To analyze the whole genome of Bacillus aryabhattai strain SK1-7 and explore its potassium solubilization characteristics and mechanism, thus providing a theoretical basis for analyzing the utilization and improvement of insoluble potassium resources in soil. Genome information for Bacillus aryabhattai SK1-7 was obtained by using Illumina NovaSeq second-generation sequencing and GridION Nanopore ONT third-generation sequencing technology. The contents of organic acids and polysaccharides in fermentation broth of Bacillus aryabhattai SK1-7 were determined by high-performance liquid chromatography and the anthrone sulfuric acid method, and the expression levels of the potassium solubilization-related genes ackA, epsB, gltA, mdh and ppc were compared by real-time fluorescence quantitative PCR under different potassium source culture conditions. The whole genome of the strain consisted of a complete chromosome sequence and four plasmid sequences. The sequence sizes of the chromosomes and plasmids P1, P2, P3 and P4 were 5,188,391 bp, 136,204 bp, 124,862 bp, 67,200 bp and 12,374 bp, respectively. The GC contents were 38.2, 34.4, 33.6, 32.8, and 33.7%. Strain SK1-7 mainly secreted malic, formic, acetic and citric acids under culture with an insoluble potassium source. The polysaccharide content produced with an insoluble potassium source was higher than that with a soluble potassium source. The expression levels of five potassium solubilization-related genes with the insoluble potassium source were higher than those with the soluble potassium source.

Burkholderia pyrrocinia JK-SH007 is a plant growth-promoting bacteria (PGPB), that can promote the growth of poplar and other trees, and, production of the plant hormone indole-3-acetic acid (IAA) is one of the reasons for this effect. Therefore, the aims of this study were to evaluate the effect of the external environment on the synthesis of IAA by B. pyrrocinia JK-SH007 and to perform a functional analysis of its IAA synthesis pathway. In this study, IAA and its synthetic intermediates indole-3-acetamide (IAM), indole-3-pyruvic acid (IPyA), tryptamine (TAM), and indole-3-acetonitrile (IAN) were detected in B. pyrrocinia JK-SH007 fermentation broth by high-performance liquid chromatography and tandem mass spectrometry (HPLC-MS/MS), and these indolic compounds were also found in the cell-free extraction of B. pyrrocinia JK-SH007, but the genomic analysis of B. pyrrocinia JK-SH007 indicated that IAA biosynthesis was mainly through the IAM and TAM pathways. The effects of L-tryptophan (L-Trp), temperature and pH on the synthesis of IAA were investigated, and the results showed that L-Trp exerted a significant effect on IAA synthesis and that 37°C and pH 7 were the optimal conditions IAA production by B. pyrrocinia JK-SH007. In addition, the protein expression of tryptophan 2-monooxygenase and indoleacetamide hydrolase, which are the key enzymes of the indole acetamide-mediated IAA synthesis pathway, was analyzed, and their activity was verified by substrate feeding experiments. The results revealed the existence of an IAA synthesis pathway mediated by IAM and indicated that this pathway plays a role in B. pyrrocinia JK-SH007. This study lays the foundation for further exploration of the specific pathway and mechanism of IAA synthesis in B. pyrrocinia JK-SH007.

Anthracnose is a foliar disease of the Euonymus shrub caused by Colletotrichum gloeosporioides. In this study, the bacterium HYEB5-6 was isolated from inside one-year-old branches of healthy Euonymus japonicus and showed significant antifungal activities against various phytopathogenic fungi, including C. gloeosporioides s.s. HYCG2-3, in dual culture experiments. The HYEB5-6 isolate significantly decreased lesion diameter and disease index caused by C. gloeosporioides inoculation on detached leaves of E. japonicus. The effects of HYEB5-6 metabolites on the invading structure of the fungus were investigated. Bacterial metabolites inhibited conidial germination, the growth of the germ tube and appressorium formation, possibly through protease and glucanase of HYEB5-6 by managing the mycelial cell wall. The HYEB5-6 isolate also produced a massive biofilm, which might facilitate leaf colonisation. These results indicate that HYEB5-6 has the potential for use as a biological control agent against C. gloeosporioides. The HYEB5-6 isolate was identified as Bacillus velezensis based on its biochemical characteristics and its 16S rRNA gene sequence.

Microbial inoculants, as harmless, efficient, and environmentally friendly plant growth promoters and soil conditioners, are attracting increasing attention. In this study, the effects of Bacillus velezensis YH-18 and B. velezensis YH-20 on Prunus davidiana growth and rhizosphere soil bacterial community in continuously cropped soil were investigated by inoculation tests. The results showed that in a pot seedling experiment, inoculation with YH-18 and YH-20 resulted in a certain degree of increase in diameter growth, plant height, and leaf area at different time periods of 180 days compared with the control. Moreover, after 30 and 90 days of inoculation, the available nutrients in the soil were effectively improved, which protected the continuously cropped soil from acidification. In addition, high-throughput sequencing showed that inoculation with microbial inoculants effectively slowed the decrease in soil microbial richness and diversity over a one-month period. At the phylum level, Proteobacteria and Bacteroidetes were significantly enriched on the 30th day. At the genus level, Sphingomonas and Pseudomonas were significantly enriched at 15 and 30 days, respectively. These bacterial phyla and genera can effectively improve the soil nutrient utilization rate, antagonize plant pathogenic bacteria, and benefit the growth of plants. Furthermore, inoculation with YH-18 and inoculation with YH-20 resulted in similar changes in the rhizosphere microbiome. This study provides a basis for the short-term effect of microbial inoculants on the P. davidiana rhizosphere microbiome and has application value for promoting the cultivation and production of high-quality fruit trees.

In the present study, the potassium-solubilizing characteristics of Bacillus aryabhattai SK1-7 and its growth-promoting effect on plants were evaluated to determine the biotechnological potential of this bacterium in alleviating soil potassium deficiency. The potassium-solubilizing activity of SK1-7 was determined by fermentation. Additionally, the fermentation broth was determined by flame spectrophotometry. The aluminum and silicon ion contents in SK1-7 fermentation broth were determined by inductively coupled plasma-atomic emission spectroscopy (ICP-AES) after digestion with nitric acid hydrogen peroxide hydrofluoric acid. Scanning electron microscopy (SEM)-based observations were performed to assess the morphological changes in potassium feldspar surfaces digested by potassium-solubilizing bacteria. In addition, the effects of SK1-7 on plant growth and soil physical and chemical properties were analyzed. After incubation for 7 days in a potassium-solubilizing medium, the concentration of potassium dissolved reached 10.8 μg/mL and the percentage of potassium released was 32.6%. The pH rapidly decreased from 7.2 to 4.321 within the first day and then further decreased to 3.90 after 7 days. After 7 days, the concentrations of aluminum and silicon in the fermentation broth were 1.01 and 24.19 μg/mL, respectively. The growth promotion assay results showed that SK1-7 has good growth-promoting effects on poplar and can effectively improve the available potassium content in poplar rhizosphere soil. The SK1-7 strain can effectively dissolve insoluble potassium to release soluble potassium ions and clearly promotes the growth of poplar after being applied to soil. Thus, the SK1-7 strain is a potassium-solubilizing microorganism with good application prospects.

Chinese fir (Cunninghamia lanceolata) is a significant timber species that has been broadly cultivated in southern China. A shoot blight disease on Chinese fir seedlings was discovered in Fujian, China and a fungus was then consistently associated with the symptoms. This fungus was determined to be causing this disease, among others by fulfilling Koch’s postulates. Based on morphological characteristics and multilocus phylogenetic analyses with the sequences of the internal transcribed spacer, partial glyceraldehyde-3-phosphate dehydrogenase gene, partial translation elongation factor 1-α gene, and partial 28S large subunit ribosomal RNA gene, the fungus was identified as Bipolaris oryzae. These characteristics and phylogenetic analyses clearly support that this pathogen is different from B. sacchari, which was, until now, considered to be the causal agent of a similar blight on Chinese fir in Guangdong, China. The fungus was also shown to be strongly pathogenic to rice, one of the most susceptible hosts to B. oryzae. Crop rotation involving rice is often carried out with Chinese fir in southern China, a practice that most likely increases the risk of shoot blight on C. lanceolata. To our knowledge, shoot blight caused by B. oryzae is reported for the first time in a gymnosperm species.
