Cadmium (Cd) is highly toxic to living organisms. This study aimed to elucidate the regulation of gene expression in rice shoots under Cd stress. Rice plants were exposed to 0, 50, 75, 100 μmol/L CdCl2 in hydroponic culture for 7 d. Transcriptional changes in rice shoots were examined by transcriptome sequencing techniques. A total of 2197 DEGs (987 up-regulated and 1210 down-regulated) were detected in rice shoots under the exposure of 75 μmol/L CdCl2. GO and KEGG enrichment analyses showed that genes encoding auxin-responsive protein IAA and peroxidase were up-regulated, while genes encoding proteins involved in signal transduction, including TIFY family, ERF and bZIP were down-regulated. Abundant ROS related terms were also identified and grouped into significantly differentially expressed GO terms, including oxidoreductase activity, catalytic activity, oxidation-reduction process, confirming the enhanced oxidative stress of Cd. Genes encoding photosystem I reaction center subunit and photosynthetic NDH subunit of luminal location were up-regulated in pathway of energy metabolism, suggesting an interference of photosynthesis by Cd stress. Our results improve the understanding of the complex molecular responsive mechanisms of rice shoots under Cd stress.

The coexistence of microplastics and benzo[a]pyrene (BaP) in the environment, and their interactions within agricultural soils in particular, have garnered widespread attention. This study focused on the early-stage interactions between microplastics and BaP, aiming to uncover their initial adsorption mechanisms. Despite the significant environmental toxicity of both pollutants, research on their mutual interactions in soil is still limited. This study conducted adsorption thermodynamics and kinetics experiments to explore the effects and mechanisms of various microplastics (polyethylene (PE), polystyrene (PS), and polyvinyl chloride (PVC)) on the adsorption of BaP. Using advanced techniques such as scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), and Fourier transform infrared (FTIR) spectroscopy, this study explored the surface characteristics of microplastics and their interactions with BaP. The results demonstrated that PVC microplastics exhibited the highest adsorption capacity for BaP, which was primarily due to π–π interactions and increased hydrophobicity. In the soil–microplastic blend systems, BaP was predominantly found on microplastics, enhancing the soil’s adsorption capacity for BaP, particularly PVC, which showed an adsorption capacity 3.69 times greater than that of soil alone. Density functional theory (DFT) simulation calculations indicated that the binding energy of BaP for PVC pretreated with soil was −59.16 kJ/mol, whereas it was −53.02 kJ/mol for untreated PVC, −39.35 kJ/mol for PE, and −48.84 kJ/mol for PS. These findings suggest that soil pretreatment enhances the adsorption stability of PVC for BaP, further elucidating the potential mechanisms behind the increased adsorption capacity in the soil–microplastic system. These findings confirm that microplastics serve as effective vectors for organic pollutants such as BaP, significantly influencing their environmental behavior in soils, and provide essential theoretical support for assessing the environmental toxicity and migration behaviors of microplastics and associated organic contaminants.

Increased application of organic fertilizer is an effective measure to improve greenhouse soil quality. However, prolonged and intensive application of organic manure has caused nutrient and certain heavy metal accumulation in greenhouse soil. Therefore, the optimal quantity of organic manure required to sustain soil fertility while mitigating the accumulation of heavy metals and other nutrients resulting from continuous application remains unclear. This study evaluated the impacts of sustained and reduced organic manure application on soil physicochemical properties, heavy metal contents, and microbial community through a 9-year greenhouse field experiment. Treatments included a control without any fertilizer (CK), conventional manure (M), and three reduced manure treatments (−25%M, −37.5%MNPK, and −50%MNPK). Compared to CK, either M treatment or manure reduction treatments either maintained or significantly elevated soil pH and soil organic matter, total nitrogen, total phosphorus, and available phosphorus. Notably, −37.5%MNPK exhibited further increases in the available nitrogen and potassium. The M treatment significantly increased in the total concentrations of cadmium, copper, lead, zinc, and the availability of chromium and zinc. However, reduced manure treatments showed no change or a significantly reduced in heavy metal availability. The −25%M and −37.5%MNPK treatments significantly improved bacterial diversity. Reducing organic manure altered microbial taxa abundance. The soil pH emerged as the primary driving factor for variation in the bacterial community structure, whereas available nitrogen, potassium, and lead were the key factors influencing fungal community structural changes. These results indicate that reducing excessive organic manure input is an effective strategy to control heavy metal accumulation, enhance soil fertility, and optimize microbial community structure.

Selenium (Se) is an essential micronutrient for the human body and is closely linked to health. Rice (Oryza sativa L.), as a major staple food globally, is the primary source of Se intake for humans. To better achieve Se biofortification in rice, it is crucial to understand the molecular mechanisms behind rice’s response to different Se concentrations. This study investigates the morphological and transcriptomic responses of rice seedlings to low (1 µM, LSe) and high (10 µM, HSe) Se concentrations compared to a control (CK). Morphological analyses revealed that LSe promoted growth, enhancing shoot and root length and biomass, whereas HSe treatment inhibited these parameters, indicating Se’s dual role in rice growth. Notably, the most significant promotion of rice growth was observed at the Se concentration of 1 µM. The organic Se content and antioxidant enzyme activities (SOD, POD and CAT) in rice seedlings also reached their maximum values simultaneously. Total RNA was extracted for transcriptome sequencing, and differential gene expression analysis was conducted using DESeq2. Transcriptomic sequencing highlighted distinct responses under LSe and HSe conditions. Gene ontology (GO) enrichment analysis revealed significant involvement in processes related to oxidoreductase activity and cellular structures. KEGG pathway analysis emphasized that Se treatments notably enhanced the glutathione metabolism pathway, which is crucial for antioxidant defense. Additionally, significant changes were observed in starch and sucrose metabolism and cysteine (Cys) and methionine (Met) metabolism pathways, showing upregulation under LSe treatment and downregulation under HSe. Six key genes were validated using qRT-PCR, confirming their differential expression under varied Se treatments. The Cys, Met and starch content assays as well as qRT-PCR data demonstrated that LSe promoted the synthesis and accumulation of Cys, Met and starch, supporting enhanced growth and antioxidant capacity. Conversely, HSe inhibited the synthesis and accumulation of Cys, Met and starch in rice seedlings, and these data were also consistent with the physiological phenotype. These findings provide insights into the molecular mechanisms by which rice seedlings adapt to varying Se levels, with implications for Se biofortification and stress management strategies in crops.
