A key element in ensuring successful immunization is the efficient delivery of vaccines. However, poor immunogenicity and adverse inflammatory immunogenic reactions make the establishment of an efficient vaccine delivery method a challenging task. The delivery of vaccines has been performed via a variety of delivery methods, including natural-polymer-based carriers that are relatively biocompatible and have low toxicity. The incorporation of adjuvants or antigens into biomaterial-based immunizations has demonstrated better immune response than formulations that just contain the antigen. This system may enable antigen-mediated immunogenicity and shelter and transport the cargo vaccine or antigen to the appropriate target organ. In this regard, this work reviews the recent applications of natural polymer composites from different sources, such as animals, plants, and microbes, in vaccine delivery systems.

Background: Mammary cancer, called breast cancer in humans, results from the abnormal growth of cells in the mammary glands that attack the surrounding tissue. The process of carcinogenesis at the molecular level can be monitored through the production of proteins as biomarkers for carcinogenesis. 7,12-Dimethylbenz[a]anthracene (DMBA) is a known carcinogenic compound. This study aimed to analyze the proteomic profile as critical data regarding DMBA-induced carcinogenesis in Sprague‒Dawley rats. Methods: Experimental animals were divided into two groups: a treatment group given DMBA at a dose of 10 mg/kg (intramammary) at intervals of 48 hours for a total of 10 doses, and a negative control group that was not given any treatment. Measurement of the total protein concentration was carried out using a spectrophotometer, and the data were analyzed using a t-test, while the characterization of protein profiles was carried out based on molecular weight data using SDS‒PAGE. Mammary gland histopathology was evaluated by hematoxylin and eosin (H&E) staining. Results: The results showed a significant (p<0.05) increase of 27% in the total protein concentration in the rat mammary cancer model. The results of proteomic characterization showed a protein profile containing proteins of 187, 169, 68, 64, 53, 41, 24, 18, and 14 kDa, which were suspected to be HER-2, Nischarin, COX-2, Albumine, Vimentin, ACTB, TNF, p16, and fatty acid binding protein 3 (FABP3), respectively. Histopathology of the mammary glands showed an irregular and indistinct arrangement of the alveoli and extensive epithelial cell proliferation from the surface to the lumen of the mammary ducts, and the mammary stroma showed the formation of new epithelial cells, which were cancer cells that spread to surrounding tissue. Conclusions: The proteomic profile was strongly associated with morphological alterations in mammary carcinogenesis in a rat model of DMBA-induced mammary cancer.

Liver has an important role in detoxification of toxins such as xenobiotic which could interfere the function of liver. Chloramphenicol is an antibiotic widely used, despite of its toxicity potentials. The enhancement of free radicals in the body could suppress antioxidant activity. Propolis of Trigona sp. has been known to contain very high amount of antioxidants. The enhanced serum glutamic oxaloacetic transaminase (SGOT) and alkaline phosphatase (ALP) levels in serum is used as marker of liver damage due to the increase of free radicals. The purpose of this study was to observe the effect of Trigona sp. propolis ethanolic extract on SGOT and ALP levels in rats (Rattus novergicus) pretreated by chloramphenicol to induce liver damage. Test animals used for this research were male rats aged 8-12 weeks divided into five treatment groups: negative controlgroup (normal), positive control group (induced by 400 mg/kgBW chloramphenicol), first therapy group, second therapy group, third therapy group induced by chloramphenicol with and propolis extract with the dose of 8 mg, 16 mg, and 24 mg, respectively. Chloramphenicol was injected subcutaneously for 14 days, whereas propolis extract were administered orally for 21 days. The level of SGOT and ALP was determined using spectophotometry. The results showed that propolis extract could reduce levels of SGOT and ALP. Dose of 24 mg/kg was the effective dose to decrease levels of SGOT and ALP significantly (p<0.01). Hence, it may be concluded that the ethanol extract of propolis could be used as herbal therapy in rats model of liver damage. Keywords : ALP, liver, chloramphenicol, propolis, SGOT

Background and Aim: Aeromonas hydrophila infections in fish result in significant financial losses within aquaculture. Previous research indicates black soldier fly (BSF) prepupae provide immunomodulatory benefits through their fatty acids, chitin, and proteins. The study evaluated the impact of hexane extract from black soldier fly prepupae (HEBP) on interleukin (IL)-4 and IL-10 cytokine expression in zebrafish, both infected and uninfected with A. hydrophila. Materials and Methods: Adult zebrafish (aged 4–5 months) was assigned to a negative control group (fed commercial feed), a positive control group (commercial feed + A. hydrophila infection at 107 colony-forming unit/mL), and three treatment groups (T1, T2, T3) that received HEBP at doses of 1000; 2000 and 4000 mg/kg feed for 30 days, respectively. A. hydrophila infection was introduced on day 31 through immersion. Analysis of IL-4 and IL-10 expression in the head kidney trunk region (body without head and tail) through quantitative polymerase chain reaction was conducted on day 33. Results: The HEBP modulated the immune response to A. hydrophila infection at a concentration of 1000 mg/kg feed, as evidenced by an increase in IL-4 and IL-10 expression in the groups not infected with the bacteria. However, these cytokines were decreased in the infected groups. Conclusion: A feed concentration of 1000 mg/kg HEBP was identified as optimal for cytokine modulation. This discovery marks a significant advancement in the development and benefit of a natural extract-based immunomodulator in a zebrafish model, which is potentially immunotherapeutic against bacterial infections in fish for the aquaculture industry. Keywords: Aeromonas hydrophilia, black soldier fly larvae, hexane extract, immune modulation, zebrafish.

Brain cancer is one of the most challenging malignancies and a major contributor to worldwide morbidity and mortality. Glioblastoma, the most aggressive adult brain tumor, is associated with poor prognosis despite conventional therapies such as surgery, chemotherapy, and radiotherapy, which often result in severe toxicity and long-term side effects. Immunotherapy holds the potential to provide durable and specific anti-tumor responses; however, its success in brain cancer is hindered by obstacles such as the blood-brain barrier, immunosuppressive tumor microenvironment, and tumor heterogeneity. Nanomedicine offers a powerful approach to overcoming these barriers through targeted and efficient drug delivery. Nanotechnology-based platforms, including lipid-based, polymeric, and inorganic nanoparticles, have demonstrated superior therapeutic efficacy compared to free drugs, with several formulations advancing into clinical trials. Among these, nanotechnology-enabled vaccines represent an emerging frontier, capable of enhancing antigen presentation, stimulating strong immune responses, and overcoming tumor-induced immunosuppression. By combining the precision of nanocarriers with the long-lasting protection of vaccines, nano-vaccines hold great potential to transform brain cancer immunotherapy. Recent studies also suggest that integrating artificial intelligence (AI) with nanotechnology could further enhance the design, targeting, and effectiveness of immunotherapies. Moreover, AI is revolutionizing treatment development by enabling the prediction of immunogenicity, immune responses, and optimizing formulation design and dosing strategies. These advancements collectively accelerate the development and enhance the precision and efficiency of immunotherapy. Hence, this review discusses current nanotechnology-based immunotherapies and highlights the emerging role of integrating AI in vaccine development as next-generation strategies for improving outcomes in brain cancer treatment.
