Thyroid cancer (THCA) is one of the commonest malignancies associated with increased recurrence. Therefore, identifying the putative molecular markers and therapeutic targets to improve the treatment of THCA is essential. The present study analyzed the potential role of tripartite motif‑containing 21 (TRIM21), a member of the TRIM family belonging to the subfamily of E3 ubiquitin ligases, in the progression of THCA. Using bioinformatics analysis and immunohistochemistry of THCA tissues, it was observed that TRIM21 is overexpressed in THCA tissues. The present study also found that TRIM21 is associated with lymph node metastasis and high‑risk recurrence of THCA. Furthermore, it identified a promotional role of TRIM21 in THCA cell migration and invasion. In addition, the present study analyzed TRIM21‑enriched pathways and co‑expressed genes in THCA. The present study suggested that TRIM21 may serve as a potential biomarker for THCA prognosis.
This study analyzes the research landscape of nanomaterials in treating hepatocellular carcinoma (HCC) and examines publication trends in this field by conducting a comprehensive bibliometric analysis within the Web of Science Core Collection (WoSCC) database. Articles published from 2000 to September 16, 2024 were retrieved using a structured search formula targeting studies on nanomaterials in HCC, including nanoparticles, nanodots, nanorods, nanosheets, and nanomedicine. Only English full-text articles and reviews relevant to nanomaterial applications in HCC were considered, excluding conference abstracts and non-research items. The analysis encompasses annual publication trends, country-wise publication distribution, prominent institutions, and key journals in the field. Statistical and graphical analyses were performed using GraphPad Prism (v8.0.2) to illustrate publication trends. CiteSpace (6.2.4R) and VOSviewer (1.6.18) software were used to visualize co-citation and keyword networks, highlighting scientific knowledge structures and research hotspots. Notable advancements have emerged as a promising strategy, enabling hepatocyte-specific drug delivery to enhance therapeutic precision and minimize off-target effects. This analysis provides a comprehensive understanding of the evolution of HCC nanomaterials research, key contributing countries, major research institutions, and frequently cited keywords. The findings offer valuable insights into the field's knowledge base, emerging trends, and future directions in HCC treatment with nanomaterials.
Immunotherapy has revolutionized cancer treatment paradigms due to its high specificity, favorable safety profile, and remarkable clinical efficacy. Immune checkpoint blockade (ICB) therapy has been widely adopted in clinical practice for various malignancies. Unfortunately, its limited response rates restrict broader application. Myeloid-derived suppressor cells (MDSCs) serve as key mediators of ICB resistance. In this study, we demonstrated that fucoidan extracted from Saccharina japonica LMWF modulated MDSCs activity to sensitize tumors to ICB therapy. Mechanistically, LMWF activated the TLR7-IRF7/STAT1 signaling axis to drive MDSCs differentiation into M1 macrophages while impairing MDSCs survival. LMWF treatment downregulated immunosuppressive mediators in MDSCs, promoted cancer cell apoptosis, and suppressed tumor cell migration and angiogenesis. In vivo studies revealed that LMWF- modulated MDSCs suppressed tumor progression through dual mechanisms: (i) immunological remodeling of the immunosuppressive tumor microenvironment (TME) and (ii) non-immunological inhibition of angiogenesis, collectively enhancing tumor sensitivity to PD-1 blockade therapy. Collectively, our work unveiled a novel strategy whereby LMWF, as a TLR7 agonist, synergizes with ICB therapy to overcome ICB resistance. These findings provide critical insights for overcoming the bottleneck of ICB refractoriness in cancer immunotherapy.
Parkinson's disease (PD) is a highly prevalent and severe neurodegenerative disease that affects more than 10 million individuals worldwide. Pathogenic mutations in LRP10 have been associated with autosomal dominant PD. Here, we report an induced pluripotent stem cell (iPSC) line generated from a PD patient harboring the LRP10 c.688C > T (p.Arg230Trp) variant. Skin fibroblasts from the PD patient were successfully reprogrammed into iPSCs that expressed pluripotency markers, a normal karyotype, and the capacity to differentiate into the three germ layers in vivo. This iPSC line is a potential resource for studying the pathogenic mechanisms of PD.
We investigated the potential correlation between abnormal blood glucose on admission and clinical outcomes of acute calculus cholecystitis in older patients. We included older patients (≥65 years) diagnosed with acute calculus cholecystitis. The first plasma glucose measurement obtained at admission was used. Overall, 280 older patients were evaluated. The results showed that ABG was significantly associated with more severe systemic inflammatory responses (elevated white blood cell count and neutrophil-lymphocyte ratio), a higher incidence of moderate to severe ACC, and the need for invasive treatment (an increased proportion of percutaneous cholecystostomy and combined surgeries). The prolonged hospital stay, 30-day readmission rate and mortality rate of patients in the ABG group were significantly increased, and the incidence of severe postoperative complications (Clavien-Dindo≥grade III) was higher. Multivariate analysis confirmed that ABG was an independent predictor of the severity of ACC (TG18 classification) and 30-day re-admission rate. In older patients with acute calculus cholecystitis, abnormal blood glucose on admission is associated with worse outcomes, serving as a novel quantifiable risk criterion to guide treatment selection, particularly for high-risk older adults ineligible for early surgery.
BACKGROUND The incidence of breast cancer is high among women, with a significant proportion of cases being familial. However, the driver genes for breast cancer can differ across families. CASE REPORT Our patient was a 37-year-old woman diagnosed with triple-negative breast cancer (TNBC) by pathology, revealing invasive ductal carcinoma of the outer upper quadrant of the breast, WHO grade 3. The maximum diameter of the microscopic invasive cancer was approximately 0.5 cm. No definite vascular tumor thrombus or nerve invasion was observed. Some (30-90%) of the tumor cells disappeared, and the remaining tumor cells showed degeneration, interstitial sclerosis, scattered lymphocyte infiltration, and hemosiderin deposition. No cancer was found in the nipple and base resection margins, or in the other quadrants. The chemotherapy response was classified as grade III according to the MP (Miller and Payen classification) scoring system. Blood samples were collected from affected family members. Whole-exome sequencing (WES) and bioinformatics analyses were used to identify potential driver genes, followed by Sanger sequencing for validation, which ultimately confirmed the pathogenic gene and the underlying mechanism in this family. CONCLUSIONS A series of analyses suggested that the co-occurrence of heterozygous deletions in BRCA1 and OBSCN was the main cause of breast cancer in this family. The simultaneous association of 2 genes with the occurrence of breast cancer was discovered for the first time in this family, which could help guide disease prevention for family.
