Lead (Pb) is a heavy metal pollutant and negatively affects agriculture and ecosystems. Pb can cause oxidative stress and abnormal plant growth. The ascorbic acid-glutathione (AsA-GSH) cycle mainly exists in chloroplasts and resists oxidative stress, scavenges reactive oxygen radicals, and maintains normal photosynthesis. However, the dosage related effects of Pb on pakchoi photosynthesis, via oxidative stress and the AsA-GSH system, remains unclear. In this study, various Pb dosage stress models were tested (low: 300 mg/kg; medium: 600 mg/kg; high: 900 mg/kg). Pb stress induced a dose-dependent increase in Pb content in pakchoi leaves (P < 0.05). Principal component analysis showed that Se, B, and Pb were significantly and negatively correlated. Pb stress also increased MDA content and decreased antioxidant enzymes SOD, GSH-Px, and T-AOC activities (P < 0.05). We also found that Vc content, as well as the GSH/GSSG ratio, decreased. Additionally, Pb stress destroyed chloroplast structure, decreased photosynthesis indicators Pn, Tr, Gs, Ci and VPD, and attenuated Fv/Fm and Fv/Fo (P < 0.05). In the high-dose group, the contents of chlorophyll a, chlorophyll b, and carotenoids decreased significantly, while the expression of chloroplast development genes (GLK, GLN2) decreased (P < 0.05). Our data suggest that Pb stress leads to dosage-dependent, aberrant photosynthesis by inhibiting the AsA-GSH system in pakchoi. This study expands the Pb toxicology research field and provides indications for screening antagonists.

Background: Integrated water and fertilizer management is important for promoting the sustainable development of agriculture. Climate-smart drip irrigation with fertilizer coupling strategies plays an important role to mitigate greenhouse gas emissions, ensuring food production, and alleviating water scarcity and excessive use of fertilizers. Methods: The greenhouse experiment consists of three drip irrigation treatments which include D1: drip irrigation (100 mm); D2: drip irrigation (200 mm); D3: drip irrigation (300 mm) under three different fertilizer management practices N1: nitrogen level (150 kg N ha−1); N2: nitrogen level (300 kg N ha−1); N3: nitrogen level (450 kg N ha−1). Results: The results showed that significantly improved soil moisture contents, quality and tomato yield, while reduced (38.6%) greenhouse gas intensity (GHGI) under the D3N3 treatment. The D2 and D3 drip irrigation treatments with 450 kg nitrogen ha−1 considerably improved NH4+-N contents, and NO3−-N contents at the fruit formation stage. The improve in net primary productivity (NPP), net ecosystem productivity (NEP), evapotranspiration (ET), and ecosystem crop water productivity (CWPeco) through D3N3 treatment is higher. The D3N3 treatment improved (28.2%) the net global warming potential (GWP), but reduced GHGI, due to improved (18.4%) tomato yield. The D3N3 treatment had significantly greater irrigation water productivity (IWP) (42.8%), total soluble sugar (TSS) (32.9%), vitamin C content (VC) (39.2%), soluble sugar content (SSC) (44.2%), lycopene content (41.3%) and nitrogen use efficiency (NUE) (52.4%), as compared to D1N1 treatment. Conclusions: Therefore, in greenhouse experiments, the D3N3 may be an effective water-saving and fertilizer management approach, which can improve WUE, tomato yield, and quality while reducing the effect of global warming.

The tomato (Solanum lycopersicum L.) is widely cultivated in yellow sand substrate-based systems in Northwest China, contributing significantly to regional agriculture. However, suboptimal water and fertilizer management hinders the balanced optimization of yield, fruit quality, and resource efficiency. In this two-year solar greenhouse experiment (2023–2024), we employed a four-factor, three-level orthogonal design [L9(34)] to examine three irrigation regimes—full irrigation (FI 100% of crop evapotranspiration, [ETc]), mild deficit irrigation (DIM 75% ETc), and severe deficit irrigation (DIS 50% ETc)—in combination with staged fertilizer applications at the seedling, flowering/fruit-set, and peak-fruit stages. A multi-objective decision-making framework, integrating an improved entropy weight method with a virtual-ideal-solution-based TOPSIS model, was established to address the complexity of water–fertilizer interactions. The results indicated that irrigation amount (IA) was the primary determinant of yield and water use efficiency (WUE), followed by fertilizer application in the second stage (FII). For fruit quality indices (moisture content, vitamin C (VC), lycopene (LC), soluble sugars (SSs), and soluble solids content (SSC)), IA remained the most influential factor, followed by FIII, FII, and FI; IA also had the largest impact on fruit hardness (Hd), soluble protein (SP), and titratable acidity (TA). An integrated scoring analysis revealed that treatment T5 achieved the highest yield, WUE, and partial factor productivity (PFP), whereas T7 excelled in fruit quality (VC, LC, SSC, and SP). Moreover, TOPSIS confirmed T5 as the optimal water–fertilizer strategy to achieve high yield, improved quality, and efficient resource utilization. Overall, these findings underscore a robust approach for optimizing water–fertilizer coupling in tomato cultivation under yellow sand substrate conditions, thereby enhancing resource use efficiency, promoting sustainable greenhouse agriculture in arid regions, and contributing to national water-saving and yield-increasing priorities.

Low-temperature stress is a major factor that limits the productivity and geographical distribution of melon (Cucumis melo L.). This study elucidates that CmDUF239-1 is a positive regulator of cold stress, and its underlying mechanisms are investigated using root-specific overexpression lines. Seedlings overexpressing CmDUF239-1 exhibited increased biomass and reduced relative electrical conductivity under cold stress. CmDUF239-1 overexpression promoted the accumulation of soluble sugar and proline, which was accompanied by enhanced activity of the proline biosynthetic enzyme Δ1-pyrroline-5-carboxylate synthase (P5CS) and suppressed activity of the proline-degrading enzyme proline dehydrogenase (PDH). Molecular analysis revealed that CmDUF239-1 overexpression upregulated antioxidant enzyme-related genes, sugar metabolism related genes, and proline-related genes. Furthermore, it activated key genes in the ICE-CBF-COR signaling pathway, including CmCBF1, CmCBF2, and the downstream effector gene CmCOR413-2. In conclusion, the CmDUF239-1 gene enhances melon cold tolerance by modulating antioxidant defense, enhancing osmolyte (sugar and proline) metabolism and activating a core signaling pathway. This study not only characterizes a novel function for a DUF family gene but also provides a promising candidate gene for the genetic improvement of cold resilience in melon and other related crops.
