Oat is a food and forage crop species widely cultivated worldwide, and it is also an important forage grass in plateau regions of China, where there is a high level of ultraviolet radiation and sunlight. Screening suitable reference genes for oat under UV-B and high-light stresses is a prerequisite for ensuring the accuracy of real-time quantitative PCR (qRT–PCR) data used in plant adaptation research. In this study, eight candidate reference genes (sulfite oxidase, SUOX; victorin binding protein, VBP; actin-encoding, Actin1; protein PSK SIMULATOR 1-like, PSKS1; TATA-binding protein 2-like, TBP2; ubiquitin-conjugating enzyme E2, UBC2; elongation factor 1-alpha, EF1-α; glyceraldehyde-3-phosphate dehydrogenase 1, GAPDH1;) were selected based on previous studies and our oat transcriptome data. The expression stability of these reference genes in oat roots, stems, and leaves under UV-B and high-light stresses was first calculated using three frequently used statistical software (geNorm, NormFinder, and BestKeeper), and then the comprehensive stability of these genes was evaluated using RefFinder. The results showed that the most stably expressed reference genes in the roots, stems, and leaves of oat under UV-B stress were EF1-α, TBP2, and PSKS1, respectively; the most stably expressed reference genes in the roots, stems, and leaves under high-light stress were PSKS1, UBC2, and PSKS1, respectively. PSKS1 was the most stably expressed reference gene in all the samples. The reliability of the selected reference genes was further validated by analysis of the expression of the phenylalanine ammonia-lyase (PAL) gene. This study highlights reference genes for accurate quantitative analysis of gene expression in different tissues of oat under UV-B and high-light stresses.

The fodder soybean (Glycine max) is an excellent leguminous forage with a high protein content and hay yield, cultivated comprehensively in alpine regions, but seasonal drought in northern regions severely impacts the growth of seedlings. Melatonin (MT) and strigolactone (SL) are critical in relieving the restraint of plant growth in water-deficient environments, but the mechanisms of MT- and SL-mediated drought resistance in fodder soybean needs to be explored. This study mainly investigated the potential morphophysiological mechanism of MT and SL treatments in protecting fodder soybean from drought stress. The fodder soybean ‘Gongnong 535’ was treated with 100 µM MT or 1 µM SL under normal, moderate, and severe water deficit conditions. The results showed that MT and SL treatments enhanced the plant growth parameters and stomatal aperture under drought stress. Moreover, the observed reductions in superoxide ion (O2.-), malondialdehyde (MDA), and relative electrical conductivity (REC), along with enhancements in the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), as well as higher levels of ascorbate (AsA), glutathione (GSH), soluble sugar (SS), soluble protein (SP), and free proline (Pro), indicated that MT and SL application effectively alleviated the oxidative damage and prevented the cell membrane disruption caused by drought stress. Additionally, MT and SL treatments improved photosynthesis and growth in fodder soybean seedlings under water stress by adjusting chlorophyll pigments, gas exchange indexes, and chlorophyll fluorescence parameters, as well as endogenous hormone levels. Simultaneously, MT and SL influenced the expression of genes associated with photosynthesis and antioxidant defenses, as well as phytohormone concentrations. Notably, the protective effect of the SL treatment was superior to that of MT in water-deficient conditions. This study contributes to further understanding the defensive mechanism of MT and SL against drought stress.

Caucasian clover (Trifolium ambiguum M. Bieb.) is an excellent perennial plant in the legume family Fabaceae, with a well-developed rhizome and strong clonal growth. Auxin is one of the most important phytohormones in plants and plays an important role in plant growth and development. Auxin response factor (ARF) can regulate the expression of auxin-responsive genes, thus participating in multiple pathways of auxin transduction signaling in a synergistic manner. No genomic database has been established for Caucasian clover. In this study, 71 TaARF genes were identified through a transcriptomic database of Caucasian clover rhizome development. Phylogenetic analysis grouped the TaARFs into six (1–6) clades. Thirty TaARFs contained a complete ARF structure, including three relatively conserved regions. Physical and chemical property analysis revealed that TaARFs are unstable and hydrophilic proteins. We also analyzed the expression pattern of TaARFs in different tissues (taproot, horizontal rhizome, swelling of taproot, rhizome bud and rhizome bud tip). Quantitative real–time RT–PCR revealed that all TaARFs were responsive to phytohormones (indole-3-acetic acid, gibberellic acid, abscisic acid and methyl jasmonate) in roots, stems and leaves. These results helped elucidate the role of ARFs in responses to different hormone treatments in Caucasian clover.

Carex meyeriana lowland meadow is an important component of natural grasslands in Hulun Buir. However, in Hulun Buir, fewer studies have been conducted on C. meyeriana lowland meadows than on other grassland types. To determine the most appropriate utilization mode for C. meyeriana lowland meadows, an experiment was conducted in Zhalantun city, Hulun Buir. Unused, moderately grazed, heavily grazed and mowed meadow sites were selected as the research objects. The analysis of experimental data from 4 consecutive years showed that relative to the other utilization modes, mowing and moderate grazing significantly increased C. meyeriana biomass. Compared with non-utilization, the other three utilization modes resulted in a higher plant diversity, and the moderately grazed meadow had the highest plant community stability. Moreover, principal component analysis (PCA) showed that among the meadow sites, the mowed meadow had the most stable plant community and soil physicochemical properties. Structural equation modeling (SEM) showed that grazing pressure was less than 0.25 hm2/sheep unit and that plant biomass in C. meyeriana lowland meadow increases with increasing grazing intensity, temperature and precipitation.

Crop productivity around the world is largely constrained by salt-induced stress, a key abiotic factor. Although oat (Avena sativa L.) can withstand challenging environmental conditions, the physiological and molecular responses underlying salt tolerance during germination and early seedling development remain insufficiently understood. To investigate these responses, 28 oat varieties were evaluated at the germination stage, and two contrasting varieties, the salt-tolerant Mengshi No. 1 (MS) and salt-sensitive Morgan (MG), were selected for detailed analysis under a severe NaCl treatment (300 mM) during early seedling stages. Under severe salt stress, the two oat varieties exhibited distinct growth and physiological responses, including changes in growth traits, chlorophyll content, membrane stability, osmotic adjustment, and antioxidant responses. Transcriptomic analysis revealed 14,109 differentially expressed genes (DEGs) between salt-treated MG and its respective control (CK), 19,405 between salt-treated MS and its CK, and 6161 between salt-treated MG and salt-treated MS, suggesting different transcriptional response patterns between the salt-tolerant and salt-sensitive varieties under severe salt stress. Weighted gene co-expression network analysis (WGCNA) revealed a salt-responsive module associated with MS, from which five hub genes, AVESA.00010b.r2.1CG0087930 (MGL), AVESA.00010b.r2.19DG0180280 (MGL), AVESA.00010b.r2.4CG1272260 (BCH1), AVESA.00010b.r2.5DG0989800 (GPAT7), and AVESA.00010b.r2.6CG1124100 (TPR10), were identified as candidate genes potentially associated with salt tolerance and stress responses.
