Single-nucleotide polymorphisms (SNPs) in the Fc gamma receptor IIB (FCGR2B) gene have recently been found to be associated with several human autoimmune diseases. We undertook the current study to investigate the influence of these polymorphisms on the risk of ankylosing spondylitis (AS). A total of 306 patients with AS from Anhui, China, fulfilling the modified New York Criteria, and 300 matched healthy controls were analysed. All subjects were genotyped for two SNPs (rs1050501, rs10917661) in the FCGR2B gene, and the SNaPshot Assay was used for genotyping. SNP rs10917661 was significantly associated with AS [C vs. T: odds ratio (OR) 1.723, 95% confidence interval (CI) 1.086-2.733, p = 0.020; genotype: p = 0.026] whereas no association was found for rs1050501. Furthermore, no haplotype was found to be associated with AS. These findings indicated that rs10917661 may be a novel SNP involved in AS genetic predisposition in the Han Chinese population.

Studies characterizing the epidemic of Mycoplasma pneumoniae ( M. pneumoniae ) in Henan Province are scarce. Our research investigated the epidemiological changes of M. pneumoniae and macrolide-resistant mutations of M. pneumonia e (MRMP) among children under 18 years from January 2017 to December 2024. The study was categorized into three stages based on the COVID-19 timeline (pre-pandemic, pandemic, and post-pandemic) to analyze the impact of nonpharmaceutical interventions (NPIs) on M. pneumoniae in Henan Province. Among 27,056 children, 28.36% tested positive for M. pneumoniae , with the lowest positivity rate in 2022 (12.56%) and the highest in 2023 (35.01%). The positivity rate was greatest in the 6–17 years (41.08%) and lowest in ≤1 year (12.38%). Epidemic seasons were predominantly in autumn (36.71%) and winter (23.12%). The MRMP rate increased from 36.68% in 2018 to 87.16% in 2023, averaging 71.00%. The positivity rate of M. pneumoniae was 26.12% during the pre-pandemic stage, significantly declined during the pandemic stage (14.97%), and rebounded to a high level in the post-pandemic stage (31.88%). The MRMP rate also increased across the three stages: 46.74%, 72.63%, and 83.31%, respectively. These results indicate a significant influence of COVID-19 NPIs on the transmission of M. pneumoniae and MRMP. IMPORTANCE: M. pneumoniae is a significant cause of community-acquired pneumonia with a high MRMP rate in China. Various NPIs in the COVID-19 pandemic have impacted the prevalence of M. pneumoniae and could have important implications for M. pneumoniae prevention and control strategies. Our study documented M. pneumoniae and MRMP detection before, during, and post the COVID-19 pandemic in Henan, China, from 2017 to 2024, aiming to investigate the impact of NPIs on M. pneumoniae transmission and continue to monitor the evolution of the epidemiological characteristics of M. pneumoniae infection after the pandemic when restrictive measures are no longer needed.

Background: Gestational diabetes mellitus (GDM) is a serious health concern that affects pregnant women worldwide and can lead to adverse pregnancy outcomes. Early detection of high-risk individuals and the implementation of appropriate treatment can enhance these outcomes. Methods: We conducted a study on a cohort of 3467 pregnant women during their pregnancy, with a total of 5649 clinical and biochemical records collected. We utilized this dataset as our training dataset to develop a web server called GDMPredictor. The GDMPredictor utilizes advanced machine learning techniques to predict the risk of GDM in pregnant women. We also personalize treatment recommendations based on essential biochemical indicators, such as A1MG, BMG, CysC, CO2, TBA, FPG, and CREA. Our assessment of GDMPredictor's effectiveness involved training it on the dataset of 3467 pregnant women and measuring its ability to predict GDM risk using an AUC and auPRC. Results: GDMPredictor demonstrated an impressive level of precision by achieving an AUC score of 0.967. To tailor our treatment recommendations, we use the GDM risk level to identify higher risk candidates who require more intensive care. The GDMPredictor can accept biochemical indicators for predicting the risk of GDM at any period from 1 to 24 weeks, providing healthcare professionals with an intuitive interface to identify high-risk patients and give optimal treatment recommendations. Conclusions: The GDMPredictor presents a valuable asset for clinical practice, with the potential to change the management of GDM in pregnant women. Its high accuracy and efficiency make it a reliable tool for doctors to improve patient outcomes. Early identification of high-risk individuals and tailored treatment can improve maternal and fetal health outcomes http://www.bioinfogenetics.info/GDM/.

Transfemoral amputees need prosthetic devices after amputation surgery, and the interface pressure between the residual limb and prosthetic socket has a significant effect on an amputee's satisfaction and comfort. The purpose of this study was to build a nonlinear finite element model to investigate the interface pressure between the above-knee residual limb and its prosthetic socket. The model was three-dimensional (3D) with consideration of nonlinear boundary conditions. Contact analysis was used to simulate the friction conditions between skin and the socket. The normal stresses up to 80.57 kPa at the distal end of the soft tissue. The longitudinal and circumferential shear stress distributions at the limb-socket interface were also simulated. This study explores the influences of load transfer between trans-femoral residual limb and its prosthetic socket.

Objective: Heat shock protein 70 (Hsp70), a highly conserved cellular stress protein, is produced in every organism from bacteria to man. The purpose of this study was to determine the difference in Hsp70 concentrations between term and preterm deliveries. Methods: In total, 30 healthy term delivery and 99 preterm delivery (PD) women were recruited, including 46 women with preterm labor and intact membranes (PTL) with or without an intra-amniotic infection (IAI) and 53 women with a preterm premature rupture of membranes (PPROMs) with or without IAI. The Hsp70 levels in the maternal and the umbilical cord sera were tested by an enzyme-linked immunosorbent assay (ELISA); the expression levels of Hsp70 in the placentas were determined by immunoblotting and real-time PCR. Results: In contrast to the expression levels in normal term controls, Hsp70 expression levels were upregulated in PD; similar changes in Hsp70 expression levels were detected in PD maternal and umbilical sera. Patients with IAI were associated with a higher Hsp70 concentration than those without IAI. Conclusion: These results suggest a probable contributing role of altered Hsp70 expression levels in the pathophysiology of PD.
