The microbiota-gut-brain axis (MGBA) represents a sophisticated communication network between the brain and the gut, involving immunological, endocrinological, and neural mediators. This bidirectional interaction is facilitated through the vagus nerve, sympathetic and parasympathetic fibers, and is regulated by the hypothalamic-pituitary-adrenal (HPA) axis. Evidence shows that alterations in gut microbiota composition, or dysbiosis, significantly impact neurological disorders (NDs) like anxiety, depression, autism, Parkinson's disease (PD), and Alzheimer's disease (AD). Dysbiosis can affect the central nervous system (CNS) via neuroinflammation and microglial activation, highlighting the importance of the microbiota-gut-brain axis (MGBA) in disease pathogenesis. The microbiota influences the immune system by modulating chemokines and cytokines, impacting neuronal health. Synbiotics have shown promise in treating NDs by enhancing cognitive function and reducing inflammation. The gut microbiota's role in producing neurotransmitters and neuroactive compounds, such as short-chain fatty acids (SCFAs), is critical for CNS homeostasis. Therapeutic interventions targeting the MGBA, including dietary modulation and synbiotic supplementation, offer potential benefits for managing neurodegenerative disorders. However, more in-depth clinical studies are necessary to fully understand and harness the therapeutic potential of the MGBA in neurological health and disease.

A potential therapeutic strategy to treat conditions brought on by the aberrant production of a disease-causing protein is emerging for targeted protein breakdown using the PROTACs technology. Few medications now in use are tiny, component-based and utilize occupancy-driven pharmacology (MOA), which inhibits protein function for a short period of time to temporarily alter it. By utilizing an event-driven MOA, the proteolysis-targeting chimeras (PROTACs) technology introduces a revolutionary tactic. Small-molecule-based heterobifunctional PROTACs hijack the ubiquitin–proteasome system to trigger the degradation of the target protein. The main challenge PROTAC’s development facing now is to find potent, tissue- and cell-specific PROTAC compounds with favorable drug-likeness and standard safety measures. The ways to increase the efficacy and selectivity of PROTACs are the main focus of this review. In this review, we have highlighted the most important discoveries related to the degradation of proteins by PROTACs, new targeted approaches to boost proteolysis’ effectiveness and development, and promising future directions in medicine.

This detailed review disclosed the NF-κB pro-inflammatory gen's activity regulation and explored the therapeutic significance, activation, and inhibition. This study uncovers the structural intricacies of the NF-κB proteins and highlights the key role of SIRT1 in NF-kB signaling pathway regulation. Particularly the Rel Homology Domain (RHD), elucidating interactions and the regulatory mechanisms involving inhibitory proteins like IκB and p100 within the NF-κB signaling cascade. Disruption of the pathway is important in uncontrolled inflammation and immune disorders. This study extensively describes the role connections of canonical and non-canonical signaling pathways of NF-κB with inflammatory and cellular responses. SIRT1 belongs to the class III histone deacetylase, via RelA/p65 deacetylation, it regulates the activity of NF-κB, closely linked with the NAD+/NADH cellular ratio, influencing stress responses, aging processes, gene regulation, and metabolic pathways. This detailed study reveals SIRT1 as a crucial avenue for uncovering the role of imbalanced NF-κB in diabetes, obesity, and atherosclerosis. This study provides valuable knowledge about the therapeutic targets of inflammatory disorders.

Addressing the challenges posed by climate change, surging population, rival demands on land for renewable fuel manufacturing, and adverse soil conditions is crucial for ensuring global food security. Achieving sustainable solutions necessitates the integration of multidisciplinary knowledge, such as materials technology and informatics. The convergence of precision agriculture with nanotechnology and artificial intelligence (AI) offers promising prospects for sustainable food production. Through real-time responsiveness to crop growth using advanced technologies, such as nanotechnology and AI, farmers can optimize resource allocation and make informed decisions. Newer opportunities for sustainable food production arise through the integration of precision agriculture, nanotechnology, and artificial intelligence. This convergence enables farmers to dynamically respond to crop growth variations using advanced techniques. By combining nanotechnology and informatics methods with existing models for nutrient cycling and crop productivity, it becomes possible to enhance critical aspects, such as precision targeting, efficient absorption, effective distribution, optimized nutrient assimilation, and long-term effects on soil microbial communities. This integration offers significant potential for improving agriculture and addressing sustainability challenges in food production. Ultimately, this synergy allows for the development of nanoscale agrochemicals that offer a balance between safety and functionality, ensuring optimal performance in agricultural systems.
