Natural bioactives are mixtures of compounds extracted from plants with physicochemical properties that are usually not favorable for penetrating the skin’s complex barrier. Nanoparticles have important advantages both in dermatology and cosmetology: improved solubility and stability of encapsulated phytocompounds, controlled and sustained skin delivery, and enhanced skin permeation, leading to an improved bioavailability. This review focuses on two generations of lipid-based nanoparticles: solid lipid nanoparticles (SLNs) and nanostructured lipid carriers (NLCs). An extensive overview on the recent studies on SLNs and NLCs entrapping essential oils, oils, herbal extracts, and phytocompounds for topical applications is presented, emphasizing their composition, physicochemical characterization, efficacy, and methodologies used to evaluate them. This review also summarizes topical systems containing natural bioactives incorporated into SLNs and NLCs, commercially available products and registered patents in the field. SLNs and NLCs turn out to be effective nanocarriers for skin applications, offering significantly improved encapsulation efficiency, stability, and bioactives delivery. However, their full potential is underexplored. Future applications should study the encapsulation potential of new natural bioactives and show more specialized solutions that address specific requirements; an improved product performance and a pleasant sensory profile could lead to increased customer compliance with the product use.

The need to develop wound healing preparations is a pressing challenge given the limitations of the current treatment and the rising prevalence of impaired healing wounds. Although herbal extracts have been used for many years to treat skin disorders, due to their wound healing, anti-inflammatory, antimicrobial, and antioxidant effects, their efficacy can be questionable because of their poor bioavailability and stability issues. Nanotechnology offers an opportunity to revolutionize wound healing therapies by including herbal compounds in nanosystems. Particularly, vesicular nanosystems exhibit beneficial properties, such as biocompatibility, targeted and sustained delivery capacity, and increased phytocompounds’ bioavailability and protection, conferring them a great potential for future applications in wound care. This review summarizes the beneficial effects of phytocompounds in wound healing and emphasizes the advantages of their entrapment in vesicular nanosystems. Different types of lipid nanocarriers are presented (liposomes, niosomes, transferosomes, ethosomes, cubosomes, and their derivates’ systems), highlighting their applications as carriers for phytocompounds in wound care, with the presentation of the state-of-art in this field. The methods of preparation, characterization, and evaluation are also described, underlining the properties that ensure good in vitro and in vivo performance. Finally, future directions of topical systems in which vesicle-bearing herbal extracts or phytocompounds can be incorporated are pointed out, as their development is emerging as a promising strategy.

This study aimed to optimize the extraction of phytocompounds intended for wound care applications from three plant species, Sambucus nigra L. flowers and Epilobium hirsutum L. and Lythrum salicaria L. aerial parts, by using a Quality by Design approach. The effects of different extraction methods (ultra-turrax and ultrasonic-assisted extraction), ethanol concentrations (30%, 50%, 70%), and extraction times (3, 5, 10 min) were studied, and during the optimization step, the polyphenol and flavonoid contents were maximized. The phytochemical profiles of the optimized HEs (herbal extracts) were assessed using LC-MS/MS methods. The antioxidant capacity of the optimized HEs was determined using DPPH (2,2-diphenyl-1-picrylhydrazyl radical scavenging capacity) TEAC (Trolox equivalent antioxidant capacity), and FRAP (ferric reducing antioxidant power) assays, while the antibacterial activity was evaluated against Escherichia coli, Pseudomonas aeruginosa, and MSSA—methicillin-sensitive Staphylococcus aureus and MRSA—methicillin-resistant Staphylococcus aureus). Cell viability and antioxidant and wound healing potential were assessed on keratinocytes and fibroblasts. The anti-inflammatory effect was assessed on fibroblasts by measuring levels of interleukins IL-6 and IL-8 and the production of nitric oxide from RAW 264.7 cells. The major compounds of the optimized HEs were rutin and chlorogenic acid. The Lythrum salicaria optimized HE showed the strongest antibacterial activity, while the Sambucus nigra optimized HE demonstrated high cell viability. Lythrum salicaria and Epilobium hirsutum optimized HEs showed increased antioxidant capacities. All extracts displayed anti-inflammatory effects, and the Epilobium hirsutum optimized HE exhibited the best in vitro wound-healing effect.

Despite the potential of film dressings for wound healing, many formulations lack an optimized design in order to ensure that the ingredients were carefully chosen to increase the product’s efficacy and stability, while also ensuring the patient’s comfort during the treatment. Moreover, commercially available film dressings do not contain herbal extracts or other active substances with wound healing properties, highlighting a gap in the market and the need for further research in this direction. The aim of this work was the development and optimization of a bio-inspired formulation of a complex herbal extract-loaded film-dressing to be used in wound care, using the quality by design approach. After setting the quality target product profile with the critical quality attributes and undergoing the risk assessment, the design of experiments was implemented. All the selected ingredients were biodegradable, aligning with the current need for a natural approach, based on their biocompatibility and reduced environmental impact. A D-optimal experimental plan was used, in which the types and concentrations of film-forming agents and plasticizers were varied: xanthan gum, acacia gum, sodium carboxymethylcellulose and glycerol, 1,3-propanediol, and xylitol, respectively. All formulations contained polyvinyl alcohol and a previously studied complex herbal extract. The films were characterized in terms of uniformity of mass, film thickness, swelling degree, folding endurance, adhesive, and mechanical properties. The optimized formulation was achieved by maximizing the swelling degree, adhesive properties, hardness, deformation at target, and elongation at break. The optimized film was characterized, and the in vitro total polyphenolic content release from the film was evaluated. Following the understanding of the influences of the formulation factors on the film characteristics, the composition of the optimized film-dressing was determined as follows: 5% polyvinyl alcohol, 0.25% xanthan gum, 10% glycerol, and 20% complex herbal extract. The optimized film exhibited high swelling degree (627.28%), high adhesive properties (adhesive force of 28.00 g and adhesiveness of 0.20 mJ), high elasticity (deformation at target of 29.80%, and elongation at break of 106.90%), as well as good mechanical properties (hardness of 2616.00 g), which are suitable characteristics for use on wounds. Moreover, the optimized film-dressing exhibited a sustained release, with a maximum release of polyphenols of 88.00% after 8 h.

Marine macroalgae, classified into three major groups, brown (Phaeophyceae), red (Rhodophyta), and green (Chlorophyta), represent a source of structurally diverse compounds relevant for topical applications. This narrative review of the peer-reviewed literature and regulatory databases targets macroalgae-derived active ingredients in cosmetic formulations and in wound-healing applications. It outlines major compound classes (polyphenols, sulfated polysaccharides, carotenoids, fatty acids, and peptides), along with their documented biological effects on skin (antioxidant, anti-inflammatory, moisturizing, photoprotective, and anti-aging activity) and regulatory/safety aspects with formulation strategies. This review also addresses the variability in compound concentrations resulting from species, environmental conditions, and seasonal factors, which impacts reproducibility and standardization. Common extraction techniques like solvent extraction, ultrasound-assisted extraction, supercritical fluid extraction, and enzyme-assisted methods are described in relation to compound class and yield. Analytical methods used for the identification and quantification of these compounds, including HPLC, GC-MS, and FTIR, are then summarized. Additionally, recent in vitro and in vivo studies evaluating the bioactivity and safety of macroalgae-derived ingredients are discussed. This review compiles relevant evidence to inform formulation strategies and ingredient evaluation in the context of marine-based topical products.

Cleansing products, particularly innovative cosmetic foams, must efficiently remove impurities with minimal impact on the skin barrier and have a favorable sensory profile. The choice of product ingredients is crucial to ensure the optimal characteristics. The current study aims to provide a comprehensive framework for understanding the variability in the characteristics of a cleansing foam to achieve desired properties. The novelty of this study lies in the combination of ingredients for their potential synergistic and complementary effects in cleansing dry skin, as well as the application of Quality by Design (QbD) elements to develop and optimize the formulation of cleansing foam. The effects of varying the concentration of mild surfactants, polyols, and gel-forming agents on the properties of the gels and of the generated foams were studied. Significant influences of the formulation factors were observed: an increased ratio of xanthan gum positively impacted the texture properties of the gel, whereas higher concentrations of surfactants had a negative impact on these parameters. Additionally, increasing the polyols ratio was found to negatively influence the foaming property and stability of the foam. The study established an optimal formulation of a cleansing foam with a ratio of 0.45% xanthan gum, 26.19% surfactants and 2.16% polyols to be used for dry skin hygiene.
