Bromodomain (BRD)-containing proteins are a class of epigenetic readers with unique recognition for N-acetyl-lysine in histones and functions of gene transcription and chromatin modification, known to be critical in various cancers. However, little is known about the roles of distinct BRD-containing protein genes in hepatocellular carcinoma (HCC). Most recently, we investigated the transcriptional and survival data of BRD1, BRD2, BRD3, BRD4, BRD7, BRD8, BRD9 in HCC patients through ONCOMINE, UALCAN, Human Protein Atlas, GEPIA, cBioPortal, STRING, TIMER databases. BRD1/2/3/4/7/8/9 were over-expressed in HCC and were significantly associated with clinical cancer stages and pathological tumor grades. High mRNA expressions of BRD4/8/9 were promising candidate biomarkers in HCC patients. The rate of sequence alternations in BRD1/2/3/4/7/8/9 was relatively high (52%) in HCC patients, and the genetic alternations were correlated with shorter overall survival and disease-free survival in HCC patients. Additionally, the mRNA expression levels of individual BRD genes were significantly positively associated with the immune infiltrating levels of B cells, CD8+ T cells, CD4+ T cells, macrophages, neutrophils, and dendritic cells. And the associations between BRD1/2/3/4/7/8/9 and diverse immune marker sets showed a significance. Overall, these results indicated that BRD4/8/9 could be potential prognostic markers and druggable epigenetic targets in HCC patients.

Purpose: To investigate the role of lymph node ratio (LNR) in young patients with gastric cancer (GC) and develop nomograms to predict the survival of young GC patients. Methods: This retrospective study enrolled stage I-III GC patients before the age of 40 between 2010 and 2016 from the Surveillance, Epidemiology, and End Results (SEER) database. Cox proportional hazards models were used to determine the prognosticators and create the nomograms incorporating LNR to predict overall survival (OS) and cancer-specific survival (CSS). The discriminating superiority of the nomograms was examined using calibration curves, C-index, receiver operating characteristic (ROC) curves, decision curve analysis (DCA), and integrated discrimination improvement (IDI) by comparing with the TNM staging. The performance of the nomograms for risk stratification was analyzed by the Kaplan–Meier method. Results: Based on the significant prognosticators identified in multivariate survival analysis, the nomograms were established and showed LNR as the third strongest predictor. The C-index of the nomograms for OS and CSS were higher than those of the TNM staging (OS: 0.773 vs 0.665; CSS: 0.769 vs 0.666). The ROC curves for the nomograms to predict survival exhibited superior sensitivity and specificity when compared with the TNM staging. The calibration plots, DCA curves, and IDI values of the nomograms also demonstrated adequate fit and ideal net benefit in prediction and clinical utility. The Kaplan–Meier analysis observed remarkable differences in patients divided into different risk subgroups ( P < .001). Conclusions: These results found the clinical outperformance of the LNR-based nomograms for predicting survival in young stage I-III GC patients. Our nomograms may improve accuracy of survival risk prediction and facilitate individualized care of young stage I-III GC patients.

Deep venous thrombosis (DVT) is a prevalent, life-threatening condition with inadequate public awareness. Social media platforms are significant sources of health information. However, they often suffer from variable content quality. This study systematically evaluated DVT-related videos on TikTok and Bilibili, which represent the dominant short-video and medium-to-long video platforms in China. The top 150 DVT-related videos from each platform, TikTok and Bilibili, sorted by default, were retrieved and screened. The basic characteristics of the included videos, as well as the user engagement metrics, were recorded. The quality of the included videos was assessed using the Global Quality Scale (GQS), the modified DISCERN (mDISCERN) score, and the Journal of the American Medical Association (JAMA) benchmark criteria. TikTok videos had higher engagement (all P < 0.001) and shorter duration (median 88s vs 233.5s, P < 0.001). Quality-wise, except for a good GQS rating for Bilibili, quality scores across both platforms were generally moderate or poor. Specifically, Bilibili scored higher on GQS (median 4 vs 3, P < 0.001), while TikTok performed better on mDISCERN and JAMA (both P < 0.001). Furthermore, user engagement was negatively correlated with GQS scores but positively correlated with both mDISCERN and JAMA scores, revealing a potential mismatch between popularity and professional quality. Despite high user engagement, DVT videos on both platforms demonstrate deficient informational quality and reliability, underscoring a significant gap in accessible public health education. Therefore, multi-stakeholder collaboration is imperative to enhance content standards and facilitate effective health dissemination.
