Soluble sugars such as sucrose, glucose and fructose in plant host cells not only play the role as donors of carbon skeletons, but they may also induce metabolic signals influencing the expression of defense genes. These metabolites function in a complex network with many bioactive molecules, which independently or in dialogue, induce successive defense mechanisms. The aim of this study was to determine the involvement of sucrose and monosaccharides as signaling molecules in the regulation of the levels of phytohormones and hydrogen peroxide participating in the defense responses of Lupinus luteus L. to a hemibiotrophic fungus Fusarium oxysporum Schlecht f. sp. lupini. A positive correlation between the level of sugars and postinfection accumulation of salicylic acid and its glucoside, as well as abscisic acid, was noted. The stimulatory effect of sugars on the production of ethylene was also reported. The protective role of soluble sugars in embryo axes of yellow lupine was seen in the limited development of infection and fusariosis. These results provide evidence for the enhanced generation of signaling molecules both by sugar alone as well as during the crosstalk between sugars and infection caused by F. oxysporum. However, a considerable postinfection increase in the level of these signaling molecules under the influence of sugars was recorded. The duration of the postinfection generation of these molecules in yellow lupine was also variable.

The aim of the research was to determine selected biochemical and physical characteristics and yielding of fruits in apple varieties (Malus domestica Borkh) such as Gala Schniga, Beni Shogun (Fuji) and Ligol, M.9 rootstock growing in the Wielkopolska region, Poland. high-performance liquid chromatography (HPLC) analysis revealed differences both in the profile and contents of soluble sugars and other metabolites detected in fruits of the tested apple varieties. The highest total saccharide content was found in fruits of cv. Gala Schniga, while leaves and shoots of this variety showed the lowest contents. Electron paramagnetic resonance (EPR) spectroscopy revealed the lowest contents of semiquinone radicals in apple fruits and the highest in leaves of apple trees. All organs of Schniga Gala apple trees were characterized by the highest levels of these radicals. Besides, gas chromatography-mass spectrometry (GC-MS) analysis of abscisic acid (ABA) revealed the highest levels of this molecule in shoots of apple trees, especially the Beni Shogun variety, while in fruits the levels were the lowest. Ligol fruits had the highest content of ABA. The percentage of injury observed after a low-temperature treatment and estimated on the basis of electrolyte leakage, was shown to be the lowest in fruits of the Beni Shogun variety. In turn, the lowest average yield of apple fruits was recorded for the Ligol and Fuji varieties, with the highest in the case of the Gala Schniga variety. At the same time, a significant variability in the average weight of fruits was observed; the highest average mass of fruits was recorded for the Ligol variety, while it was lowest for Gala Schniga. Additionally, firmness evaluation of the fruits revealed that the firmest fruits were harvested from Gala apple trees, before the Beni Shogun and Ligol varieties. A significant variability was recorded in the red blush on fruit surfaces of the tested varieties, with Ligol fruits having the weakest blush. Altogether, these results indicate that fruits of the analyzed apple varieties differed both in terms of their biochemical composition and physical characteristics.

The main aim of this study was to understand the regulation of the biosynthesis of phytohormones as signaling molecules in the defense mechanisms of pea seedlings during the application of abiotic and biotic stress factors. It was important to identify this regulation at the molecular level in Pisum sativum L. seedlings under the influence of various concentrations of lead—i.e., a low concentration increasing plant metabolism, causing a hormetic effect, and a high dose causing a sublethal effect—and during feeding of a phytophagous insect with a piercing-sucking mouthpart—i.e., pea aphid (Acyrthosiphon pisum (Harris)). The aim of the study was to determine the expression level of genes encoding enzymes of the biosynthesis of signaling molecules such as phytohormones—i.e., jasmonates (JA/MeJA), ethylene (ET) and abscisic acid (ABA). Real-time qPCR was applied to analyze the expression of genes encoding enzymes involved in the regulation of the biosynthesis of JA/MeJA (lipoxygenase 1 (LOX1), lipoxygenase 2 (LOX2), 12-oxophytodienoate reductase 1 (OPR1) and jasmonic acid-amido synthetase (JAR1)), ET (1-aminocyclopropane-1-carboxylate synthase 3 (ACS3)) and ABA (9-cis-epoxycarotenoid dioxygenase (NCED) and aldehyde oxidase 1 (AO1)). In response to the abovementioned stress factors—i.e., abiotic and biotic stressors acting independently or simultaneously—the expression of the LOX1, LOX2, OPR1, JAR1, ACS3, NCED and AO1 genes at both sublethal and hormetic doses increased. Particularly high levels of the relative expression of the tested genes in pea seedlings growing at sublethal doses of lead and colonized by A. pisum compared to the control were noticeable. A hormetic dose of lead induced high expression levels of the JAR1, OPR1 and ACS3 genes, especially in leaves. Moreover, an increase in the concentration of phytohormones such as jasmonates (JA and MeJA) and aminococyclopropane-1-carboxylic acid (ACC)-ethylene (ET) precursor was observed. The results of this study indicate that the response of pea seedlings to lead and A. pisum aphid infestation differed greatly at both the gene expression and metabolic levels. The intensity of these defense responses depended on the organ, the metal dose and direct contact of the stress factor with the organ.

The main goal of this study was to investigate the effect of lead (Pb) at various concentrations, as an abiotic factor, and the cross-talk between Pb and pea aphid (Acyrthosiphon pisum (Harris)) (Hemiptera: Aphididae), as a biotic factor, on the defence responses of pea seedlings (Pisum sativum L. cv. Cysterski). The analysis of growth parameters for pea seedlings demonstrated that Pb at a low concentration, i.e., 0.025–0.0625 mM Pb(NO3)2, caused a hormesis effect, i.e., stimulation of seedling growth, whereas Pb at higher concentrations, i.e., 0.01–0.325 mM Pb(NO3)2, inhibited growth, which manifested as the inhibition of length and fresh biomass. The differences in the level of the main defence-related phytohormones, such as abscisic acid (ABA), jasmonic acid (JA) and salicylic acid (SA), and indole-3-acetic acid (IAA)—an auxin stimulating plant cell growth—depended on the dose of Pb, aphid infestation and direct contact of the stress factor with the organ. A high accumulation of soluble sugars in the organs of pea seedlings both at sublethal doses and hormetic doses at early experimental time points was observed. At 0 h and 24 h of the experiment, the hormetic doses of Pb significantly stimulated invertase activities, especially in the roots. Moreover, an increase was observed in the pisatin concentration in pea seedlings growing in the presence of different concentrations of Pb and in the case of cross-talk between Pb and A. pisum in relation to the control. Additionally, a significant induction of the expressions of isoflavone synthase (IFS) and 6α-hydroxymaackiain 3-O-methyltransferase (HMM) genes, which participate in the regulation of the pisatin biosynthesis pathway, in pea seedlings growing under the influence of sublethal 0.5 mM Pb(NO3)2 and hormetic 0.075 mM Pb(NO3)2 doses of Pb was noted. The obtained results showed that the response of P. sativum seedlings depends on the Pb dose applied, direct contact of the stress factor with the organ and the duration of contact.

The main aim of this study was to investigate metabolic responses of fruits, leaves, and shoots of pistachio trees (Pistacia vera L. cv. Kirmizi) during their development. Electron paramagnetic resonance spectroscopy revealed significant increase in generation of semiquinone radicals in fruits and leaves of pistachio, while the flower cluster thinning application was conducted in relation to the control; especially at the second term of the plant material collection. Moreover, flower abscission caused an increase in the level of phytohormones such as indole-3-acetic acid and abscisic acid in fruits at the first term of fruits sampling. In turn, high-performance liquid chromatography analysis revealed differences both in the profile as well as the contents of soluble sugars detected in pistachio organs. The highest total sugar content was found in fruits of pistachio where the flower cluster thinning application was made early. Total sugar levels were higher also in leaves and shoots of the above-mentioned pistachio plants in relation to the control until the third sampling time. The importance of high levels of fructose in the fruits following the flower abscission was observed. Additionally, analyses of mineral elements in organs showed that copper and phosphorus contents in fruits were higher after the flower abscission in relation to the control. To conclude, our findings signal on contribution of semiquinone radicals, paramagnetic manganese ions, phytohormones, nutrients, and sugars in pistachio organs development on the background of the flower cluster thinning which was applied before fructification.
