This review will focus on the structural modifications, structure–activity relationships, pharmacology, and clinical development of triptolide in the last forty years.

Direct C-2 amidation of indoles was reported using sulfonyl azides as the amino source to release N2 as the single byproduct. This reaction exhibits high functional group tolerance and regioselectivity, providing a variety of 2-amino substituted indoles in high to excellent yield. The procedure is robust, reliable, and compatible with water and air.

About Me
Yuanchao Li joined Sun Yat-sen University in 2017 and is currently an Associate Professor at the School of Materials Science and Engineering. Prior to this, he was a Postdoc at the Department of Chemical and Biomolecular Engineering, North Carolina State University (2014–2017), and at the Department of Chemistry, the University of North Carolina at Chapel Hill (UNC-Chapel Hill) (2013–2014). He graduated from the Department of Polymer Science and Engineering, University of Science and Technology of China, with a bachelor’s in 2008 and completed his Ph.D. at the Department of Chemistry, UNC-Chapel Hill, in 2013. His main research area is focused on polymer mechanochemistry and force-responsive materials.
