Flavonoid profile and antioxidant activity of citrus peels, pulps, and juices from 27 local citrus cultivars in China were investigated. Flavonoid composition and content were determined using UPLC-PDA. Total phenolic content (TPC) and total flavonoid content (TFC) were measured using a Folin–Ciocalteau reagent and Al(NO3)-NaNO2 complexometry, respectively. The antioxidant capacities of the extracts were evaluated by DPPH, ABTS and FRAP method, respectively. Citrus peel not only exhibited better antioxidant potential, but also presented more composition diversity and contained higher concentrations of flavonoids than pulp and juice. Different citrus species were characterized by their individual predominant flavonoids, contributing largely to the antioxidant activity, such as mandarin was characterized by hesperidin, nobiletin and tangeretin, while pummelo and papeda were characterized by naringin. The peel of Guihuadinanfeng (Citrus reticulata) had the highest TPC of 23.46 mg equivalent gallic acid/g DW (dry weight) and TFC of 21.37 mg equivalent rutin/g DW. Shiyueju (C. reticulata) peel showed the highest antioxidant capacity based on the antioxidant potency composite (APC) analysis. Overall, mandarin (C. reticulata) fruits peel contained more TPC and TFC, exhibiting higher antioxidant capacities than other species, and were good natural sources of flavonoids and antioxidants.

A major citrus postharvest pathogen, Penicillium italicum (P. italicum), causes substantial economic losses in citrus. In this study, a citral nanoemulsion containing polymethoxylated flavonoids (PMFs), the antimicrobial compounds from citrus, was prepared. The antifungal activity and potential antifungal mechanisms of the nanoemulsion against P. italicum were evaluated. The results showed that the growth of P. italicum was effectively inhibited by the nanoemulsion, with a minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) of 62.5 and 250 mg L−1, respectively. The nanoemulsion significantly inhibited spore germination and mycelial growth, and it altered the morphology of P. italicum. In addition, the permeability of the cell membrane increased with increasing nanoemulsion concentrations, as evidenced by a rapid rise in extracellular electric conductivity and stronger red fluorescence from mycelia (propidium iodide staining). Compared with the control, the nanoemulsion treatment induced a decrease in total lipid and ergosterol contents in P. italicum cells by 64.61% and 60.58%, respectively, demonstrating that membrane integrity had been disrupted. The results indicated that the PMFs-loaded nanoemulsion exerted antifungal activity against P. italicum by disrupting cell membrane integrity and permeability; such a nanoemulsion may be used as a potential fungicide substitute for preservation in citrus fruits.

The anti-aging activity of many plant flavonoids, as well as their mechanisms of action, have been explored in the current literature. However, the studies on the synergistic effects between the different flavonoid compounds were quite limited in previous reports. In this study, by using a high throughput assay, we tested the synergistic effects between different citrus flavonoids throughout the yeast’s chronological lifespan (CLS). We studied the effect of four flavonoid compounds including naringin, hesperedin, hesperitin, neohesperidin, as well as their different combinations on the CLS of the yeast strain BY4742. Their ROS scavenging ability, in vitro antioxidant activity and the influence on the extracellular pH were also tested. The results showed that neohesperidin extended the yeast’s CLS in a concentration-dependent manner. Especially, we found that neohesperidin showed great potential in extending CLS of budding yeast individually or synergistically with hesperetin. The neohesperidin exhibited the strongest function in decreasing the reactive oxygen species (ROS) accumulation in yeast. These findings clearly indicated that neohesperidin is potentially an anti-aging citrus flavonoid, and its synergistic effect with other flavonoids on yeast’s CLS will be an interesting subject for future research of the anti-aging function of citrus fruits.

Plant extracellular vesicles (PEVs) have attracted increasing attention due to their rich composition, good antioxidant and anti-inflammatory activity, and ability to transport drugs. As a common fruit, citrus is an ideal material for extracting PEVs because of the diversity and abundance of bioactive substances in it. In our study, citrus-derived extracellular vesicles (CEVs) were extracted from red mandarin (Citrus reticulata Blanco cv. ‘Dahongpao’) and it was found that they contain high levels of lipids, proteins, and carbohydrates. The high levels of total phenols and total flavonoids suggest that CEVs have good chemical antioxidant properties. We also demonstrated through cell experiments that CEVs have significant antioxidant and anti-inflammatory effects. Furthermore, we found that CEVs have an encapsulation rate of 71.5 ± 0.19% and a high drug-carrying capacity of 4.96 ± 0.22% and can enhance antioxidant and anti-inflammatory activity when loaded with tangeretin. Our results show that CEVs contain abundant bioactive components, have low toxicity, exhibit good antioxidant and anti-inflammatory properties, and can serve as drug delivery agents. This study has important implications for utilizing citrus materials and developing natural anti-oxidative and anti-inflammatory biomaterials.

As one of the most widely consumed citrus fruits, Citrus reticulata Blanco cv. Ponkan is known to have a number of dietary and medicinal values. In this work, we developed a new method using ultra performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS) for qualitative analysis and profiling of chemical compositions in Ponkan peel methanol extract. With this method, the elution and separation of chemical components in Ponkan peel could be easily completed within seven minutes. The method was much faster than other analytical methods previously used with citrus fruits. According to the chromatography retention time, UV spectrum, and exact molecular weight and high energy fragment ions combined with information from reference standards or literature, a total of 32 bioactive compounds were unambiguously identified or tentatively characterized in the Ponkan peel extract. The compounds included 8 flavonoid-O-glycosides, 5 flavone-C-glycosides, 1 dimethoxyflavone, 10 polymethoxyflavones and 8 other compounds. Among these compounds, caffeoyl-glucose, dihydrophaseic acid glycosil ester, ferulic acid hexose, luteolin-6,8-di-C-glucoside, luteolin-8-C-glucoside, narirutin 4′-glucoside, nomilinic acid glucoside, isolimonic acid, dihydroxy-dimethoxyflavone, and 5,7,4′-trimethoxyflavone were characterized for the first time in Ponkan peel to our knowledge. These results showed the UPLC-Q-TOF-MS was a powerful tool to analyze the structures of chemical compounds in Ponkan peel. And our data will provide useful information for further evaluating the bioactivity of Ponkan peel and the utilization of Ponkan resource.
