast, and green synthesis method to prepare a viable carbon quantum dots/gold (CQDs/Au) nanocomposite fluorescence immunosensing solution for the detection of CA 19-9 was reported. The present method is conducted by preparing glucose-derived CQDs using a microwave-assisted method. CQDs were employed as reducing and stabilizing agents for the preparation of a CQDs/Au nanocomposite. The immobilized anti-CA 19-9-labeled horseradish peroxidase enzyme (Ab–HRP) was anchored to the surface of a CQDs/Au nanocomposite by a peptide interaction between the carboxylic and amine active groups. The CA 19-9 antigen was trapped by another monoclonal antibody that was coated on the surface of microtiter wells. The formed sandwich capping antibody–antigen–antibody enzyme complex had tunable fluorescence properties that were detected under excitation and emission wavelengths of 420 and 530 nm. The increase in fluorescence intensities of the immunoassay sensing solution was proportional to the CA 19-9 antigen concentration in the linear range of 0.01–350 U mL−1 and had a lower detection limit of 0.007 U mL−1. The proposed CQDs/Au nanocomposite immunoassay method provides a promising tool for detecting CA 19-9 in human serum.
Two noble metals, such as silver and gold alloy nanoparticles, were successfully synthesized by the microwave assisted method in the presence of the Asparagus racemosus root extract and were used as an antibacterial and immunomodulatory agent. The nanostuctures of the synthesized nanoparticles were confirmed by various spectroscopic and microscopic techniques. The UV-vis spectrum exhibits a distinct absorption peak at 483 nm for the bimetallic alloy nanoparticles. The microscopic analysis revealed the spherical shaped morphology of the biosynthesized nanoparticles with a particle size of 10–50 nm. The antibacterial potential of the green synthesized single metal (AgNPs and AuNPs) and bimetallic alloy nanoparticles was tested against five bacterial strains. The bimetallic alloy nanoparticles displayed the highest zone of inhibition against P. aeurgnosia and S.aureus strains when compared to single metal nanoparticles and plant extract. In addition, the inmmunomodulatory potential of the root extract of A. racemosus, AgNPs, AuNPs, and Ag-Au alloy NPs is achieved by measuring the cytokine levels in macrophages (IL-1β, IL-6, and TNF-α) and NK cells (IFN-γ) of NK92 and THP1 cells using the solid phase sandwich ELISA technique. The results showed that the root extract of A. racemosus, AgNPs, and AuNPs can reduce the pro-inflammatory cytokine levels in the macrophages cells, while Ag-Au alloy NPs can reduce cytokine responses in NK92 cells. Overall, this study shows that the microwave assisted biogenic synthesized bimetallic nanoalloy nanoparticles could be further explored for the development of antibacterial and anti-inflammatory therapies.
This work describes an environmental-friendly preparation of ZnO nanoparticles using aqueous oat extract. The advanced electrochemical and optical features of green synthesized ZnONPs displayed excellent antibacterial activity and exhibited an important role in pharmaceutical determinations. The formation of nanoscale ZnO was confirmed using various spectroscopic and microscopic investigations. The formed nanoparticles were found to be around 100 nm. The as-prepared ZnONPs were monitored for their antibacterial potential against different bacterial strains. The inhibition zones for ZnONPs were found as Escherichia coli (16 mm), Pseudomonas aeruginosa (17 mm), Staphylococcus aureus (12 mm) and Bacillus subtilis (11 mm) using a 30-µg mL−1 sample concentration. In addition, ZnONPs exhibited significant antioxidant effects, from 58 to 67%, with an average IC50 value of 0.88 ± 0.03 scavenging activity and from 53 to 71% (IC50 value of 0.73 ± 0.05) versus the scavenging free radicals DPPH and ABTS, respectively. The photocatalytic potential of ZnONPs for Rhodamine B dye degradation under UV irradiation was calculated. The photodegradation process was carried out as a function of time-dependent and complete degradation (nearly 98%), with color removal after 120 min. Conclusively, the synthesized ZnONPs using oat biomass might provide a great promise in the future for biomedical applications.
The rapid detection of lung cancer in early stages using the antigen cytokeratin-19 fragment (CYFRA 21-1) as a tumor marker in human serum plays an important role in the survival of patients and taking a fast surgical reaction. This study aimed to employ the green synthesized carbon quantum dots conjugated zinc oxide nanocomposite as a highly sensitive fluorescence immunosensing solution for fast determination of CYFRA 21-1 antigen in human serum. The suggested method was conducted by applying a hydrothermal method to prepare carbon quantum dots using Citrus lemon pericarp. The formed carbon quantum dots were used in the reduction and stabilization of zinc acetate to synthesize carbon quantum dots-zinc oxide nanocomposite. To form a sandwich capping antibody-antigen-antibody immunosensing system, a CYFRA 21-1 antigen was trapped by immobilizing a non-conjugated monoclonal antibody BM 19.21 on the surface of carbon quantum dots-zinc oxide nanocomposite and another monoclonal antibody KS 19.1, which was coated on the microtiter well surface. This system has a tunable fluorescence feature recorded at excitation and emission of λex = 470 and λem = 520 nm, respectively. The suggested nanocomposite fluorescence immunosensing system displayed a linear relationship of 0.01–100 ng mL−1 with a limit of detection of 0.008 ng mL−1. The suggested immunosensing system based on carbon quantum dots-zinc oxide nanocomposite provides a promising approach for rapid diagnoses of lung cancer by detecting CYFRA 21-1 in human serum.
Early diagnosis and clinical treatments of breast cancer provide a highly successful chance for patients to survive. Herein, the detection of CA 27-29 breast cancer antigen was conducted using a label-free ultrasensitive electrochemical immunosensor Au/MoS2/rGO nanocomposite system. The high electrochemical sensitivity of the as-prepared nanocomposites was provided as a result of the high synergistic effect between AuNPs, MoS2 and rGO. Signal amplification of the amperometric detection was evaluated by monitoring the electrocatalytic current response of hydrogen peroxide reduction through the immunoreaction between the anti-CA 27-29 and CA 27-29 antigen on the surface of the Au/MoS2/rGO nanocomposites. The immunosensor displayed an excellent specificity and high sensitivity towards the detection of CA 27-29 breast cancer antigen in human serum with low detection limits of 0.08 U mL−1. The reproducibility, stability and specificity of the suggested immunosensor were evaluated and the recorded results indicated that this type of immunosensor can play an important role in the clinical diagnosis of different types of tumor markers.
