Anisakidosis is a foodborne zoonotic infection induced by members of the family Anisakidae via the consumption of raw or undercooked fish such as sushi and sashimi. Identifying anisakid larval species is critical for the epidemiology and diagnosis of diseases caused by them. This study aimed at identifying Anisakis larvae collected from marine fish in Egyptian waters based on morphological characteristics and molecular analysis. Thirty marine fish coral trout, Plectropomus areolatus, were collected from Hurghada, Red Sea, Egypt, to investigate larval nematodes of the genus Anisakis. The larvae were detected encapsulated in the peritoneal cavity and muscle of the fish host. This examination revealed that anisakid larvae naturally infected 19 fish specimens with a prevalence of 63.33% and a mean intensity of 4.1 ± 0.40. Most of them (68 larvae: 71.57%) were found in the musculature. Morphological and morphometric analyses using light and scanning electron microscopy revealed a head region with a prominent boring tooth, inconspicuous lips, and a characteristic protruded cylindrical mucron. All larvae in this study possessed the same morphology as Anisakis Larval type I. Molecular analysis based on ITS region using maximum likelihood and Bayesian phylogenetic methods confirmed them as Anisakis typica. This is the first study to identify A. typica larvae from the commercial fish coral trout P. areolatus in Egyptian waters using morphological and molecular methods.

Natural products extracted from animal sources have many biological activities, such as chitosan, which is being researched for its medicinal or therapeutic potential. Syphacia muris is the most well-known intestinal nematode, infecting laboratory rats and influencing their immune systems. In this study, we looked at the anthelminthic activity of chitosan particles against S. muris infection using biochemical, immunological, and histopathological methods. Chitosan particles were characterized using Fourier-transform infrared spectroscopy (FTIR). Rats were separated into four groups, each consisting of seven individuals (n = 7). The first group was the control (non-infected), the second group was infected, and both groups received 0.5 ml of 1% glacial acetic acid orally. The third group was the infected group (treated), and the fourth group (normal) received 0.5 ml of 30 mg/kg/day chitosan dissolved in 1% glacial acetic acid for 14 days using gavage. Liver and kidney parameters, oxidative stress markers, serum levels of cytokines (IFN-γ, IL-5, IL-13, IL-33, and IL-10), as well as immunoglobulins (total IgE and IgG), were assessed. Histological examinations of host tissues (intestine, liver, kidney, and spleen) were also performed. Following chitosan treatment, a significant decrease in worm count (P < 0.05) was indicated; this was associated with an enhancement of biochemical and oxidative stress biomarkers, which were altered due to infection. Moreover, immunological analysis revealed a significant drop in INF-γ, IL-5, IL-13, and IL-33 levels and total immunoglobulins (IgE and IgG) as well as an improvement in rat tissues. Conclusively, this study showed the anthelminthic effect of chitosan against S. muris infection.

Worldwide, particularly in developing nations, helminth infections are the leading causes of livestock illness and mortality. Parascaris (P.) equorum, a parasitic worm from the Ascarididae family, significantly impacts the production, health, and working performance of equines. This study aimed to investigate the impact of intraperitoneal sensitization of P. equorum on the immune system, oxidative stress, and histology in Wistar rats. After acclimatization for 7 days, we divided the rats into five groups, each consisting of six rats. Group I, serving as the control, was administered distilled water, followed by groups II (day 7), III (day 14), IV (day 21), and V (day 33). The rats were euthanized every day mentioned (Days 7–33). On day 0, a dosage of 1ml/100 gm rat (containing 500 μg/ml protein content) emulsified crude antigen extract with an incomplete Freund’s adjuvant (1:1 volume), followed by a second dose of the same antigen concentration on day 7. To assess the allergenicity of this nematode, we measured a whole blood profile, serum levels of IFN-γ, IL-5, IL-10, IL-13, and IL-33, total immunoglobulins IgE and IgG, and oxidative stress markers. Also, we examined histological changes in the liver, kidney, and spleen. The results showed that values of total leukocyte count, granulocytes, monocytes, and lymphocytes were significantly (P < 0.05) increased on day 14 post-infection relative to other days of investigation. It was found that the levels of total immunoglobulins (IgE and IgG) and cytokines (INF-γ, IL-5, IL-13, and IL-33) on days 14 and 21 were significantly higher than in the control group. At all periods of the experiment, the injected group exhibited significantly higher concentrations of MDA and NO compared to the control group (P < 0.05). Conversely, GSH and CAT levels (P < 0.05) dropped significantly on days 7, 14, and 21. Different rat tissues showed alterations. Ultimately, this study described the detrimental effects of P. equorum crude antigen administration on the immune system, oxidative states, and histological changes of Wistar rats at various intervals.

Schistosomiasis is a severe illness carried by snails of the genus Biomphalaria, which have a large distribution in Egypt. Control of the intermediate host snail is critical in limiting schistosomiasis spread. On the topic of snails’ management, bioactive products extracted from aquatic invertebrates have gained more interest. Here, for the first time, the current research intended to identify the bioactive compounds present in hemolymph collected from the freshwater crayfish Procambarus clarkii and to assess its molluscidal activity against Biomphalaria alexandrina snails. Results showed that following exposure to the sub-lethal concentrations (LC10 34.9 and LC25 42.8 mg/l) of hemolymph, a reduction in survival and reproductive rates of B. alexandrina was detected, as well as a substantial increase in transaminases (AST and ALT) activities, while a reduction in total protein and albumin levels. Also, levels of testosterone and 17β-estradiol hormones decreased compared to the control group. Histopathological examination of the digestive gland of treated B. alexandrina displayed a shrinkage of the tubules and an increase in the inter-tubular spaces, and also, degeneration in the oocytes and spermatocytes of the hermaphrodite glands with a loss of connective tissues between the acini was observed. Thus, based on these results, a new strategy should be developed for a better use of these agents in schistosomiasis control.

As one of the neglected tropical diseases, schistosomiasis is responsible for various social and economic issues in numerous developing countries. The intermediate host of Schistosoma mansoni is the Biomphalaria alexandrina snails. A promising approach to mitigate the transmission of this disease is to use medicinal plants loaded with nanomaterials to control these snails. This research aimed to investigate the molluscicidal activity of Orobanche aegyptiaca- chitosan nanocomposite on B. alexandrina snails. The phytochemical screening of the tested plant verified its abundance of saponins, tannins, and flavonoids, which may be responsible for its cytotoxic effects. Chitosan nanoparticles were produced using the ionotropic gelation technique, while the O. aegyptiaca-chitosan nanocomposite was synthesized using the sonochemical approach. The characterization of the nanocomposite was carried out using TEM, XRD, and Zeta potential. The results demonstrated that the survival, fecundity (eggs/snail/week), and reproductive rates of B. alexandrina snails were substantially reduced upon exposure to the sub-lethal concentrations LC10 or LC25 of O. aegyptiaca-chitosan nanocomposite. Biochemically, it adversely affected some parameters, where it considerably (P < 0.05) raised ALT, AST, and ALP levels, while urea, uric acid, and cholesterol were significantly decreased compared to control snails. Furthermore, the antioxidant markers CAT, SOD, and GSH exhibited a substantial (P < 0.05) reduction, while MDA and NO levels increased at both sublethal concentrations. Histopathological examinations of the digestive gland of the treated B. alexandrina revealed a decrease in the size of the tubules, and the digestive and secretory cells degenerated. The hermaphrodite glands exhibited substantial damage in the reproductive tubules, with extensive damage to the gonadal cells, resulting in the loss of ova and sperm shapes. Also, the comet analysis showed a genotoxic effect of the nanocomposite, evidenced by significant changes in crucial comet assay parameters: tail length (TL), tail DNA percentage (TD), tail moment (TM), and olive tail moment (OTM). Conclusively, these findings confirmed the potential activity of O. aegyptiaca- chitosan nanocomposite as a molluscicidal agent against B. alexandrina to decrease schistosomiasis transmission.
