Phoebe sheareri is an excellent roadside tree with a wide distribution range and high ornamental value. Excessive moisture can affect the external morphology, the microstructure, and the ultrastructure of the leaf. Little is known at present regarding the leaf structure of P. sheareri under waterlogging stress. In this paper, the external morphology of leaves, the microstructure of leaf epidermis, and the ultrastructure of mesophyll cells of P. sheareri seedlings under waterlogging stress and drainage were dynamically observed. Waterlogging stress contributed to the yellowing and wilting of P. sheareri seedling leaves, the gradual closing of leaf epidermal stomata, increasing density of leaf stomata, gradual loosening of the arrangement of leaf cell structure, and merging of leaf palisade tissue cells. Waterlogging stress forced the structure of the chloroplast membranes to blur, gradually causing swelling, and deformation, with plasmolysis occurring in severe cases. During waterlogging, the basal lamellae were disorganized, and the mitochondrial membrane structure was damaged. The damaged state of the leaves was not relieved after drainage. Waterlogging stress not only inhibited the growth of leaves, but also accelerated the closure of stomata, disordered the arrangement of palisade tissue and spongy tissue gradually, and damaged the internal organelles of mesophyll cells.

The seeds of Tilia miqueliana Maxim. exhibit deep dormancy, which is categorized as combinational dormancy. This study utilized a comprehensive treatment involving magnetic fields, gibberellin (GA3), and cold stratification to promote the release of seed physiological dormancy and enhance germination rates. After being soaked in 98% H2SO4 for 15 min, mature seeds of Tilia were exposed to magnetic field treatments (150 MT, 250 MT) for different durations (25 min, 45 min, 65 min, and 85 min), as well as GA3 solution soaking (concentration: 0 μmol·L−1, 1443 μmol·L−1). Subsequently, cold stratification (0–5 °C) was applied to investigate the effects of these treatments on seed dormancy release and nutrient transformation. The results indicated that the comprehensive treatment involving magnetic field, GA3 solution soaking, and cold stratification effectively released the physiological dormancy of Tilia seeds and improved germination rates. Among the treatments, M150T85G1443 (magnetic field intensity: 150 MT, magnetic field treatment time: 85 min, GA3 soaking concentration: 1433 μmol·L−1) exhibited the most favorable outcome. After 75 days of cold stratification following the comprehensive treatments, the germination rate of M150T85G1443 seeds reached 89%. Additionally, the levels of storage substances such as starches and crude fats within the seeds decreased, while the utilization of soluble sugars and soluble proteins increased. The M150T85G1443 treatment exhibited the highest degree of variation, leading to gradual increases in metabolic activities of the seeds and a transition from dormancy to germination.

Machilus thunbergii Siebold & Zucc. is recognized as an excellent tree species for landscaping and shelter forest. Excessive drought can affect the changes of physiological and biochemical substances in plants. However, little is known at present regarding the drought stress of M. thunbergii seedlings. In this paper, matrix water content, the anatomical structure of leaves, relative water content of leaves, and physiological characteristics index of leaves under droughting stress were dynamically observed. Droughting stress led to the wilting of M. thunbergii leaves, gradual closure of stomata on leaf epidermis, increases in stomatal density, gradual loosening of leaf cell structure arrangement, a thickening in leaf palisade tissue, and reductions in spongy tissue. Droughting stress caused the relative water content of the cultivation substrate to decline, the cultivation substrate reached the moderate drought level, and the seedlings began to die. Droughting stress led to the destruction of activity and balance of the leaf protective enzyme system, excessive accumulation of free radicals, the destruction of enzyme structure and function, and the production of lipid peroxidation product MDA. Droughting stress reduced the relative water content of leaves as a whole, the content of osmotic adjustment substances proline and soluble protein continued to decline, and a large number of electrolyte leakage in cells, causing serious damage to seedlings.

Quercus acutissima seeds exhibit high desiccation sensitivity, posing significant challenges for long-term preservation. This study investigates the physiological and metabolic responses of soluble osmoprotectants—particularly soluble proteins and proline—during the desiccation process. Seeds were sampled at three critical moisture content levels: 38.8%, 26.8%, and 14.8%, corresponding to approximately 99%, 52%, and 0% germination, respectively. We measured germination ability, soluble protein content, and proline accumulation, and we performed untargeted metabolomic profiling using LC-MS. Soluble protein levels increased early but declined later during desiccation, while proline levels continuously increased for sustained osmotic adjustment. Metabolomics analysis identified a total of 2802 metabolites, with phenylpropanoids and polyketides (31.12%) and lipids and lipid-like molecules (29.05%) being the most abundant. Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis showed that differentially expressed metabolites were mainly enriched in key pathways such as amino acid metabolism, energy metabolism, and nitrogen metabolism. Notably, most amino acids decreased in content, except for proline, which showed an increasing trend. Tricarboxylic acid cycle intermediates, especially citric acid and isocitric acid, showed significantly decreased levels, indicating energy metabolism imbalance due to uncoordinated consumption without effective replenishment. The reductions in key amino acids such as glutamic acid and aspartic acid further reflected metabolic network disruption. In summary, Q. acutissima seeds fail to establish an effective desiccation tolerance mechanism. The loss of soluble protein-based protection, limited capacity for proline-mediated osmotic regulation, and widespread metabolic disruption collectively lead to irreversible cellular damage. These findings highlight the inherent metabolic vulnerabilities of recalcitrant seeds and suggest potential preservation strategies, such as supplementing critical metabolites (e.g., TCA intermediates) during storage to delay metabolic collapse and mitigate desiccation-induced damage.

By measuring the changes in phytohormones and related enzyme activities of Malus ‘Huabiao’ scions and different rootstocks during the healing process of grafting, this study aims to analyze the effects of different rootstocks on the physiological metabolism of the grafting healing site, thereby providing reference for the selection of M. ‘Huabiao’ grafting rootstocks. The live seedlings of 1-0 M. hupehensis, M. robusta and M. baccata were chosen as rootstocks, and M. ‘Huabiao’ scions were grafted with these seedlings in spring by pasting method. The dynamic changes in activities of indole acetic acid, zeatin riboside, methyl jasmonate content and peroxidase, polyphenol oxidase and L-phenylalanine ammonia-lyase at the healing site of grafting were measured, and the effects of different grafting combinations on the phytohormone and related enzyme activities at the grafted site were investigated. It was found that the M. ‘Huabiao’/M. hupehensis and M. ‘Huabiao’/M. robusta grafting combinations had significantly higher survival rate than M. ‘Huabiao’/M. baccata which only had a success rate of 63%. At the beginning of graft healing, the contents of three phytohormones in healing sites of all the three grafting combinations exhibited a significant decrease. However, in the mid and late stages, great differences were shown in phytohormone content. During the grafting healing process, the changes in three enzyme activities in healing sites of the three grafting combinations showed some regularities. In conclusion, M. hupehensis and M. robusta have strong grafting compatibility with M. ‘Huabiao’; thus, they are suitable to be selected as rootstocks for grafting and propagation of M. ‘Huabiao’. In addition, the changes in phytohormone content and enzyme activity in different grafting combinations reflect the strong affinity relationship between rootstock and scion grafting.

The narrow crown, a distinct form of canopy architecture and a specialized phenotypic expression of branch angle traits in woody plants, significantly influences key physiological processes such as photosynthesis, transpiration, and biomass accumulation. This phenotype holds substantial application value in forestry production. This article aims to elucidate the biological traits and formation mechanisms of the narrow-crown phenotype in woody plants by analyzing the contributions of plant hormones, their related genes, and specific key genes to the formation and determination of branch angles. Furthermore, the review explores applications of the narrow-crown phenotype in timber forestry, orchard production, breeding of new ornamental woody varieties, and forest tree genetic improvement. Finally, we summarize recent advances in research on narrow-crowned woody plants. On this basis, the future research directions of woody plants with narrow-crown phenotype were prospected, in order to provide reference for promoting the study of branching angle formation of woody plants and the production and application of woody plants with narrow-crown phenotype.
