The Hippo‑yes‑associated protein (YAP) signaling pathway was previously identified to serve an important role in controlling chondrocyte differentiation and post‑natal growth. Growth plate cartilage tissue is avascular, and hypoxia‑inducible factor (HIF)‑1α is essential for chondrocytes to maintain their chondrogenic phenotype in a hypoxic environment. In the present study, the role of hypoxia and HIF‑1α in the regulation of YAP in chondrocytes was investigated. The data demonstrated that hypoxia promoted the maintenance of the chondrogenic phenotype, HIF‑1α expression and YAP activation in chondrocytes in a time‑dependent manner. Hypoxia promoted YAP activation in a Hippo‑independent manner. Inhibiting the expression of HIF‑1α decreased the activation of YAP and downregulated the expression of sex‑determining region‑box 9 protein (SOX9) under hypoxic conditions, while the upregulation of HIF‑1α by cobalt chloride promoted the expression and nuclear translocation of YAP and upregulated the expression of SOX9 and collagen II chain under normoxic conditions. In addition, inhibition of YAP expression under hypoxia did not affect the expression of the HIF‑1α signaling pathway, but inhibited the up‑regulation of SOX9 expression caused by hypoxia. In addition, reoxygenation following hypoxia inhibited the activation of YAP caused by hypoxia in chondrocytes, whereas the upregulation of SOX9 and collagen II chain also appeared to be inhibited. In conclusion, the results of the present study demonstrated that hypoxia promoted YAP activation via HIF‑1α. Therefore, the HIF‑1α/YAP signaling axis may serve an important role in controlling growth plate chondrocyte differentiation and the maintenance of the chondrogenic phenotype in growth plate chondrocytes.

Lung adenocarcinoma (LUAD) is one of the most commonly malignant tumors, and major challenges remain in the treatment of LUAD. Budding uninhibited by benzimidazole 1/3 (BUB1/3) play significant roles in the process of spindle-assembly checkpoint (SAC) during mitosis. However, their roles in LUAD have not been established. Here, we performed an immunological analysis of BUB1/3 in LUAD using a comprehensive bioinformatics approach, quantitative real-time-PCR and Western blotting technique. Our results indicated that the expression levels of BUB1 and BUB3 in LUAD samples were higher than the expression levels in the control groups and were associated with some clinicopathologic parameters in patients with LUAD. BUB1/3 and their related genes were enriched in cell immune, and the immune infiltration analysis revealed that the BUB1/3 expression profile was significantly correlated with characteristics of immune cell infiltration. Survival analysis showed that the disease-free survival and overall survival of patients with LUAD decreased with an increase in the BUB1/3 expression levels. The mRNA and protein expression levels of BUB1 and BUB3 in each of the LUAD cell lines were upregulated to varying degrees. BUB1 and BUB3 are the potential immunological therapeutic intervention targets for patients with LUAD.

Background: Osteosarcoma is an aggressive malignancy of bone that poses significant treatment challenges and has been a focal point of extensive research due to its complex pathogenesis. Despite advances in traditional therapeutic approaches, the intricate genetic and cellular landscape of osteosarcoma remains inadequately understood, emphasizing the need for innovative research methodologies to unravel its underlying mechanisms. Objective: This study aims to leverage the power of single-cell transcriptome sequencing technology to elucidate the cellular heterogeneity, gene expression patterns, intercellular communication networks, and critical genetic pathways implicated in osteosarcoma. By doing so, we intend to contribute valuable insights into the biogenesis of this malignancy, which may ultimately inform precision treatment strategies. Methods: Utilizing single-cell sequencing, we conducted a comprehensive analysis of osteosarcoma samples to identify diverse cellular subpopulations within the tumor microenvironment. Our focus on gene expression profiles revealed significant differences across these subpopulations. Moreover, we employed bioinformatics approaches to explore the intercellular communication networks and identified key ligand-receptor pairings, substantiating the role of angiogenesis-related genes prominently expressed in osteoblasts and their proliferative counterparts. Results: Our findings underscore the critical involvement of angiogenesis in osteosarcoma pathogenesis, with notable pathway activity variations among distinct cellular subpopulations. Additionally, protein interaction network mapping has unveiled significant discrepancies in pathway activities and highlighted the potential functional roles of key genes involved in tumor progression. Conclusion: This study offers a comprehensive exploration of the biological characteristics of osteosarcoma through single-cell sequencing technology, thereby establishing a robust theoretical foundation that may facilitate the development of targeted and effective therapeutic strategies. However, it is essential to recognize that these findings are preliminary, necessitating further validation through expanded sample sizes and integration of multi-omics data. Future research will delve deeper into the mechanisms of the identified key pathways and genes, with the aspiration of enhancing the prognostic outcomes and quality of life for patients with osteosarcoma.

This case report describes the medical journey of a 9-year-old girl who was diagnosed with Rosai-Dorfman disease (RDD), a rare disorder characterized by the overproduction of histiocytes within the lymph nodes. The patient was diagnosed with RDD after experiencing left calf pain and discomfort. Initial imaging revealed abnormal signals and localized low-density foci in the left tibia, and subsequent MRI scans indicated a possible tumor. The diagnosis was confirmed through a puncture biopsy, which revealed granulomatous tissue consistent with RDD. The patient's treatment focused on immobilizing the affected limb without surgical intervention. Follow-up imaging showed a reduction in the extent of the initial abnormalities and periosteal reaction. This case emphasizes considering Rosai-Dorfman disease (RDD) in pediatric bone lesion diagnoses and highlights the effectiveness of conservative management. The study provides a comprehensive overview of RDD in the skeletal system, including etiology, clinical manifestations, imaging features, diagnostics, and therapeutic strategies, serving as a valuable reference for clinicians and future research.
