Environmental enrichment can improve animal welfare. As a method of environmental enrichment, the effect of different auditory stimulations on the behavior response and welfare of laying hen chicks has yet to be investigated. Therefore, this study was aimed at exploring the impact of various auditory exposures on the behavior, learning ability, and fear response of 4-week-old laying hen chicks. A total of 600 1-day-old chicks were randomly assigned to five different groups: C (control group), LM (Mozart’s String Quartets, 65 to 75 dB), LN (recorded ventilation fans and machinery, 65 to 75 dB), HN (recorded ventilation fans and machinery, 85 to 95 dB), and HM (Mozart’s String Quartets, 85 to 95 dB). The experiment was conducted from day 1 until the end of the experiment on day 28. Groups LM and LN were exposed to music and noise stimulation ranging from 65 to 75 dB. Groups HN and HM, meanwhile, received noise and music stimulation ranging from 85 to 95 dB. The control group (C) did not receive any additional auditory stimuli. During the experimental period, continuous behavioral recordings were made of each group of chicks from day 22 to day 28. On day 21, the PAL (one-trial passive avoidance learning) task was conducted. On days 23 and 24, OF (open field) and TI (tonic immobility) tests were performed, and the levels of serum CORT (corticosterone) and DA (dopamine) were measured. The results indicated that exposure to music and noise at intensities ranging from 85 to 95 dB could reduce comforting, preening, PAL avoidance rate, the total number of steps and grid crossings of OF, and the concentration of DA in 4 WOA chicks (p < 0.05), increase the freezing times of OF (p < 0.05); 65 to 75 dB of noise stimulation could reduce preening and total number steps of OF in 4 WOA chicks (p < 0.05), increase the freezing times of OF (p < 0.05); and 65 to 75 dB of music exposure could reduce the concentration of CORT in 4 WOA chicks (p < 0.05). Therefore, 65 to 75 dB of music exposure could produce positive effects on chicks and showed relatively low CORT level, whereas 85 to 95 dB of music and noise exposure could reduce comforting and preening behavior, impair learning ability, and increase the fear responses of chicks.

A 28-d experiment was conducted to investigate the effects of feed-conditioning temperature on the pellet quality, growth performance, intestinal development, and blood parameters of geese. A total of 180 one-day-old White Yuzhou goslings were randomly allotted to 5 treatment groups, with 6 replicates containing 6 birds each. Five diets were conditioned at 65, 70, 75, 80, and 85°C. Body weight and feed intake per pen basis were recorded from the arrival to the end of the trial. Blood and small intestine samples were collected on d 28 for analysis. The results showed that the pellet durability index (PDI), pellet hardness, and gelatinisation degree of starch (GDS) increased with increasing conditioning temperature (P < 0.05). The final body weight (FBW), average daily gain (ADG) and average daily feed intake (ADFI) of goslings significantly increased when conditioning temperature increased from 65 or 70°C to 80 or 85°C (P < 0.05), accompanied by unaffected feed conversion ratio (FCR) (P > 0.05). The villus height to crypt depth ratio (VH/CD) in the duodenum and ileum improved with increasing conditioning temperature (P < 0.05). Additionally, trypsin and amylase activity were enhanced when the conditioning temperature increased from 65 to 85°C (P < 0.05). No significant differences in the carcass traits and blood parameters of goslings were observed among the groups (P > 0.05). Overall, under the present experimental conditions, increasing the steam-conditioning temperature of pelleted feed improved pellet quality, growth performance, intestinal morphology, and digestive enzyme activity in goslings. Based on broken-line regression analysis, the lower critical conditioning temperature for ADG in geese from 1 to 28 d of age was 80.95°C.

Transcription factor Homeobox C8 (HOXC8) is identified as a white adipose gene as revealed by expression profile analysis in fat tissues. However, the specific role of HOXC8 in fat accumulation remains to be identified. This study was designed to reveal the effects of HOXC8 on preadipocyte proliferation and differentiation. We first make clear that the expression of HOXC8 is associated with fat contents in muscles, highlighting a role of HOXC8 in fat accumulation. Next, it is demonstrated that HOXC8 promotes the proliferation and differentiation of preadipocytes through gain- and loss-of-function assays in primary cultured porcine preadipocytes. Then, mechanisms underlying the regulation of HOXC8 on preadipocyte proliferation and differentiation are identified with RNA sequencing, and a number of differentially expressed genes (DEGs) in response to HOXC8 knockdown are identified. The top GO (Gene Ontology) terms enriched by DEGs involved in proliferation and differentiation, respectively, are identical. IL-17 signaling pathway is the common one significantly enriched by DEGs involved in preadipocyte proliferation and differentiation, respectively, indicating its importance in mediating fat accumulation regulated by HOXC8. Additionally, we find that the inhibition of proliferation is one of the main processes during preadipocyte differentiation. The results will contribue to further revealing the mechanisms underlying fat accumulation regulated by HOXC8.

This study aimed to evaluate the potential of fermented sweet potato residue (FSPR) as a partial substitute for corn in goose diets, determine its optimal inclusion level, and assess its effects on intestinal morphology, immune status, and cecal microbiota. A 42-day feeding trial was conducted with 144 twenty-eight-day-old male Sichuan white geese randomly assigned to one of four dietary treatments: a corn-soybean meal basal diet (CON) and three test diets replacing corn with 5%, 8%, or 12% of the corn with FSPR on a dry matter basis. Growth performance, carcass traits, meat quality, serum biochemistry, intestinal morphology, and cecal microbiota were comprehensively assessed. Dietary FSPR inclusion induced a significant, dose-dependent reduction in average daily feed intake (P < 0.001). However, feed conversion ratio was linearly improved (P < 0.001), which compensated for the reduced intake and sustained growth performance. Despite reduced intake, the 8% FSPR group achieved the highest final body weight, and the feed conversion ratio was significantly improved in the 8% and 12% FSPR groups (P < 0.001). Additionally, FSPR improved meat quality by maintaining higher postmortem thigh muscle pH. Systemically, supplementation enhanced immunoglobulins (IgM, IgG) and anti-inflammatory IL-10 without inducing pro-inflammatory markers. Ileal morphology was optimized, evidenced by a significantly increased villus height-to-crypt depth ratio (P < 0.001). Cecal microbiota analysis revealed that FSPR enriched beneficial taxa (e.g., Phocaeicola, Lachnospiraceae), enhanced microbial diversity, and upregulated metabolic pathways for carbohydrate and amino acid biosynthesis. In conclusion, FSPR effectively replaces corn in goose diets, enhancing feed efficiency, meat quality, and immunity, likely associated with through gut microbiota modulation, with 8% identified as the optimal inclusion level.

In applying a hierarchical mixed model to genome-wide association analysis (GWAS) of longitudinal data, dimensionality reduction through modeling repeated measurements improves both computational efficiency and statistical power. Legendre polynomials can flexibly fit population growth trajectories, but higher orders substantially increase computational complexity. Instead of using Legendre polynomials, we first estimated fewer individual-specific parameters using biologically meaningful non-linear models and then associated these phenotypic regressions with genetic markers using a multivariate linear mixed model (mvLMM). After performing a canonical transformation of the regressions based on the pre-estimated covariance matrices under the null genomic mvLMM, we decomposed the mvLMM into mutually independent univariate models and incorporated EMMAX to enable rapid genome-wide mixed-model associations for each transformed phenotype. Simulations for longitudinal association analysis in maize and GWAS for the growth trajectories of body weights in mice demonstrated the advantages of hierarchical non-linear mixed models in computing efficiency and statistical power for detecting quantitative trait loci (QTL), compared with mvLMM for multiple growth points and the hierarchical random regression model using Legendre polynomials as sub-models.
