As an exogenous carbon input, microplastics (MPs), especially biodegradable MPs, may significantly disrupt soil microbial communities and soil element cycling (CNPS cycling), but few studies have focused on this. Here, we focused on assessing the effects of conventional low-density polyethylene (LDPE), biodegradable polybutylene adipate terephthalate (PBAT), and polylactic acid (PLA) MPs on rhizosphere microbial communities and CNPS cycling in a soil-soybean system. The results showed that PBAT-MPs and PLA-MPs were more detrimental to soybean growth than LDPE-MPs, resulting in a reduction in shoot nitrogen (14.05% and 11.84%) and shoot biomass (33.80% and 28.09%) at the podding stage. In addition, dissolved organic carbon (DOC) increased by 20.91% and 66.59%, while nitrate nitrogen (NO3--N) significantly decreased by 56.91% and 69.65% in soils treated with PBAT-MPs and PLA-MPs, respectively. PBAT-MPs and PLA-MPs mainly enhanced copiotrophic bacteria (Proteobacteria) and suppressed oligotrophic bacteria (Verrucomicrobiota, Gemmatimonadota, etc.), increasing the abundance of CNPS cycling-related functional genes. LDPE-MPs tended to enrich oligotrophic bacteria (Verrucomicrobiota, etc.) and decrease the abundance of CNPS cycling-related functional genes. Correlation analysis revealed that MPs with different degradation properties selectively affected the composition and function of the bacterial community, resulting in changes in the availability of soil nutrients (especially NO3--N). Redundancy analysis further indicated that NO3--N was the primary constraining factor for soybean growth. This study provides a new perspective for revealing the underlying ecological effects of MPs on soil-plant systems.

Soil heavy metal pollution is one of the most serious environmental problems in China, especially cadmium (Cd), which has the most extensive contaminated soil coverage. Therefore, more economical and efficient remediation methods and measures are needed to control soil Cd contamination. In this study, different amendments (biochar (B), organic fertilizer (F), lime (L)) and actinomycetes (A) inoculants were applied to Cd contaminated farmland to explore their effects on wheat growth. Compared with Control, all treatments except A treatment were able to significantly increase the underground parts dry mass of wheat, with the highest increase of 57.19 %. The results showed that the B treatment significantly increased the plant height of wheat by 3.45 %. All treatments increased wheat SOD activity and chlorophyll content and reduced the MDA, which contributes to wheat stress resistance under Cd contamination. F, L and AF treatments can significantly reduce the Cd content in wheat above- and underground parts by up to 56.39 %. Soil amendments can modify the physical and chemical properties of the soil, which in turn affects the absorption of Cd by wheat. Moreover, the addition of soil amendments significantly affects the composition and structure of the rhizospheric soil bacterial community at the wheat jointing stage. The application of organic fertilizer increases the richness and diversity of the bacterial community, while lime makes it significantly decreases it. T-test and microbiome co-occurrence networks show that actinomycetes could not only effectively colonize in local soil, but also effectively enhance the complexity and stability of the rhizosphere microbial community. Considering the practical impact of different treatments on wheat, soil microorganisms, economic benefits and restoration of soil Cd contamination, the application of organic fertilizer and actinomycetes in Cd contaminated soil is a more ideal remediation strategy. This conclusion can be further verified by studying larger repair regions and longer consecutive repair cycles to gain insight into the repair mechanism.

Orobanche cumama wallr. is the sunflower root parasitic weed with special life stage in which seed germination and parasitism take place in the soil. In practice, applying microbial agents and trapping crop rotation are utilized separately, or just one of them is selected to control O. cumana. The development of the sunflower industry is severely constrained on the farmland, where there is high density of O. cumana’s seed banks. In this study, two biological control methods were combined to solve the problem of O. cumana parasitism. The bioassay experiment showed that the high concentration fermentation filtrates of Streptomyces rochei D74 could effectively inhibit the germination and growth of the germ tube of O. cumana seeds. As the concentration was increased to 3.1 mg/mL, O. cumana was almost unable to sprout. A two-year pot experiment revealed that the use of D74 agents and sunflower–maize–sunflower rotation together promoted sunflower growth, as shown by the biomass accumulation, plant height, and denser root systems. The combined method resulted in a significant decrease in the number of O. cumana parasitism, compared to one method alone. Additionally, it affected the bacterial community composition of sunflower rhizosphere, mostly leading to an increase in Streptomyces and Brevibacterium and a decrease in Arthrobacter. This experiment, combined with multiple biological control, means significantly reducing the parasitism of O. cumana, which provides an effective foundation for practical application.

Orobanche cumana Wallr. is a root parasitic plant that causes considerable yield losses of up to 50% in sunflower Helianthus annuus plantations. The holoparasite fulfills its entire demand for water, minerals, and organic nutrients from the host’s vascular system. Agronomic practices alone are not effective in controlling this pest. This study investigated the mechanism of a verified plant growth-promoting strain, Streptomyces rochei D74, on the inhibition of the parasitism of O. cumana in a co-culture experiment. We conducted potted and sterile co-culture experiments using sunflower, O. cumana, and S. rochei D74. Our results suggest that the inoculated bacteria invoked the sunflower systemic resistance (SAR and ISR) by increasing the activity of resistance-related enzymes (SOD, POD, PPO, and PAL), the gene expression of systemic resistance marker genes (PR-1 and NPR1), ethylene synthesis genes (HACS. 1 and ACCO1), and JA synthesis genes (pin2 and lox). The expression levels of ISR marker genes (lox, HACS. 1, ACCO1, and pin2) increased by 1.66–7.91-fold in the seedling stage. Simultaneously, S. rochei D74 formed a protective layer on the sunflower root surface, preventing O. cumana from connecting to the vascular system of the sunflower roots. In addition, S. rochei D74 reduced 5DS synthesis of the strigol precursor substance, resulting in a reduction in O. cumana germination. These results demonstrated that the S. rochei D74 strain improved systemic resistance and decreased seed germination to prevent O. cumana parasitism.

Sunflower broomrape (Orobanche cumana Wallr.) has severely restricted the development of the sunflower industry in China, and more efficient and convenient control methods are urgently needed. In this experiment, we investigated the effects of N, P and silica fertilizers on the parasitism rate of O. cumana, as well as on the yield of sunflower and native microbial communities in the field. Firstly, pot experiments were conducted to select the most effective fertilization method and to determine the physiological and biochemical indexes of sunflowers. Subsequently, field application studies were carried out to determine the physiological indexes, yield, O. cumana parasitism on sunflower, and the effect on the indigenous microbial community. The results demonstrate that compared with the CNP treatment (Control), the number of parasites under the N1P5 treatment significantly decreased by 66.15%. The exogenous application of silica can significantly reduce the number of O. cumana parasites. The treatments with N1P5 (N/P = 1:5) and available SiO2 content higher than 40 mg/kg (NS2, NS3, SF2 and SF3) significantly increased superoxide dismutase (SOD) activity and decreased malondialdehyde (MDA) content in sunflower leaves. In the field, fertilization significantly decreased the number of O. cumana parasites. The S treatment improved the native microbial community structure and enriched beneficial microorganisms, including Vicinamibacteria and Pyrinomonadaceae. Additionally, applying the S treatment significantly increased sunflower yield by 23.82% and crude protein content by 20.20%. In summary, the application of silicon fertilizer can effectively improve the host microbial community, reduce O. cumana parasitism and improve the yield and quality of sunflower.

Both organic matter and iron oxide (FeO) dynamics pose key roles in soil cadmium (Cd) bioavailability. However, the microbially driven transformation of soil organic matter and FeO and their linkages to Cd fractions remain unclear under reductive soil disinfestation (RSD) with different organic sources, which limits our mechanistic understanding of Cd immobilization by RSD. To address this gap, we conducted a 45 day microcosm experiment using a paddy soil contaminated with 22.8 mg/kg Cd. Six treatments were established: untreated control (CK), waterlogged (WF), and RSD-amended soils with 0.7% or 2.1% wheat straw (LWD, HWD) or soybean meal (LSD, HSD). We systematically assessed soil Cd fractionation, organic carbon and FeO concentrations, and bacterial community structure, aiming to clarify differences in Cd immobilization efficiency and the underlying mechanisms between wheat straw and soybean meal. For strongly extractable Cd, wheat straw RSD reduced the soil Cd concentrations from 6.02 mg/kg to 4.32 mg/kg (28.2%), whereas soybean meal RSD achieved a maximum reduction to 2.26 mg/kg (62.5%). Additionally, the soil mobility factor of Cd decreased from 44.6% (CK) to 39.2% (HWD) and 32.5% (HSD), while the distribution index increased from 58.5% (CK) to 62.2% (HWD) and 66.8% (HSD). Notably, the HWD treatment increased soil total organic carbon, humus, and humic acid concentrations by 34.8%, 24.6%, and 28.3%, respectively. Regarding amorphous FeO, their concentrations increased by 19.1% and 33.3% relative to CK. RSD treatments significantly altered soil C/N ratios (5.91–12.5). The higher C/N ratios associated with wheat straw stimulated r-strategist bacteria (e.g., Firmicutes, Bacteroidetes), which promoted carbohydrate degradation and fermentation, thereby enhancing the accumulation of humic substances. In contrast, the lower C/N ratios of soybean meal increased dissolved organic carbon and activated iron-reducing bacteria (FeRB; e.g., Anaeromyxobacter, Clostridium), driving iron reduction and amorphous iron oxide formation. PLS-PM analysis confirmed that wheat straw RSD immobilized Cd primarily through humification, whereas soybean meal RSD relied on FeRB-mediated FeO amorphization. These findings suggest that Cd immobilization in soil under RSD may be regulated by microbially mediated organic matter transformation and iron oxide dynamics, which was affected by organic materials of different C/N ratios.
