Despite improvements in therapeutic approaches like immunotherapy and gene therapy, cancer still remains a serious threat to world health due to its high incidence and mortality rates. Limitations of conventional therapy include suboptimal targeting, multidrug resistance, and systemic toxicity. A major challenge in current oncology therapies is the development of new delivery methods for antineoplastic drugs that act directly on target. One approach involves the complexation of antitumor drugs with cyclodextrins (CDs) and chitosan (CS) as an attempt to counteract their primary limitations: low water solubility and bioavailability, diminished in vitro and in vivo stability, and high dose-dependent toxicity. All those drawbacks may potentially exclude some therapeutic candidates from clinical trials, thus their integration into smart delivery systems or drug-targeting technologies must be implemented. We intended to overview new drug delivery systems based on chitosan or cyclodextrins with regard to the current diagnosis and cancer management. This narrative review encompasses full-length articles published in English between 2019 and 2025 (including online ahead of print versions) in PubMed-indexed journals, focusing on recent research on the encapsulation of diverse antitumor drugs within those nanosystems that exhibit responsiveness to various stimuli such as pH, redox potential, and folate receptor levels, thereby enhancing the release of bioactive compounds at tumor sites. The majority of the cited references focus on the most notable research, studies of novel applications, and scientific advancements in the field of nanostructures and functional materials employed in oncological therapies over the last six years. Certainly, there are additional stimuli with research potential that can facilitate the drug’s release upon activation, such as reactive oxygen species (ROS), various enzymes, ATP level, or hypoxia; however, our review exclusively addresses the aforementioned stimuli presented in a comprehensive manner.

All acute and chronic wound management strategies have limitations. Therefore, there is an urgent need to develop new treatment options for wound healing. Hydrogels based on natural polymers offer advantages in wound management because they can reduce patients’ pain, fight infection, and carry targeted drugs to speed up the healing process. In this study, we aimed to develop and investigate an alginate-grafted N-vinylcaprolactam-based matrix for a modified release of dexketoprofen (DEX), which is potentially useful in wound healing. Free radical polymerization and grafted techniques were used to prepare thermo-responsive hydrogels. The obtained hydrogels, unloaded hydrogel (HY) and dexketoprofen-loaded hydrogel (DEXHY), were characterized and analyzed. The concentration of DEX encapsulated in the polymer matrix was 4 mg/mL. The IC50 values found for the samples tested by us were 607.4 µg/mL for HY, 950.4 µg/mL for DEXHY, and 2239 µg/mL for DEX. The average value of cell viability (%) after the exposure of cells to DEXHY hydrogel was 75.4%. DEXHY exhibited a very good in vitro wound closure rate, given its ability to modify DEX release kinetics. The hydrogel developed in this study has shown considerable potential to facilitate and even accelerate wound healing, including surgical wounds, by inhibiting the overexpressed inflammation process.

Hyaluronic acid (HA) hydrogels have attracted increasing interest for biomedical applications due to their tunable properties and biocompatibility. Methods: In this study, hyaluronic acid HA-based hydrogels were developed using two distinct crosslinking strategies: physical crosslinking through poly(vinyl alcohol) (PVA) incorporation and covalent crosslinking via DCC/NHS-mediated reactions. Piroxicam (Px) was included as a model drug to evaluate the drug delivery potential of the resulting systems. The hydrogels were characterized in terms of morphology, swelling behaviour, adhesion, enzymatic degradation, drug release, and in vitro cytocompatibility. Results: The results indicate that formulation parameters significantly influence the overall performance of the systems. PVA-containing hydrogels exhibited higher swelling capacity and improved adhesive properties, while covalently crosslinked networks showed reduced swelling and enhanced structural stability and resistance to enzymatic degradation. Drug release profiles were dependent on network structure, with more compact systems displaying slower release behaviour. In vitro assays suggested that the developed hydrogels are cytocompatible and that drug incorporation influences both release kinetics and cellular response. However, it should be noted that the biological evaluation was performed under simplified in vitro conditions, which primarily reflect specific aspects such as cell viability and migration. Conclusions: This study provides a comparative analysis of physical and covalent crosslinking strategies within a HA platform and highlights how formulation variables influence key physicochemical and biological properties. These findings contribute to the rational design of HA-based hydrogels, although further studies are required to establish their performance in more complex biological environments.

When developing a topical formulation, it is essential to evaluate it from the perspective of its carrier capacity for the controlled release of the encapsulated drug. The drug should be simply quantified through an accurate and reproducible method. This paper presents a rapid, simple, sensitive, and reproducible high-performance liquid chromatography method with UV detection for evaluating the ability of chitosan-graft-β-cyclodextrin/PVA (CS-g-β-CD/PVA) hydrogels as carriers for the controlled release of vemurafenib (VEM). The chromatographic separation was achieved using a Waters CORTECS C18 column (100 × 2.1 mm ID, 2.7 µm). The mobile phase was a mixture of: A (water/formic acid - 99.9/0.1, v/v), B (acetonitrile), and C (methanol) in the ratio of 40:55:5 (v/v/v). The injection sample amount was 10 μL, and the run time was 9 minutes in isocratic mode at a flow rate of 1 mL/min. The analyte was detected using UV absorption at 252 nm. The standard calibration curve was linear over the concentration range of 0.78-100 mg/L. The values for LOD and LOQ were 0.5 mg/L and 0.75 mg/L, respectively. The intra- and inter-day precision of measurements were lower than the accepted criteria (RSD ≤ 2%). The high value of recoveries obtained for VEM indicates that the proposed method was found to be accurate. The stability of VEM solutions was assessed, indicating that the drug remained stable under all relevant conditions. Finally, the validated method was successfully applied to evaluate the ability of chitosan-graft-β-cyclodextrin/PVA hydrogels to load and sustain release of VEM. The drug entrapment efficiency (DEE%) was between 65±0.08% and 70±0.05%.

Diabetes mellitus represents a growing global health challenge, establishing oral antidiabetic drugs as a therapeutic class of paramount importance. Consequently, to ensure drug safety, therapeutic efficacy, and accurate therapeutic drug monitoring, robust, selective and highly reproducible analytical methods are indispensable. This review provides a comprehensive overview of instrumental analytical techniques developed between 1985 and 2026 for quantifying oral antidiabetic agents. Based on a literature search across major databases—including PubMed, ScienceDirect and Springer—we systematically evaluated the application of UV-VIS spectrophotometry, liquid chromatography, and advanced electroanalytical methods. The reviewed literature encompasses methodologies ranging from routine quality control of bulk powders and pharmaceutical formulations to complex bioanalytical and pharmacokinetic applications. Liquid chromatography–tandem mass spectrometry (LC-MS/MS) emerged as the gold standard for precise bioanalysis due to its superior selectivity and sensitivity in complex biological matrices. Conversely, UV-VIS spectrophotometry and electrochemical methods remain highly relevant for cost-effective routine quality control in industrial manufacturing. Methodologies are categorized by drug therapeutic class, including biguanides, sulfonylureas, DPP-4 inhibitors, SGLT2 inhibitors, meglitinides, highlighting key validation parameters such as linearity, accuracy, and limits of detection. Finally, this review outlines future directions in the field of antidiabetic drug quantification.

Four decades have elapsed since orally disintegrating tablets (ODTs) were first formulated as the emulsion/type Lyoc tablet, a porous mass intended to rapidly disperse in saliva. Following the lyophilization process, new formulations of ODTs were designed, intending to make a simpler and more reproducible formulationZydis, LBL-Flash, Quicksolv, and, more recently, Zydis Ultra. Lyophilization is widely recognized as an effective technique for the development of ODTs, due to its ability to produce highly porous structures that enable rapid disintegration and improved patient compliance. However, its advantages should be considered in relation to other manufacturing methods, as each technology presents specific trade-offs in terms of cost, scalability, mechanical strength, drug loading capacity, and process robustness. In line with the modern sustainable and green pharmacy trend, new raw materials have gained attention as excipients for lyophilized ODTs; these materials include certain plant derivatives, but also performant excipients with newly discovered functionalities. At present, a new generation of ODTs is available in the form of Self-Nanoemulsifying Lyophilized Tablets (SNELTs), which bring the advantages of Self-Nanoemulsifying Drug Delivery Systems (SNEDDS) into ODTs via the lyophilization method. The technique is mostly applicable to low-solubility drugs formulated as nanoemulsions, which are absorbed onto solid carriers and further lyophilized, forming the final ODT. Despite its limitations (expensive, time-consuming, and high product friability), lyophilization is being continuously developed nowadays, in combination with other techniques (3D printing, mucoadhesion, or electrospinning), building hybrid platforms for the modern ODTs of the future.
