To better understand the genetic control of resin yield, growth traits and morphologic traits for Pinus elliottii families, genetic relationships among these traits were examined in three 27-year-old progeny trials located in Jingdezhen, Jian and Ganzhou, Jiangxi Province, China. In total, 3695 trees from 112 families were assessed at the three sites. Significant site, family and family × site effects were found for resin yield, growth traits and morphologic traits. Resin yield and growth traits were found to be under moderate genetic control for the three sites combined, with family heritability and individual narrow-sense heritability ranging from 0.41 to 0.55 and 0.11 to 0.27, respectively. The coefficient of genotypic variation (CVG) of stem volume (SV) and crown surface area (CSA) were higher than those of other traits at each site. Genetic correlation estimates indicated that selection for growth traits might lead to a large increment in resin yield (RY), and most morphologic traits had moderate to strong correlations with growth traits at each individual site. One possible strategy in tree breeding would be to maximize resin production through selection for growth traits.

Slash pine (Pinus elliottii Engelmann) is a pine species widely cultivated for its high oleoresin production capacity. However, little is known about the underlying molecular mechanism of oleoresin biosynthesis between high and low oleoresin-yielding slash pines. In this study, the terpenoid compositions of oleoresin harvested from high- and low-yielding slash pines were identified using gas chromatography/mass spectrometry (GC-MS) analysis. The monoterpenes and diterpenes are the major constituents, of which the α- and β-pinenes are the overwhelming majority of turpentines, and abietic acid, levopimaric acid, and neoabietic acid are the most abundant in rosin. The transcriptomic analysis was also performed with secondary xylem tissues of high- and low-yielding slash pines. After functional annotation, the DEGs of RNA-seq data between high- and low-yielding pines in April, July, and October were screened, and many key enzyme genes were found to be implicated in terpenoid backbone biosynthesis. Moreover, weighted gene correlation network analysis (WGCNA) was carried out to uncover the gene modules highly related to α- and β-pinene biosynthesis in slash pine. Twenty-three modules were attained in this study. Focusing on the total oleoresin yield, the MEblue module exhibited the highest positive correlation, while the MEgreen module exhibited the highest negative correlation. A total of 20 TFs were identified in gene modules. Among these genes, the c215396.graph_c0 encoding an MYB TF is the key differentially expressed gene (DEG) between high- and low-yielding pines. The subsequent one-hybrid yeast assay verified that c215396.graph_c0 can activate the transcription of Apetala 2 (AP2) and 1-deoxy-d-xylulose 5-phosphate synthase (dxs), which are also two DEGs between high- and low-yielding pines. Thus, our study identified a set of key enzymes and TFs that are involved in regulating oleoresin and composition between high- and low-yielding slash pines and provided us a deep insight into oleoresin biosynthesis.

A firm understanding of the early selection efficiency of ring width and wood quality traits is a prerequisite for breeding of higher growth rates and wood quality of slash pine (Pinus elliottii Engelm.). To accomplish effective early selection for the studied traits, wood core samples were collected from 360 trees of 20 open-pollinated families from three slash pine rotation-aged (harvest age) genetic trials. Wood cores were measured by SilviScan, and the annual patterning, heritability, age-age genetic correlation and efficiency of early selection for ring width, modulus of elasticity (MOE), microfibril angle (MFA) and wood density for early to reference age (age 27) were investigated. The greatest annual ring width was observed at age 4, and this trait decreased gradually toward the bark. Different radial trends were found for MOE and MFA. Wood density was lowest near the pith and then increased steadily with age. Overall, heritability was moderate to high for all area-weighted wood quality traits. Very high age-age genetic correlation values were observed for all traits researched. Estimates of efficiency for early selection indicated that the optimal age of early selection for ring width was 9–12 years, while that for area-weighted MOE, MFA and wood density was 7–8 years.

Plants do need a fixed proportion of carbon (C), nitrogen (N) and phosphorus (P) elements to maintain normal metabolic activities. The P fertilizers are widely used to supplement nutrients in subtropical plantations. Stoichiometric homeostasis reflects the strategy of plants to cope with various environments (including P fertilizer supply rate). It is thus of great significance to understand C:N:P stoichiometry in the plant-litter-soil system under P addition and stoichiometric homeostasis of plant tissues for fertilization management of slash pine (Pinus elliottii Engelm) plantations. We measured the C, N and P contents in root, branch, needle, litter and soil in slash pine plantations fertilized with four treatments - P1 (25 kg P ha−1 yr−1), P2 (50 kg P ha−1 yr−1), P3 (100 kg P ha−1 yr−1), and a control (CK) in subtropical China and calculated stoichiometric homeostasis of plant tissues. Results showed that P addition increased the capacity of needle to obtain C, N and P elements and altered the C:N:P stoichiometry of plant tissues, as well promoted the accumulation of C and P elements in soil, but had no significant effect on soil stoichiometry. The nutrient contents of needle and branch were higher than those of root and litter, indicating that slash pine was more inclined to allocate nutrient to the aboveground tissues. The stoichiometric homeostasis of C, N and P among plant tissues was ranked as follows: root > branch > needle, and homeostasis of nutrient elements in the needle was C > N > P. This indicated that the C, N and P stoichiometric homeostasis was various among plant tissues and elements types under P addition. These findings suggest P addition would alleviate the P limitation of slash pine growth in subtropical regions. In the future, long-term observation experiments should be conducted and the trade-off between P addition rates and economic and ecological benefits should be considered.

This study aimed to compare and assess the genetic diversity and trends among the introduced family provenance, first-cycle superior trees breeding provenance, and improved-generation superior trees breeding provenance of Pinus elliottii using EST-SSR markers. The goal was to provide a foundation for advanced genetic improvement and sustainable utilization of P. elliottii in Jiangxi Province. A total of 417 individuals were analyzed for their genetic diversity and population structure using 19 pairs of SSR markers. The analysis identified 103 alleles across all the samples, with an average of 5.421 alleles per locus. Compared to other coniferous species, P. elliottii exhibited a moderate to high level of genetic diversity (I = 0.862, He = 0.457). Analysis of the molecular variance (AMOVA) revealed that 97.90% of the genetic variation occurred within provenances, consistent with a low genetic differentiation coefficient (Fst = 0.016 < 0.05) and high gene flow (Nm = 15.715) among provenances. In addition, analysis using STRUCTURE v. 2.3.4 software divided the 417 germplasm samples into two distinct groups, corroborating the results of the principal coordinates analysis (PCoA) and the unweighted pair group method with arithmetic (UPGMA) clustering analysis. Overall, the germplasm resources of P. elliottii exhibited rich genetic diversity, with the majority of the genetic variation occurring within provenances. For the genetic improvement of high-resin-yielding slash pines, breeding programs should prioritize populations with high genetic diversity while carefully selecting superior individuals from within those populations. These findings provide a solid foundation for breeding high-resin-yielding varieties and for future research on the sustainable utilization of these valuable resources.
