Intrathecal methotrexate in children with leukemia is known to cause seizures, dementia, leukoencephalo pathy, and cognitive dysfunction after long-term treatment. To investigate the cognitive dysfunction, male Wistar rats were given multiple intracerebroventricular injections of methotrexate. Its effect on behaviour was tested in the two-compartment conditioned avoidance task and dark–bright arena test. Levels of brain amines in the hippocampal region of the brain were estimated by HPLC. The qualitative and quantitative histopathological changes in the different regions of the hippocampus were studied by cresyl violet staining. Multiple injections (1 or 2 mg/kg) produced convulsions and learning and memory impairment but did not induce anxiolytic activity. They also reduced concentrations of all three brain amines (norepinephrine, dopamine, and serotonin) and the serotonin metabolite 5-hydroxyindoleacetic acid. The CA4 region of the hippocampus was severely affected by intraventricular methotrexate. Disruption of brain monoamines has been proposed as a cause of brain dysfunction from this chemotherapy, and that disruption may in turn involve cytotoxic effects of methotrexate on brain tissue. The outcomes of this study may have therapeutic implications in the management of cancer conditions, particularly in childhood lymphoblastic leukemia.Key words: methotrexate, hippocampus, norepinephrine, dopamine, serotonin, learning and memory.

To assess the effects of sodium valproate on rat sperm morphology, sperm count, motility, and histopathological changes in testis. Male Wistar rats (12 week old) were treated with sodium valpraote and sacrificed at the end of 2(nd), 4(th), 5(th), 7(th), 10(th) and 15(th) week after the last exposure to sodium valproate. Epididymal sperm count, sperm motility, sperm morphology, and histopathology of testes were analyzed. Sperm count and sperm motility were decreased significantly by sodium valproate. The percentage of abnormal sperms increased in a dose-dependent manner. A histopathological study revealed that sodium valproate had caused sloughing of epithelial cells in testes. Sodium valproate causes reversible change in sperm motility, sperm count, morphology, and cytoarchitecture of testes.

To study possible inhibition of oxidative stress and cataract formation by single combined formulation of grape seed extract and Zincovit tablets against sodium selenite-induced age-related cataract in Wistar rat pups. Oxidative stress and consequent cataract formation was induced by subcutaneous administration of a single dose of sodium selenite (10 µmoles/kg) to Wistar rat pups on day 7 post-natally. In experiments designed to inhibit such cataract formation, the pups were pretreated subcutaneously with combined formulation of grape seed extract and Zincovit tablets (40, 80 and 160 mg/kg), one day prior to the administration of selenite and continuing such treatment till day 20, when the experiments were terminated. The extent of tissue damage caused by the selenite was assessed biochemically by measurements of the levels of reduced glutathione, glutathione peroxidase, glucose-6-phosphate dehydrogenase, protein thiol, catalase, superoxide dismutase, malondialdehyde, aldose reductase, sorbitol dehydrogenase and adenosine triphosphate in the isolated lenses. Cataract formation and its prevention were monitored by examining the eye with pen light illumination and subsequent photography of the isolated lenses. Injection of selenite led to a significant loss of lens clarity due to cataract formation. In the group treated with combined formulation of grape seed extract and Zincovit tablets, the formation of cataract was significantly prevented. In the normal and selenite induced senile cataract control group, the levels of lens oxidative stress markers, G6PD and ATP were substantially lower than in the grape seed extract with Zincovit tablets treated group (p < 0.05). Over all, the results suggest that single combined formulation of grape seed extract and Zincovit tablets may offer a prophylactic measure against onset and progression of age- related cataract of human subjects as nutritional food supplement.

This study was aimed to investigate the hepatoprotective potential of dapagliflozin and silymarin alone and in combination to combat carbon tetrachloride (CCl4)-induced hepatotoxicity and the anticipated mechanisms. Thirty female Wistar rats were randomly allocated into five different groups. All the experimental animals except the normal control (Group I) were administered CCl4. Additionally, Groups II, III, IV, and V were treated with gum acacia, silymarin, dapagliflozin, and a combination of dapagliflozin and silymarin, respectively, for 14 days. Dapagliflozin, silymarin alone, and in combination, significantly reduced (p < 0.05) serum levels of ALT, AST, AST:ALT ratio, and total bilirubin compared to CCl4-intoxicated control rats. There was a notable reduction (p < 0.05) observed in the levels of IL-1beta, IL-6, TNF-alpha, nitrites, and 4-hydroxynonenal, accompanied by an elevation in catalase, superoxide dismutase, glutathione peroxidase, nuclear erythroid 2-related factor 2 (Nrf2), and heme oxygenase-1 (HO-1) in liver homogenates of the groups treated with dapagliflozin, silymarin alone, and in combination, as compared to the CCl4-intoxicated control group. Dapagliflozin in combination with silymarin showed a synergistic hepatoprotective effect. Our study reveals the profound hepatoprotective potential of dapagliflozin alone and in combination with silymarin in CCl4-intoxicated Wistar rats by modulating the Nrf2 and HO-1 signaling pathways.

Neuropathologies, such as neuroinflammaging, have arisen as a serious concern for preserving the quality of life due to the global increase in neurodegenerative illnesses. Nowadays, neuronutraceuticals have gained remarkable attention. It is necessary to investigate the bioavailability, off-target effects, and mechanism of action of neuronutraceuticals. To comprehend the comprehensive impact on brain health, well-designed randomized controlled trials testing combinations of neuronutraceuticals are also necessary. Although there is a translational gap between basic and clinical research, the present knowledge of the molecular perspectives of neuroinflammaging and neuronutraceuticals may be able to slow down brain aging and to enhance cognitive performance. The present review also highlights the key emergent issues, such as regulatory and scientific concerns of neuronutraceuticals, including bioavailability, formulation, blood–brain permeability, safety, and efficacy.
