Phytophthora capsici is a notorious fungus which infects many crop plants at their early and late growth stages. In the present study, twelve P. capsici isolates were morphologically characterized, and based on pathogenicity assays; two highly virulent isolates causing post-emergence damping-off on locally cultivated chilli pepper were screened. Two P. capsici isolates, HydPak1 (MF322868) and HydPk2 (MF322869) were identified based on internal transcribed spacer (ITS) sequence homology. Plant growth promoting rhizobacteria (PGPR) play a significant role in disease suppression and plant growth promotion in various crops. Out of fifteen bacterial strains recovered from chilli rhizosphere, eight were found potential antagonists to P. capsici in vitro. Bacterial strains with strong antifungal potential were subjected to biochemical and molecular analysis. All tested bacterial strains, were positive for hydrogen cyanide (HCN), catalase production and indole-3-acetic acid (IAA) production (ranging from 6.10 to 56.23 µg ml−1), while siderophore production varied between 12.5 and 33.5%. The 16S rRNA sequence analysis of tested bacterial strains showed 98–100% identity with Pseudomonas putida, P. libanensis, P. aeruginosa, Bacillus subtilis, B. megaterium, and B. cereus sequences available in the National Center for Biotechnology Information (NCBI) GenBank nucleotide database. All sequences of identified bacteria were submitted to GenBank for accessions numbers (MH796347-50, MH796355-56, MH801129 and MH801071). Greenhouse studies concluded that all tested bacterial strains significantly suppressed the P. capsici infections (52.3–63%) and enhanced the plant growth characters in chilli pepper. Efficacy of many of these tested rhizobacteria is being first time reported against P. capsici from Pakistan. Plant growth promoting rhizobacteria (PGPR) exhibiting multiple traits may be used in the development of new, eco-friendly, and effective bioformulations as an alternative to synthetic fungicides.

In South Asian countries, the lucrative production of peaches has been seriously threatened by an assortment of biotic stresses especially nematodes. This situation compromises the achievement of sustainable development goals (SDGs) related to food security and zero hunger. Recently under changing climate, root-knot nematodes of the genus Meloidogyne have emerged as the most damaging phytopathogenic nematodes, while the efficacy of chemical control has remained limited. Therefore, a study was executed to assess the efficacy of four biocontrol agents including Pochonia chlamydosporia, Purpureocillum lilacinum, Trichoderma harzianum, and T. viride (at concentrations of 2.5 × 103, 5 × 103, 7.5 × 103, and 1 × 104) along with nematicide Rugby and a control treatment against Meloidogyne javanica on peach. The response variables included nematode infestations in terms of number of galls, egg masses, and reproductive factors. P. lilacinus and T. harzianum (1 × 104 concentration) reduced the number of galls by 18% and 16%, respectively, than the control. All biocontrol agents exhibited their effectiveness by significantly reducing number of egg masses, eggs per egg mass, and reproductive factors, while these remained statistically at par to each other. The study proved that application of these biocontrol agents holds potential for controlling root-knot nematodes and might be developed as a potent strategy to replace or at least reduce the use of traditional chemicals for avoiding environmental pollution and contamination.

Sugarcane crop constitutes one of the most vital sources of sugar and bioenergy globally; however, higher level of polyploidy makes its genome editing an intricate task. Recently, genome editing has become easier with CRISPR/Cas9 system that uses Cas9 to target sequence-specific regions and introduce double-strand breaks into the target region. This technique has been successfully employed to develop new varieties of sugarcane having desired phenotypic and physiological traits. Several genes can be fused with the CRISPR/Cas9 system leading to successful metabolic engineering and biological improvement for ensuring sustainable enhancement in sugarcane production. This simple RNA-guided genome editing technique has become a revolutionary tool and innovative application in biology that might be effectively employed for inducing specified genomic modifications in plant tissues. This review attempts to synthesize fundamentals of genome editing techniques with an ultimate aim to find out opportunities and challenges of sugarcane genome editing. It also tends to summarize the advances and achievements of gene editing via CRISPR-based genome editing technique for numerous field crops. Lastly, the enormous potential of CRISPR/Cas9 for gene editing in sugarcane, major challenges and future perspectives have been objectively elaborated.

Drought variability analysis is of utmost concern for planning and efficiently managing water resources and food security in any specific area. In the current study, drought spell occurrence has been investigated in the Balochistan province of Pakistan during the past four decades (1981-2020) using standardized precipitation index (SPI), reconnaissance drought index (RDI), and precipitation deciles (PD) at an annual timescale. Precipitation and temperature data collected from 13 synoptic meteorological stations located in Balochistan were used to calculate the SPI, the RDI, and the PD for calculation of drought severity and duration. Based on these indices, temporal analysis shows adverse impacts of drought spells in Nokkundi during 1991-1993, in Barkhan, Dalbandin, Quetta stations during 1999-2000, whereas Barkhan, Dalbandin, Lasbella, Sibi during 2002-2003, Zhob during 2010-2011, Kalat and Khuzdar during 2014-2015, and Panjgur during 2017-2018. Also, the aridity index for each station was calculated based on the UNEP method shows that major part of Balochistan lies in the arid zone, followed by the hyper-arid in the southwestern part and the semi-arid zones in the northeastern part of the province. SPI and RDI results were found more localized than PD, as PD shows extensive events. Furthermore, principal component analysis shows a significant contribution from all the indices. For SPI, RDI, and PD, the first three principal components have more than 70% share, contributing 73.63%, 74.15%, and 72.30% respectively. By integrating drought patterns, long-term planning, and preparedness to mitigate drought impacts are only possible. The RDI was found more suitable and recommended in case of temperature data availability.

The potential interactions of rhizobium bacteria in enhancing nodulation, nitrogen (N) fixation for boosting N availability, and the yield of black gram under a temperate environment continue to remain unexplored. Therefore, this study aimed to evaluate the agronomic performance of black gram cultivars, their yield comparisons, and shoot–grain–soil N dynamics in a prevalently rainfed farming system. Two black gram cultivars, NARC Mash-I and NARC Mash-II, were subjected to rhizobia inoculation combined with different N doses (0, 25, 50, 75, 100 kg ha−1). The response variables included root nodulation, agronomic yield attributes, grain yield, shoot–grain and soil N dynamics, and biological productivity. Black gram cultivar NARC Mash-II showed the maximum nodule formation (41 per plant), while each nodule obtained 0.69 g weight in response to RI combined with 25 kg N ha−1. Additionally, this combination showed the highest pods per plant and thousand grain weight, which maximized the grain yield (1777 kg ha−1) and biological productivity (3007 kg ha−1). In contrast, NARC Mash-I under 50 kg N recorded the highest shoot N content, while the same cultivar under 100 kg N exhibited the maximum soil N content. The correlation analyses indicated a significantly robust association among the nodule numbers, grain weight, and N contents in different plant organs. These results give mechanistic insights into plant–microbe interactions based on the eco-friendly, sustainable, and smart agricultural practice of black gram production in a temperate environment.
