Patrick Dansette is Director of Research Emeritus at CNRS UMR 8601, University of Paris. He is 76, a chemical ingeneer and PhD in biochemistry 1972). Post Doc at NIH in the group of JW Daly and Don Jerina, involved in the study of xenobiotic metabolizing enzymes and of reactive metabolites (Arene oxides, Thiophene sulfoxides and other electrophiles). He has published about 160 papers. He has a strong background in studying irreversible binding to cytochrome P450s and their mechanism based inactivation. In the past he has synthetised or identified by nucleophile trapping a number of reactive intermediates. He studied the active site topology of several CYP 2Cs, using family of substrates and inhibitors, NMR relaxation methods, and computermodelling and cristallisation. He demonstrated the pharmacological activation pathway of antithrombotic agents Clopidogrel and Prasugrel via a sulfenic acid reactive intermediate. He also worked on the metabolism of ferrocens, ferrocenyl analogs of tamoxifen, as anticancer compounds. He also studied the metabolism of anisyl-dithiole thione, and on the mechanism of production of H2S from dithiole thiones.
