Drought stress during sorghum emergence significantly affects seedling establishment, adversely affecting both emergence and population growth. This study investigates drought tolerance mechanisms during sorghum germination by analyzing physiological changes and transcriptomic data from two lines: W069 (drought tolerant) and W040 (drought sensitive). Under drought conditions, a phenotypic analysis revealed that W069 exhibited longer shoots and roots than W040. Additionally, physiological data indicated higher osmotic substance and lower malondialdehyde levels in W069. Using Kyoto Encyclopedia of Genes and Genome analyses, we identified three key pathways (starch and sucrose metabolism, phenylpropanoid biosynthesis, and phytohormone signaling) as pivotal in the drought response during seed germination in sorghum plants. Expression profiling revealed that most drought tolerance-related genes in the three key pathways were expressed at higher levels in the drought-tolerant cultivar W069, possibly explaining its greater stress tolerance. These findings enhance our understanding of drought-responsive gene networks in sorghum seed germination, offering potential target genes and strategies for enhancing drought tolerance in this crop.

The flavour of foxtail millet (Setaria italica (L.) P. Beauv.) is an important indicator for evaluating the quality of the millet. The volatile components in steamed millet porridge samples were analysed using electronic nose (E-Nose) and gas chromatography–ion mobility spectrometry (GC-IMS) techniques, and characteristic volatile fingerprints were constructed to clarify the differences in the main flavour substances in different foxtail millet varieties (two hybrids and two conventional foxtail millets). After sensory evaluation by judges, Jingu 21 (JG) scored significantly higher than the other varieties, and the others were, in order, Jinmiao K1 (JM), Changzagu 466 (CZ) and Zhangzagu 3 (ZZ). E-Nose analysis showed differences in sulphides and terpenoids, nitrogen oxides, organosulphides and aromatic compounds in different varieties of millet porridge. A total of 59 volatile components were determined by GC-IMS in the four varieties of millet porridge, including 23 aldehydes, 17 alcohols, 9 ketones, 4 esters, 2 acids, 3 furans and 1 pyrazine. Comparative analyses of the volatile components in JG, JM, ZZ and CZ revealed that the contents of octanal, nonanal and 3-methyl-2-butenal were higher in JG; the contents of trans-2-butenal, 2-methyl-1-propanol, trans-2-heptenal and trans-2-pentenal were higher in JM; and the contents of 2-octanone, hexanol, 1-octen-3-ol, 2-pentanone and butyraldehyde were higher in ZZ. The contents of 2-butanol, propionic acid and acetic acid were higher in CZ. A prediction model with good stability was established by orthogonal partial least squares discriminant analysis (OPLS-DA), and 25 potential characteristic markers (VIP > 1) were screened out from 59 volatile organic compounds (VOCs). These volatile components can be used to distinguish the different varieties of millet porridge samples. Moreover, we found conventional foxtail millet contained more aldehydes than the hybridised foxtail millet; especially decanal, 1-nonanal-D, heptanal-D, 1-octanal-M, 1-octanal-D and 1-nonanal-M were significantly higher in JG than in the other varieties. These results indicate that the E-Nose combined with GC-IMS can be used to characterise the flavour volatiles of different foxtail millet, and the results of this study may provide some information for future understanding of the aroma characteristics of foxtail millet and the genetic improvement of hybrid grains.

Background/Objectives: Cyclic nucleotide-gated channel (CNGC) genes play crucial roles in plant growth, development, and stress responses, yet their functions in sorghum remain poorly understood. Methods: This study systematically analyzed sorghum CNGC genes through genome-wide identification, encompassing chromosomal mapping, phylogenetic relationships, gene structure, cis-acting elements, miRNA regulation, and GO/KEGG annotation. Results: A total of 23 sorghum CNGC genes were identified and classified into five subclasses (I–IV-b), exhibiting high evolutionary conservation with rice and maize. Promoter and miRNA analyses revealed multi-level regulation involving light, hormones (ABA, JA), and stress response elements. Several SbCNGC genes were predicted to be regulated by multiple miRNAs. Expression profiling and qRT-PCR validation indicated that most SbCNGC genes responded to both high-temperature and low-temperature stress. Expression analysis revealed tissue specificity and stress-induced transcriptional responses. Notably, SbCNGC1 remains consistently upregulated under both cold and heat stress, suggesting a potential key role in Ca2+-mediated signaling. Conclusions: This study systematically characterizes SbCNGC genes for the first time, reveals their potential role in abiotic stress tolerance, and provides a valuable resource for sorghum functional genomics and molecular breeding.

Background: Sorghum (Sorghum bicolor L.) is an adversity-tolerant crop, but the function of the DUF966 gene family in its growth, development, and stress tolerance is unclear. Methods: The SbDUF966 gene was identified and analyzed using bioinformatics methods in this study. We also analyzed the expression pattern of SbDUF966 in different tissues and stress conditions using RNA-seq and RT-qPCR. We explored its post-transcriptional regulatory mechanism by combining it with miRNA analysis. Results: A total of six SbDUF966 genes were identified and categorized into two groups (Group I and Group II). Gene expression analysis showed that SbDUF966 exhibited specific expression in different tissues and developmental stages, and the expression response to abiotic stresses such as low temperature, high temperature, salinity, and flooding varied over time. In addition, 12 sorghum miRNAs were predicted as potential regulators of SbDUF966. Conclusions: The SbDUF966 gene family likely regulates sorghum’s growth, development, and stress tolerance.

Continuous sorghum cropping causes yield decline in semi-arid regions. In a two-year field experiment conducted on plots previously cropped with sorghum for two years, four fertilisation treatments—no fertiliser (SCK), chemical fertiliser (SCF), organic–inorganic compound fertiliser (SOIF) and compound microbial fertiliser (SCMF; SOIF supplemented with Bacillus subtilis and B. licheniformis)—were compared in terms of grain yield, soil physicochemical properties, potential enzyme activities and rhizosphere bacterial and fungal communities. Year, treatment and year × treatment effects were tested using mixed models that accounted for repeated measurements of the same plots. The complete SCMF formulation was associated with the highest grain yield (10,108.13 kg ha−1, in 2025), exceeding those unde SCK, SCF, and SOIF by 42.95%, 11.47%, and 3.59%, respectively. The SCMF-SOIF comparison, in which N, P2O5, K2O and organic-matter inputs were identical, isolates the microbial inoculants; its yield increment was small and not significant in either year (4.27% and 3.59%), so an inoculant-specific yield benefit was not demonstrated. Compared with SOIF, SCMF had higher alkaline phosphatase (ALP) activity in both years and higher sucrase (SUS) activity (+36.80%) and available nitrogen (AN; +21.11%) in 2025. In 2025, SOIF and SCMF reduced the topsoil bulk density relative to SCK and SCF, and SCMF shifted the aggregate-size distribution towards the 2–0.25-mm fraction (+47.45–112.62%). Fertilisation altered the composition, but not the alpha diversity, of rhizosphere communities; the treatment effect was larger and more independent of multivariate dispersion for fungi (R2 = 0.167, p = 0.009) than for bacteria (R2 = 0.113, p = 0.049). The fungal response represents an overall treatment-associated compositional difference with weaker statistical evidence in the second year, because treatment differences were significant only in 2024 when each year was tested separately. Nitrosospira, a genus containing members previously associated with ammonia oxidation, was consistently enriched under SCMF, although nitrification was not measured. Overall, the yield benefit was attributable mainly to organic–inorganic fertilisation, whereas inoculant-associated differences were found in potential enzyme activities and fungal community composition rather than in grain yield.
