Tantalum (Ta) implants fabricated by current processing techniques inevitably contain more or less oxygen impurities due to the extremely high melting point and high affinity of oxygen for Ta. Therefore, in this study we investigated whether oxygen impurities cause any effects on the bioactivity of Ta. EDS analysis demonstrated the surface oxygen content difference among different fabricated Ta samples, and the surface water contact angle (WCA) of Ta with high oxygen content (HO-Ta) was significantly higher than that of Ta with medium (MO-Ta) and low (LO-Ta) oxygen content. The in vitro cellular experiments showed that MC3T3-E1 cells on Ta with lower oxygen content exhibited better adhesion, growth, morphological development and in vitro osteogenic ability. Similarly, the in vivo animal experiments indicated the better bone regeneration and ingrowth performances of Ta with lower oxygen content. In addition, the highest ROS production was detected in the HO-Ta group, while the lowest in the LO-Ta group. This study suggests that the oxygen content within Ta, which occurs unavoidably due to technical limitations, negatively affects the bioactivity of Ta in a dose-dependent manner, indicating the need to develop techniques to produce orthopedic all-Ta implants.

Background: The incidence of insufficiency fracture (IF) at femoral neck is low, accounting for about 5% of all insufficiency fractures, and IF at bilateral femoral neck is less common with more occurrence in athlete or serviceman. With the aging of populations, more cases of bilateral femoral neck IF have occurred recently, while the standard clinical treatment still remains lacking due to the complexity of these patients. Case presentation: A 55-year-old male patient complained pain in his bilateral hip, with no history of trauma, glucocorticoid hormone consumption or radiotherapy, and imaging examination revealed fracture nonunion and shortening in his left femoral neck, and double fracture line on the right femoral neck. The patient received a cementless THA for the left femoral neck fracture and conservative treatment for the right side, followed by Elcatonin injection and oral administration of Carbonate D3 Granules. After 4 months of fellow-up, the patient presented improved functional scorings in bilateral hip joints, with no signs of prothesis infection or loosening. Conclusion: We present a rare case of bilateral femoral neck IF in a middle-aged male and the treatment is successful. The timely CT and MRI examinations of bilateral hip joints for patients was necessary for orthopedists to select proper therapeutic regimen. In addition, the choice for therapeutic regimen of bilateral femoral IF should not only be based on the professional judgement of orthopedists, but also on the wishes of patients.

While the prognostic value of autophagy-related genes (ARGs) in OS patients remains scarcely known, increasing evidence is indicating that autophagy is closely associated with the development and progression of osteosarcoma (OS). Therefore, we explored the prognostic value of ARGs in OS patients and illuminate associated mechanisms in this study. When the OS patients in the training/validation cohort were stratified into high- and low-risk groups according to the risk model established using least absolute shrinkage and selection operator (LASSO) regression analysis, we observed that patients in the low-risk group possessed better prognosis ( ). Univariate/Multivariate COX regression and subgroup analysis demonstrated that the ARGs-based risk model was an independent survival indicator for OS patients. The nomogram incorporating the risk model and clinical features exhibited excellent prognostic accuracy. GO, KEGG, and GSVA analyses collectively indicated that bone development-associated pathway mediated the contribution of ARGs to the malignance of OS. Immune infiltration analysis suggested the potential pivotal role of macrophage in OS. In summary, the risk model based on 12 ARGs possessed potent capacity in predicting the prognosis of OS patients. Our work may assist clinicians to map out more reasonable treatment strategies and facilitate individual-targeted therapy in osteosarcoma.
