While chemotherapy significantly improves the prognosis of breast cancer patients, it also damages otherwise healthy organs, such as the ovaries. Gonadotropin-releasing hormone (GnRH) agonists may have a protective effect against chemotherapy-induced ovarian toxicity in premenopausal women being treated for breast cancer; however, studies of its clinical efficacy have reported conflicting results. This meta-analysis was designed to assess the collective data from previous studies of GnRH agonists administered concurrently with chemotherapy to prevent chemotherapy-induced ovarian toxicity in premenopausal women with breast cancer. Electronic literature databases (Cochrane Library, Medline, and Embase) were searched for relevant randomized controlled trials (RCTs) published prior to April 2012. Only RCTs that compared GnRH agonists plus chemotherapy to chemotherapy alone for premenopausal women with breast cancer were selected. A random-effects model was used to calculate the risk ratios (RRs) for premature ovarian failure (POF) within one year after chemotherapy treatment and rates of resumed menses and spontaneous pregnancy during the follow-up period after cessation of treatment. Five RCTs composed of 528 patients (GnRH agonist combination, n = 274; chemotherapy alone, n = 254) were included in the meta-analysis. Significantly fewer women treated with GnRH agonist experienced post-chemotherapy POF, yielding a RR of 0.40 (vs. chemotherapy alone, 95% confidence interval [CI] 0.21-0.75). In contrast, both treatment groups experienced similar rates of resumed menses (RR = 1.31, 95% CI 0.93-1.85) and spontaneous pregnancy (RR = 0.96, 95% CI 0.20-4.56). Concurrent administration of GnRH agonists during chemotherapy treatment of breast cancer in premenopausal women appears to protect against chemotherapy-related POF in the first year after treatment, but appears to have no effect on resumed menses or spontaneous pregnancy rates.

Cardiovascular diseases arc common diseases in Sweden as in most countries. In 2016, 25,700 persons suffered from coronary heart disease (CHD) and 25% of these died within 28 days. The present study investigated whether dioscin may exert protective effects against CHD-induced heart apoptosis, oxidative stress and inflammation in a pig model and the potential underlying mechanisms. Adult pigs were used to establish a CHD model group and 80 mg/kg dioscin was administered for 4 weeks. Histological analysis and measurement of serum levels of heart injury markers demonstrated that 80 mg/kg dioscin markedly alleviated CHD, while left ventricular ejection fraction and left ventricular systolic internal diameter measurements indicated that 80 mg/kg dioscin also increased heart function in the CHD pig model. Furthermore, western blotting demonstrated that 80 mg/kg dioscin significantly reduced protein levels of apoptosis markers in the heart of CHD model pigs, including Bcl-2-associated X and caspase-3, potentially via the suppression of poly (ADP-ribose) polymerase 1 (PARP)/p53 expression. Additionally, the results of ELBA and western blotting demonstrated that 80 mg/kg dioscin may reduce oxidative stress and inflammation in CHD model pigs through the promotion of sirtuin 1 (Sirt1)/nuclear factor erythroid 2-related factor 2 (Nrf2) protein expression and the suppression of PARP/p53 and p38 mitogen-activated protein kinase (MAPK) expression. The results of the current study indicate that dioscin may protect against CHD by regulating oxidative stress and inflammation via Sirt1/Nrf2 and p38 MAPK pathways.

To investigate the performance of methylated SEPT9 (mSEPT9) in assessing the surgical therapeutic effect and prognosis of colorectal cancer (CRC). Blood samples before surgery and 1 and 7 days after surgery were obtained from 120 CRC patients, and mSEPT9 and carcinoembryonic antigen (CEA) assays were performed. The mean plasma mSEPT9 level showed 57.6-times and 131.1-times decrease 1 day and 7 days after surgery, respectively. In contrast, mean CEA levels showed 2.51-and 2.70-times decrease 1 and 7 days after surgery. 86.7% of patients can be assessed by mSEPT9 while 44.2% can be assessed by CEA. Positive mSEPT9 detection before surgery correlated with higher risk of death after surgery. mSEPT9 is effective for CRC postsurgical assessment and prognosis prediction.
