High temperatures are considered one of the most significant limitations to subtropical fishery production. Largemouth bass (Micropterus salmoides) is an economically important freshwater species grown in subtropical areas, which are extremely sensitive to heat stress (HS). However, comprehensive transcriptomic data for the livers of largemouth bass in response to HS are still lacking. In this study, a comparative transcriptomic analysis was performed to investigate the gene expression profiles of the livers of largemouth bass under HS treatment. As a result, 6114 significantly differentially expressed genes (DEGs), which included 2645 up-regulated and 3469 down-regulated genes, were identified in response to HS. Bioinformatics analyses demonstrated that the ‘ECM-receptor interaction’ pathway was one of the most dramatically changed pathways in response to HS, and eight DEGs assigned to this pathway were taken as hub genes. Furthermore, the expression of these eight hub genes was determined by quantitative reverse transcription PCR, and all of them showed a significant change at the transcriptional level, suggesting a crucial role of the ‘ECM-receptor interaction’ pathway in the response of largemouth bass to HS. These findings may improve our understanding of the molecular mechanisms underlying the response of largemouth bass to HS.

Chito-oligosaccharides (COS) and β-glucan are gradually being applied in aquaculture as antioxidants and immunomodulators. However, this study examined the effects of dietary supplementation of COS and β-glucan on the water quality, gut microbiota, intestinal morphology, non-specific immunity, and meat quality of Chinese soft-shell turtle. To investigate the possible mechanisms, 3-year-old turtles were fed basal diet (CK group) and 0.1%, 0.5%, and 1% COS or β-glucan supplemented diet for 4 weeks. Colon, liver, blood and muscle tissues, colon contents, water and sediment of paddy field samples were collected and analyzed after feeding 2 and 4 weeks. The results indicated that COS and β-glucan altered microbial community composition and diversity in Chinese soft-shell turtles. The relative abundance of Cellulosilyticum, Helicobacter and Solibacillus were increased after feeding COS, while Romboutsia, Akkermansia and Paraclostridium were increased after feeding β-glucan, whereas Cetobacterium, Vibrio and Edwardsiella were enriched in the control group. Furthermore, colon morphology analysis revealed that COS and β-glucan improved the length and number of intestinal villi, and the effect of 0.5% β-glucan was more obvious. Both β-glucan and COS significantly improved liver and serum lysozyme activity and antibacterial capacity. COS significantly increased the total antioxidant capacity in the liver. Further, 0.1% β-glucan significantly increased the activity of hepatic alkaline phosphatase, which closely related to the bacteria involved in lipid metabolism. Moreover, dietary supplementation with 1% COS and 1% β-glucan significantly enhanced the content of total amino acids, especially umami amino acids, in muscle tissue, with β-glucan exerting a stronger effect than COS. Additionally, these two prebiotics promoted the quality of culture water in paddy fields and reshaped the bacterial community composition of aquaculture environment. All these phenotypic changes were closely associated with the gut microbes regulated by these two prebiotics. In summary, the findings suggest that dietary supplementation with COS and β-glucan in Pelodiscus sinensis could modulate the gut microbiota, improve intestinal morphology, enhance non-specific immunity and antioxidant capacity of liver and serum, increase meat quality, and improve the culture water environment. This study provides new insights and a comprehensive understanding of the positive effects of COS and β-glucan on Pelodiscus sinensis.

The Chinese soft-shelled turtle (Pelodiscus sinensis) is an important aquatic species in southern China that is threatened by many serious diseases. Edwardsiella tarda is one of the highly pathogenic bacteria that cause the white abdominal shell disease. Yet, little is known about the immune and metabolic responses of the Chinese soft-shelled turtle against E. tarda infection. In the paper, gene expression profiles in the turtle liver were obtained to study the immune responses and metabolic regulations induced by E. tarda infection using RNA sequencing. A total of 3908 differentially expressed unigenes between the experimental group and the control group were obtained by transcriptome analysis, among them, were the significantly upregulated unigenes and downregulated unigenes 2065 and 1922, respectively. Further annotation and analysis revealed that the DEGs were mainly enriched in complement and coagulation cascades, phagosome, and steroid hormone biosynthesis pathways, indicating that they were mainly associated with defense mechanisms in the turtle liver against E. tarda four days post infection. For the first time, we reported on the gene profile of anti-E. tarda response in the soft-shelled turtle, and our research might provide valuable data to support further study on anti-E. tarda defense mechanisms in turtles.

The Chinese soft-shelled turtle (Pelodiscus sinensis) is an aquatic reptile prized for its nutritional and health benefits. Given its adaptability to various culturing modes including the greenhouse, pond and rice culturing modes, we conducted a comparative analysis of the morphology, organ trait and nutritional composition of turtles cultured in three culturing modes. This study investigated the plasticity of morphology and physiology, as well as the variations in nutritional composition across varying culturing modes. The results demonstrated that after approximately 120 days of cultivation, significant changes were observed in the morphology, physiology and nutritional composition of turtles from each culturing mode. In terms of morphology, rice turtles exhibited an arched shell shape, broad plastron, elongated limbs, narrow interocular distance and slender head and neck. Pond turtles displayed similar morphological characteristics to rice turtles, with the additional features of a flattened body shape and narrower plastron. Greenhouse turtles presented a flattened shell shape, narrow plastron, shortened limbs, wider interocular distance and stocky head and neck. Regarding the organ characteristics, the specific weights of liver, viscera, internal fat lumps and condition factors were significantly higher in greenhouse turtles compared to rice turtles and pond turtles (p < 0.05). Conversely, the specific weights of the back carapace, calipash and edible part were significantly lower than those in rice turtles and pond turtles (p < 0.05). Nutritional analysis revealed that crude protein, total amino acid, essential amino acid, flavor amino acid, pharmacodynamic amino acid, collagen and EPA+DHA contents were significantly higher in rice turtles and pond turtles than greenhouse turtles (p < 0.05). However, crude fat and unsaturated fatty acid contents were significantly higher in greenhouse turtles than in rice turtles and pond turtles (p < 0.05). In summary, Chinese soft-shelled turtles exhibited significant morphological and organ plasticity in response to different culturing modes. While the rice and pond culturing modes could enhance the nutritional quality of turtles to some extent, the impact of commercial feed on fatty acid profiles must be carefully considered.

The barbel steed ( Hemibarbus labeo ), an economically important freshwater fish in China, belongs to the order Cypriniformes and the family Cyprinidae. At present, research has been conducted on the basic biology, artificial breeding, aquaculture, and nutritional requirements of H. labeo . However, genome assembly and exploration of the unique genomic composition of the barbel steed are still in their infancy. Previously available genome assemblies were fragmented and incomplete, hindering advances in genomic research and breeding applications. In this study, we generated a near complete telomere-to-telomere (T2T) genome assemblies for a male H. labeo using PacBio HiFi, ONT ultra-long, and Hi-C data. The resulting assemblies span 1,040.46 Mb with contig N50 of 41.15 Mb, and 25 chromosomes were assembled. The quality value (QV) scores were 60.02, and Benchmarking Universal Single-Copy Orthologs (BUSCO) completeness scores were 98.3%. The genome contains a total of 37,023 coding genes, of which 35,589 (96.13%) were functionally annotated. The high-quality near-T2T assemblies provide invaluable resources for molecular breeding and advancing biological and evolutionary research in H. labeo .
