The silkworm Bombyx mori is an economically important species. White muscardine caused by Beauveria bassiana is the main fungal disease in sericulture, and understanding the silkworm responses to B. bassiana infection is of particular interest. Herein, we investigated the molecular mechanisms underlying these responses in two silkworm strains Haoyue (HY, sensitive to B. bassiana) and Kang 8 (K8, resistant to B. bassiana) using an RNA-seq approach. For each strain, three biological replicates for immersion treatment, two replicates for injection treatment and three untreated controls were collected to generate 16 libraries for sequencing. Differentially expressed genes (DEGs) between treated samples and untreated controls, and between the two silkworm strains, were identified. DEGs and the enriched Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways of the two strains exhibited an obvious difference. Several genes encoding cuticle proteins, serine proteinase inhibitors (SPI) and antimicrobial peptides (AMP) and the drug metabolism pathway involved in toxin detoxification were considered to be related to the resistance of K8 to B. bassiana. These results revealed insight into the resistance and susceptibility of two silkworm strains against B. bassiana infection and provided a roadmap for silkworm molecular breeding to enhance its resistance to B. bassiana.

Bombyx batryticatus is a well-known animal in traditional Chinese medicine. The aim of the research was to reveal the quality formation mechanism of B. batryticatus and to screen out the characteristic component used for the quality control. The anticonvulsant effects of B. batryticatus with a stiff time of one, five, and nine days (D1, D5 and D9, respectively) and healthy silkworm of the same developmental stage (SW) were determined by animal experiment. The dynamic changes in chemical composition were analyzed using UPLC-Q-TOF-MS-based metabolomics. D5 and D9 B. batryticatus exhibited significant anticonvulsant effects (p < 0.05 and p < 0.01, respectively). Accordingly, principal component analysis (PCA) and partial least squares discrimination analysis (PLS-DA) indicated that the chemical composition of D5 and D9 B. batryticatus changed significantly. The different metabolites mainly consisted of primary metabolites such as lipids and amino acids and secondary metabolites such as flavonoids, beauvericin, and glycolipids. Interestingly, the relative abundance of quercetin-7-O-β-d-4-O-methylglucoside, the characteristic component of B. batryticatus, increased with stiff time and was promised to be used as an index component of quality control. The results expand our understanding of the quality formation mechanism of B. batryticatus. In addition, it highlights the potential of UPLC-Q-TOF-MS-based metabolomics for the quality control purpose of TCMs.

The pupal diapause of univoltine Antheraea pernyi hampers sericultural and biotechnological applications, which requires a high eclosion incidence after artificial diapause termination to ensure production of enough eggs. The effect of pupal diapause termination using 20-hydroxyecdysone (20E) on the eclosion incidence has not been well-documented in A. pernyi. Here, the dosage of injected 20E was optimized to efficiently terminate pupal diapause of A. pernyi, showing that inappropriate dosage of 20E can cause pupal lethality and a low eclosion incidence. The optimal ratio of 20E to 1-month-old pupae was determined as 6 μg/g. Morphological changes showed visible tissue dissociation at 3 days post-injection (dpi) and eye pigmentation at 5 dpi. Comprehensive transcriptome analysis identified 1,355/1,592, 494/203, 584/297, and 1,238/1,404 upregulated and downregulated genes at 1, 3, 6, and 9 dpi, respectively. The 117 genes enriched in the information processing pathways of “signal transduction” and “signaling molecules and interaction” were upregulated at 1 and 3 dpi, including the genes involved in FOXO signaling pathway. One chitinase, three trehalase, and five cathepsin genes related to energy metabolism and tissue dissociation showed high expression levels at the early stage, which were different from the upregulated expression of four other chitinase genes at the later stage. Simultaneously, the expression of several genes involved in molting hormone biosynthesis was also activated between 1 and 3 dpi. qRT-PCR further verified the expression patterns of two ecdysone receptor genes (EcRB1 and USP) and four downstream response genes (E93, Br-C, βFTZ-F1, and cathepsin L) at the pupal and pharate stages, respectively. Taken together, these genes serve as a resource for unraveling the mechanism underlying pupal-adult transition; these findings facilitate rearing of larvae more than once a year and biotechnological development through efficient termination of pupal diapause in A. pernyi in approximately half a month.

This study examined the effects of dietary silkworm pupae powder (SWP) supplementation on the growth performance, muscle fatty acid composition, and intestinal function in mandarin fish (Siniperca chuatsi) during a 60-day feeding trial. A total of 360 fish (initial body weight 52.44 ± 0.06 g) were distributed randomly into three groups with three replicates per group and 40 fish per replicate. The three groups were fed either the basal diet, supplementation with 1.0 % fermented SWP (T1), or supplementation with 1.0 % fermented induced SWP (T2, induced by Beauveria bassiana). Results showed that fish fed T1 had significantly higher (P < 0.05) condition factor, C16:0, C20:4n-6, C20:5 n3, C22:6n3 C22:5n6, C22:5n-3, and n-3/n-6 levels, but lower (P < 0.05) fat body ratio, C18:1, C18:2n-6 and crude lipid levels than those fish fed basal diet. All fish had similar (P > 0.05) weight gain rate, specific growth rate, feed conversion rate, feed intake, and hepatosomatic index. Fish fed T2 had higher (P < 0.05) trypsin activity than those fish fed basal diet. Compared with the basal diet, fish fed T1 or T2 had higher (P < 0.05) villus length, muscular layer thickness, glutathione peroxidase, alkaline phosphatase, and lysozyme activities, but lower (P < 0.05) triglyceride and total cholesterol levels. Comparative transcriptomic analysis indicated that T1 or T2 enhanced immunity and lipid metabolism ability, as manifested in the up-regulated expression of il12rb1, scdb, tlr2, and FABP7, and the down-regulated expression of EBP and hsd17b7. These findings suggest that dietary supplementation with fermented SWP could enhance the health and quality of mandarin fish, potentially offering a valuable strategy for improving aquaculture productivity.

Chronic heat stress (HS) induces oxidative damage, low immunity, and intestinal flora disturbance of fish, posing great challenges to the aquaculture industry. As an important plant extract, mulberry leaf extract (MLE) has been shown to have antioxidant and immune-boosting properties. This study evaluated the protective effect of dietary MLE on HS-induced liver injury and intestinal flora disturbance in Largemouth bass (Micropterus salmoides) based on oxidative damage parameters, immune parameters, and intestinal flora composition. Fish were randomly assigned into three groups: CON group (ambient temperature, 26°C, basal diet), HS group (33°C, basal diet), and HS + MLE group (33°C, basal diet supplemented with 9 g/kg MLE). HS significantly decreased the final body weight, specific growth rate, intestinal villus length, muscular layer thickness, ACE and Chao1 indices, and lipase, trypsin, total superoxide dismutase (T-SOD), glutathione peroxidase (GPx), alkaline phosphatase (AKP), and lysozyme (LZM) activities, but significantly increased the feed conversion rate, aspartate aminotransferase (AST) activity, reactive oxygen species (ROS), malondialdehyde (MDA), complement 3 (C3) contents, and the relative abundances of Proteobacteria and Plesiomonas. Compared with the HS group, dietary MLE significantly improved lipase, trypsin, GPx, ACP, and LZM activities, and ACE and Chao1 indices in intestine, but significantly decreased the numbers of vacuoles and inflammatory cells, AST activity, and the GLU, MDA, and ROS contents. Dietary MLE also significantly up-regulated the mRNA expressions of gpx, tgf-β, il-10, jnk2, and bcl-2, but down-regulated the mRNA expressions of il-8, tnf-α, caspase-3, atf4, chop, ire1, traf2, jnk1, TRPV, MCU, and VDAC in liver. In conclusion, dietary supplementation with 9 g/kg MLE could improve the growth, immunity, and diversity of intestinal flora in heat-stressed largemouth bass, and alleviate hepatic injuries by regulating inflammation, oxidative stress, and apoptosis, providing a theoretical basis for the development of MLE as a treatment against HS.

Beauveria bassiana (B. bassiana) is a broad-spectrum entomopathogenic fungus that can control pests in agriculture and forestry. In this study, encoding ecdysteroid uridine diphosphate glucosyltransferase gene (egt) was successfully screened in B. bassiana on the medium containing 500μg/mL G418 sulfate solution through the protoplast transformation method. This enzyme has the function of 20E (20-hydroxyecdysone) inactivation, thus increasing the mortality of the early instar larvae infected with B. bassiana. In this study, we transformed B. bassiana with the egt gene, which deactivates 20-hydroxyecdysone, a key hormone in insect development. The results showed that transgenic B. bassiana killed more silkworms of the 2nd instar larvae than the wild-type with a shorter LT50 time, which was reduced by approximately 20% (day 1 of the 2nd instar silkworm infection of B. bassiana) and 26.4% (day 2 of the 2nd instar silkworm infection of B. bassiana) compared to the wild-type, and also showed a higher mortality number before molting. The transgenic B. bassiana had a higher coverage of the body surface of silkworms compared to the wild type on the 3rd instar. In summary, improving entomopathogenic fungi using biological methods such as genetic engineering is feasible.
