Natural competence for genetic transformation in Streptococcus pneumoniae is achieved directly by specific proteins that are involved in DNA uptake and chromosomal recombination, and is regulated indirectly by a quorum‐sensing system encoded by two loci, comAB and comCDE. The alternative sigma factor, ComX, is thought to be the unique link between quorum sensing and competence‐specific genes. To test this hypothesis, we replaced the quorum‐sensing inducible promoter (PQ) of the comX gene with either a constitutive promoter (PC) or a raffinose‐inducible promoter (PR), so that comX transcription would be independent of quorum sensing. Surprisingly, both competence and expression of late genes, such as ssbB, cglA or celB, were found to depend on CSP in these mutants. An unknown, CSP‐dependent regulator was needed when comX was expressed from these ectopic promoters, and it appears to act post‐transcriptionally. However, when a multicopy nisin‐inducible ComX‐overexpressing plasmid was introduced, pneumococcal cells developed competence in the presence of nisin even despite deletion of comE. At 1% of the normal protein peak level, ComX protein stimulated competence without the participation of the pheromone response circuit. Thus, ComX is a unique link to competence‐specific genes, but depends on multiple outputs of quorum sensing for maximal expression.

The problem of soil salinization has seriously hindered agricultural development. Cotton is a pioneering salinity-tolerant crop, so harvesting its key salinity-tolerant genes is important for improving crop salt tolerance. In this study, we analyzed changes in the transcriptome expression profiles of the salt-tolerant cultivar Lu Mian 28 (LM) and the salt-sensitive cultivar Zhong Mian Suo 12 (ZMS) after applying salt stress, and we constructed weighted gene co-expression networks (WGCNA). The results indicated that photosynthesis, amino acid biosynthesis, membrane lipid remodeling, autophagy, and ROS scavenging are key pathways in the salt stress response. Plant–pathogen interactions, plant hormone signal transduction, the mitogen-activated protein kinase (MAPK) signaling pathway, and carotenoid biosynthesis are the regulatory networks associated with these metabolic pathways that confer cotton salt tolerance. The gene-weighted co-expression network was used to screen four modules closely related to traits, identifying 114 transcription factors, including WRKYs, ERFs, NACs, bHLHs, bZIPs, and MYBs, and 11 hub genes. This study provides a reference for acquiring salt-tolerant cotton and abundant genetic resources for molecular breeding.
Drought is one of the most serious environmental stresses affecting crop production. NAC transcription factors play a crucial role in responding to various abiotic stresses in plants. Here, we identified a maize NAC transcription factor, ZmSNAC06, between drought-tolerant and drought-sensitive inbred lines through RNA-seq analysis and characterized its function in Arabidopsis. ZmSNAC06 had five transcripts, of which ZmSNAC06-T02 had a typical NAC domain, while ZmSNAC06-P02 was localized in the nucleus of maize protoplasts and had transactivation activity in yeasts. The expression of ZmSNAC06 in maize was induced by drought. The overexpression of ZmSNAC06-T02 in Arabidopsis resulted in hypersensitivity to abscisic acid (ABA) at the germination stage, and overexpression lines exhibited higher survival rates and higher antioxidant enzyme activities compared with the wild-type under drought stress. These results suggest that ZmSNAC06 acts as a positive regulator in drought tolerance and may be used to improve drought tolerance in crops.
Drought stress is seriously affecting the growth and production of crops, especially when agricultural irrigation still remains quantitatively restricted in some arid and semi-arid areas. The identification of drought-tolerant genes is important for improving the adaptability of maize under stress. Here, we found that a new member of the actin-depolymerizing factor (ADF) family; the ZmADF5 gene was tightly linked with a consensus drought-tolerant quantitative trait locus, and the significantly associated signals were detected through genome wide association analysis. ZmADF5 expression could be induced by osmotic stress and the application of exogenous abscisic acid. Its overexpression in Arabidopsis and maize helped plants to keep a higher survival rate after water-deficit stress, which reduced the stomatal aperture and the water-loss rate, as well as improved clearance of reactive oxygen species. Moreover, seventeen differentially expressed genes were identified as regulated by both drought stress and ZmADF5, four of which were involved in the ABA-dependent drought stress response. ZmADF5-overexpressing plants were also identified as sensitive to ABA during the seed germination and seedling stages. These results suggested that ZmADF5 played an important role in the response to drought stress.
