Inflammatory bowel disease remains a significant challenge in clinical settings. This study investigated the therapeutic potential of sea cucumber ovum hydrolysates (SCH) in a dextran sulfate sodium (DSS)-induced colitis mouse model. SCH, defined by its elevated stability and solubility, with a molecular weight below 1000 Da, significantly alleviated DSS-induced colitis, as evidenced by enhanced splenic index, reduced colonic damage, and diminished serum pro-inflammatory cytokines. Furthermore, macrogenomic analysis demonstrated that SCH increased beneficial gut microbes and decreased pro-inflammatory bacteria. Furthermore, metabolomic analysis of colonic tissues identified elevated levels of anti-inflammatory metabolites, such as Phenyllactate, 2-Hydroxyglutarate, and L-Aspartic acid, in colitis mice after oral administration of SCH. In conclusion, SCH represents a promising candidate for the treatment of colitis.

This study aimed to assess the impact of alcalase, trypsin, flavourzyme, and neutrase on the characteristics, antioxidant activity stability, and anti-fatigue activity of hydrolysates derived from Cucumaria frondosa tentacles (CFTHs). The results demonstrate that favourzyme hydrolysates exhibited the highest degree of hydrolysis (DH). Zeta potential and particle size measurements indicated that hydrolyzed peptides treated with favourzyme appeared aggregated and exhibited larger particle sizes. The antioxidant properties of CFTHs demonstrated good thermal stability, pH stability, and enhanced simulated gastrointestinal digestive stability. The anti-fatigue activity of CFTHs was examined using an acute exercise fatigue model. The results indicate that CFTHs extended the exhaustive swimming time of mice to 17.81 min. Additionally, CFTHs significantly elevated (p < 0.01) blood glucose (Glu) and liver glycogen (LG) levels, while also decreasing (p < 0.05) the concentrations of metabolites such as lactic acid (LA), urea nitrogen (BUN), creatine kinase (CK), lactate dehydrogenase (LDH), and ammonia (NH3). This reduction contributed to the alleviation of fatigue in the body. Furthermore, the levels of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were significantly increased (p < 0.05), which aided in mitigating oxidative damage induced by strenuous exercise. These findings strongly support the potential utilization of CFTHs in food products as natural antioxidant and anti-fatigue alternatives.

Squid pen (SP) was found to contain 64.41% protein and 26.03% chitin. The amino acid composition revealed that Met was the most abundant amino acid in SP, with a concentration of 13.67 g/100 g. To enhance the stability and bioavailability of SP hydrolysates, microcapsules were developed using ultrasonic emulsification techniques with SP trypsin hydrolysates (SPTH) and SP β-chitosan (SPC). The optimal preparation conditions involved using a 2% concentration of SPC, a 4 mg/mL concentration of SPTH, a core-to-wall ratio (v/v) of 1:3 for SPTH/SPC, and subjecting them to ultrasonic treatment for 20 min. These microcapsules had a loading capacity of 58.95% for SPTH under these conditions. The successful encapsulation of SPTH in the SPC complex to form SPC-SPTH microcapsules was confirmed by FTIR, XRD, DSC, and SEM, exhibiting good thermal stability, small particle size, and high encapsulation efficiency. In vitro digestion studies demonstrated a release of 15.61% in simulated gastric fluid and 69.32% in intestinal fluid, achieving targeted release in the intestines. The digested products exhibited superior antioxidant activity compared to free SPTH digests, suggesting that microencapsulation effectively preserves SPTH bioactivity. This study enhances the bioavailability of SPTH and offers a promising delivery system for natural compounds with low bioavailability and stability.

Sea cucumber ovum are high-value compounds that remain after the processing of sea cucumbers, and their optimal utilization has long posed a challenge. In this research, we systematically examined the therapeutic effects of sea cucumber ovum hydrolysate (SCH) on premature ovarian failure (POF) and its underlying mechanism. We utilized a model of ICR mice induced with 100 mg/kg cyclophosphamide (CP) to evaluate the therapeutic influence of SCH on ovarian performance. The ovarian and uterine indices were significantly decreased in the POF group compared to the control group; however, these trends were notably reversed following SCH intervention. The therapeutic effects of SCH were positively reflected by the alterations induced by CP in levels of estradiol (E2), follicle-stimulating hormone (FSH), testosterone (T), luteinizing hormone (LH), and anti-Müllerian hormone (AMH). Regarding oxidative stress, SCH was found to enhance superoxide dismutase (SOD) activity and decrease malondialdehyde (MDA) levels, while also alleviating apoptosis in ovarian granulosa cells. Metabolomics analysis revealed hypoxanthine, mannitol, neocnidilide, tryptophan, palmitoleic acid, and protoporphyrinogen IX as potential biomarkers. In conclusion, SCH effectively improves POF induced by CP, thereby reinforcing the potential application of SCH in the domain of functional foods.

Inflammatory bowel disease continues to pose substantial therapeutic challenges in modern gastroenterology. This study systematically evaluated the anti-colitis efficacy of Cucumaria frondosa tentacles hydrolysates (CFTHs) using a dextran sulfate sodium (DSS)-induced murine colitis model. Characterized by enhanced stability and solubility with molecular weights below 1000 Da, administration of CFTHs demonstrated a significant mitigation in colitis pathology. Therapeutic outcomes included an improved splenic index, attenuated colonic mucosal damage, and substantial decreases in serum pro-inflammatory cytokines. Relative to the DSS group, the MPO value in the CFTHs-H group decreased by 27.6%, and the IL-6 value exhibited a reduction of 33%. Metagenomic profiling revealed that CFTHs mediated gut microbiota modulation, particularly the enrichment of beneficial Bacteroidetes and suppression of pro-inflammatory Proteobacteria. Metabolomic analysis identified elevated colonic concentrations of anti-inflammatory metabolites such as gamma-linolenic acid and prostaglandin I2, suggesting a microbiome–metabolome crosstalk in the therapeutic mechanism. These multi-omics findings in a murine model suggest that CFTHs may represent a promising candidate for future studies as a nutraceutical intervention for inflammatory bowel disorder. This intervention may operate through mechanisms that include simultaneous immunomodulation, microbiota restoration, and metabolic reprogramming.
