Nano- and microemulsions are colloidal systems that are widely used in various fields of biomedicine, including wound and burn healing, cosmetology, the development of antibacterial and antiviral drugs, oncology, etc. The stability of these systems is governed by the balance of molecular interactions between nanodomains. Microemulsions as a colloidal form play a special important role in stability. The microemulsion is the thermodynamically stable phase from oil, water, surfactant and co-surfactant which forms the surface of drops with very small surface energy. The last phenomena determines the shortage time of all fluid dispersions including nanoemulsions and emulgels. This review examines the theory and main methods of obtaining nano- and microemulsions, particularly focusing on the structure of microemulsions and methods for emulsion analysis. Additionally, we have analyzed the main preclinical and clinical studies in the field of wound healing and the use of emulsions in cancer therapy, emphasizing the prospects for further developments in this area.

The colloidal long-storage stability of nanosized drugs is a crucial factor for pharmacology, as they require much time for robust estimation. The application of bioavailable magnetic nanosuspensions in theranostics is limited by incomplete information about their colloidal stability in the internal media of human organisms. A method for the accelerated temperature stress “aging” of magnetic nanosized suspensions is proposed for the rapid assessment and prediction of the colloidal stability over time of nanosized iron oxide suspensions stabilized by albumin HSA. Colloidal stability is assessed using dynamic light scattering (DLS), fluorescence spectroscopy, electrophoresis, and ion monitoring methods during short- and long-term storage. Rapid assessment is achieved by short high-temperature (70 °C) processing of carboxymethyl-dextran-coated nanosol in the presence of albumin. The role of albumin in the sustained stability of superparamagnetic iron oxide particles (SPIONs) was studied under conditions mimicking blood plasma (pH = 7.4) and endolysosomal cell compartments (pH = 5.5). According to the fluorescence quenching and DLS data, colloidal stability is ensured by the formation of an HSA corona on carboxymethyl-dextran-coated SPIONs and their process of clustering. In the presence of albumin, the colloidal stability of nanoparticles is shown to increase from 80 to 121 days at a storage temperature of 8 °C The prognostic shelf life of magnetic nanosol is estimated by calculating the Van’t Hoff’s relation for the rate of chemical reactions. The validity of using the Van’t Hoff’s rule is confirmed by the agreement of the calculated activation energy at 8 °C and 70 °C. The developed method of the accelerated aging of nanoparticles can not only be employed for the estimation of the shelf life of magnetic nanoparticles coated with HSA in vitro but also for assessing the stability of SPIONs applied in vivo.

Type I interferons, particularly IFNα-2b, play essential roles in eliciting adaptive and innate immune responses, being implicated in the pathogenesis of various diseases, including cancer, and autoimmune and infectious diseases. Therefore, the development of a highly sensitive platform for analysis of either IFNα-2b or anti-IFNα-2b antibodies is of high importance to improve the diagnosis of various pathologies associated with the IFNα-2b disbalance. For evaluation of the anti-IFNα-2b antibody level, we have synthesized superparamagnetic iron oxide nanoparticles (SPIONs) coupled with the recombinant human IFNα-2b protein (SPIONs@IFNα-2b). Employing a magnetic relaxation switching assay (MRSw)-based nanosensor, we detected picomolar concentrations (0.36 pg/mL) of anti-INFα-2b antibodies. The high sensitivity of the real-time antibodies’ detection was ensured by the specificity of immune responses and the maintenance of resonance conditions for water spins by choosing a high-frequency filling of short radio-frequency pulses of the generator. The formation of a complex of the SPIONs@IFNα-2b nanoparticles with the anti-INFα-2b antibodies led to a cascade process of the formation of nanoparticle clusters, which was further enhanced by exposure to a strong (7.1 T) homogenous magnetic field. Obtained magnetic conjugates exhibited high negative MR contrast-enhancing properties (as shown by NMR studies) that were also preserved when particles were administered in vivo. Thus, we observed a 1.2-fold decrease of the T2 relaxation time in the liver following administration of magnetic conjugates as compared to the control. In conclusion, the developed MRSw assay based on SPIONs@IFNα-2b nanoparticles represents an alternative immunological probe for the estimation of anti-IFNα-2b antibodies that could be further employed in clinical studies.

Clinical experience indicates that enhanced level of heat shock protein 70 (Hsp70) and p53 correlates with poor prognosis due to malignant cell overexpression of these proteins in tumor progression. Cadmium selenide quantum dots (QDs) were synthesized in aqueous solution using mercaptopropionic acid and L-cysteine (L-Cys) as ligands. They were conjugated with a monoclonal antibody (Ab) to p53 and cmHp70.1 to Hsp70 for detection of cancer cell apoptosis that was demonstrated in the experiment by fluorescent confocal microscopy both for breast carcinoma cells and for thyroid tissue. It is shown that in comparison with organic dyes, quantum dots have superior photostability of tracking apoptosis in cancer cells for longer time.

The development of highly sensitive approaches for detecting tumor cells in biological samples remains a critical challenge in laboratory and clinical oncology. In this study, we investigated the structural and magnetic properties of iron oxide nanoparticles incorporated into cellulose microspheres of two size ranges (~100 and ~700 μm) and evaluated their potential for targeted tumor cell isolation. In the smaller microspheres, magnetite-based magnetic nanoparticles (MNPs) were synthesized in situ via co-precipitation, whereas pre-synthesized MNPs were embedded into the larger microspheres. The geometrical characteristics of the resulting magnetic cellulose microspheres (MSCMNs) were assessed by confocal microscopy. Transmission electron microscopy and X-ray diffraction analyses revealed an average magnetic core size of approximately 17 nm. Magnetic properties of the MNPs within MSCMNs were characterized using a highly sensitive nonlinear magnetic response technique, and their dynamic parameters were derived using a formalism based on the stochastic Hilbert–Landau–Lifshitz equation. To evaluate their applicability in cancer diagnostics and treatment monitoring, the MSCMNs were functionalized with a TKD peptide that selectively binds membrane-associated Hsp70 (mHsp70), yielding TKD@MSCMNs. Magnetic separation enabled the isolation of tumor cells from biological fluids. The specificity of TKD-mediated binding was confirmed using Flamma648-labeled Hsp70 and compared with control alloferone-conjugated microspheres (All@MSCMNs). The ability of TKD@MSCMNs to selectively extract mHsp70-positive tumor cells was validated using C6 glioma cells and mHsp70-negative FetMSCs controls. Following co-incubation, the extraction efficiency for C6 cells was 28 ± 14%, significantly higher than that for FetMSC (7 ± 7%, p < 0.05). These findings highlight the potential of TKD-functionalized magnetic cellulose microspheres as a sensitive platform for tumor cell detection and isolation.
