Statins are included in the category of high-frequency prescription drugs, and their use is on an upward trend worldwide. In 2012, the FDA issued a warning about possible cognitive adverse drug reactions (ADRs) related to statins, some of which are listed in the Summary of Product Characteristics, but there are still concerns about their potential risk of psychiatric events. The aim of this research was to investigate spontaneous reports containing psychiatric ADRs associated with statins by analyzing the EudraVigilance (EV) database. From January 2004 to July 2021, a total of 8965 ADRs were reported for the Systems Organ Class (SOC) “psychiatric disorders”, of which 88.64% were registered for atorvastatin (3659), simvastatin (2326) and rosuvastatin (1962). Out of a total of 7947 individual case safety reports (ICSRs) of the 3 statins mentioned above, in 36.3% (2885) of them, statins were considered the only suspected drug, and in 42% (3338), no other co-administered drugs were mentioned. Moreover, insomnia has been reported in 19.3% (1536) of cases, being the most frequent adverse reaction. A disproportionality analysis of psychiatric ADRs was performed. The Reporting Odds Ratio (ROR) and 95% confidence interval (95% CI) were calculated for simvastatin, atorvastatin and rosuvastatin compared with antiplatelets and antihypertensive drugs. The reporting probability for most ADRs of these statins compared to antiplatelets was higher. The reporting probability for insomnia, nightmares and depression produced by statins compared to antihypertensive drugs was also higher. The results of this analysis augment the existing data about a possible correlation between the administration of statins and the occurrence of psychiatric side effects.

Background/Objectives: Worldwide, colon cancer is a major cause of cancer-related mortality, with an increasing incidence influenced by genetic, environmental, and lifestyle factors. Despite advances in diagnosis and personalized treatments, challenges remain in improving patient prognosis, particularly in metastatic colorectal cancer (mCRC). Bevacizumab (BEV), a monoclonal antibody, is widely used in colorectal cancer treatment. This study aimed to analyze adverse events associated with BEV compared with other therapies based on data from the EudraVigilance (EV) database. Methods: A descriptive and disproportionality analysis was conducted on signals reported in the EV database related to BEV. The study included comparisons with other antineoplastic treatments, such as chemotherapy, targeted therapy, and immunotherapy. Patient demographics, severity of adverse drug reactions (ADRs), and distribution patterns were analyzed to assess the safety profile of BEV in colorectal cancer treatment. Results: The majority of the signals for BEV were from patients aged 18–64 years (39.42%) and 65–85 years (34.08%). Hypertension, thromboembolism, proteinuria, and gastrointestinal disorders have been the most frequently reported. Serious ADRs, including gastrointestinal perforations, hemorrhage, and arterial thromboembolism, were observed in 93.74% of Individual Case Safety Reports. BEV was associated with a higher likelihood of vascular and endocrine disorders compared with chemotherapy and other targeted therapies. Immunotherapy was linked to increased immunological ADRs, while BEV demonstrated fewer immune-related toxicities. Conclusions: Continuous monitoring is necessary to optimize patient management, particularly in elderly patients or those with cardiovascular comorbidities. Understanding BEV’s safety profile allows for better personalization of treatment strategies, minimizing risks while enhancing therapeutic outcomes.

Background/Objectives: Microplastics (MPs) are emerging environmental contaminants with potential implications for human health. Experimental studies suggest that MPs accumulate in the liver and promote inflammatory and fibrotic changes, but evidence from human tissues remains limited. This study investigated the presence of suspected MP-like particles in post-mortem human liver tissue and their associations with clinical, biochemical, hematological and histopathological parameters. Methods: Post-mortem liver tissue samples were collected from 55 adults. Suspected MP-like particles were extracted using hydrogen peroxide digestion, filtration, Nile Red staining and image-based quantification. Histopathological evaluation assessed inflammation, fibrosis, necrosis and fatty liver degeneration. Polarized light microscopy was used as a supportive morphological assessment method. Statistical analyses included Mann–Whitney U tests, Fisher’s exact tests and Spearman correlation analysis. Results: Detectable hepatic suspected MP-like particles were identified in 9 of 55 individuals (16.4%). Individuals with detectable suspected MP-like particles had significantly lower alanine aminotransferase (ALT) levels and leukocyte counts. The hepatic suspected MP-like particle burden showed weak positive correlation with liver fibrosis and inflammation and weak inverse correlation with ALT levels and leukocyte count. Fisher’s exact test showed that liver fibrosis and liver inflammation were significantly associated with detectable hepatic suspected MP-like particles, with higher unadjusted odds observed in the corresponding 2 × 2 contingency tables. No statistically significant associations were found for liver necrosis or fatty liver degeneration. Conclusions: Detectable suspected MP-like particles were identified in post-mortem human liver tissues and were more closely associated with fibrotic and inflammatory histopathological changes than with routine biochemical abnormalities. Larger studies using standardized detection methods are needed to confirm these results.

Background: In metastatic colorectal cancer (mCRC), systemic inflammation and routine laboratory parameters may reflect host–tumor interactions and provide additional prognostic information. This study evaluated the association between baseline clinicopathological and laboratory parameters, including the derived neutrophil-to-lymphocyte ratio (dNLR), and overall survival (OS) in patients with stage IV colorectal adenocarcinoma. Methods: We conducted a retrospective cohort study including 92 patients diagnosed with metastatic colorectal adenocarcinoma and treated at a single oncology center between February 2022 and December 2024. Baseline laboratory parameters were collected at diagnosis. Survival was analyzed using Kaplan–Meier estimates with log-rank testing. Prognostic associations were evaluated using univariable and multivariable Cox proportional hazards regression models adjusted for relevant clinical and treatment-related factors. Results: The cohort was predominantly male (62%) and younger than 70 years (66%), with 80 deaths recorded during follow-up. In univariable analyses, primary tumor resection, irinotecan-based first-line therapy, elevated AST, and dNLR tertiles were associated with OS. However, after multivariable adjustment, only irinotecan-based first-line therapy remained independently associated with poorer survival (HR 2.10, 95% CI 1.16–3.81; p = 0.022). Continuous dNLR, anemia (WHO sex-specific), and AST elevation did not retain independent prognostic significance. Conclusions: In this cohort of patients with mCRC, inflammation-related laboratory markers demonstrated associations with survival in unadjusted analyses but did not remain independent predictors after adjustment for clinical and treatment-related confounders. These findings highlight the importance of rigorous multivariable modeling when evaluating inflammatory biomarkers in metastatic colorectal cancer.

Background: Obesity is strongly associated with type 2 diabetes, but clinical and metabolic characteristics vary among adults with obesity. This cross-sectional study evaluated whether routinely available nonglycemic variables could classify biochemically defined diabetes status at the time of evaluation. Methods: This retrospective study included 351 adults with obesity, comprising 185 with diabetes and 166 without diabetes. Diabetes was defined by two fasting plasma glucose measurements ≥ 126 mg/dL obtained on separate occasions or an HbA1c value ≥ 6.5%. Complete numerical fasting glucose and HbA1c values were not retained as continuous variables in the analytical database, and insulin-related measures were unavailable. Multivariable logistic regression and four machine learning classifiers were evaluated. Results: Participants with diabetes were older, more frequently male, and more frequently had hypertension, fatty liver, and prior cardiovascular interventions. They had higher triglyceride and CRP concentrations, lower LDL-C concentrations, and no statistically significant difference in HDL-C. Age, hypertension, fatty liver, and LDL-C were associated with diabetes status in the multivariable model. Gradient Boosting and Random Forest showed similar discrimination (AUC-ROC = 0.807 and 0.803, respectively), with overlapping confidence intervals. Conclusions: The models classified prevalent diabetes status within this cohort using variables other than the glycemic criteria used to define the outcome. They were not designed to predict incident diabetes or progression from obesity to diabetes. The findings are exploratory and require external validation before clinical application.
