Probiotics have been shown to possess several properties, depending on the strain. Some probiotics have important roles in preventing infection and balancing the immune system due to the interaction between the intestinal mucosa and cells in the immune system. This study aimed to examine the properties of three probiotic strains using the tumor necrosis factor-alpha (TNF-α) inhibition test in colorectal adenocarcinoma cells (Caco-2 cells). It was revealed that the viable cells and heat-killed cells of the probiotic L. paracasei strain MSMC39-1 dramatically suppressed TNF-α secretion in Caco-2 cells. The strongest strains were then chosen to treat rats with colitis induced by dextran sulfate sodium (DSS). Viable cells of the probiotic L. paracasei strain MSMC39-1 reduced aspartate transaminase and alanine transaminase in the serum and significantly inhibited TNF-α secretion in the colon and liver tissues. Treatment with the probiotic L. paracasei strain MSMC39-1 alleviated the colon and liver histopathology in DSS-induced colitis rats. Furthermore, supplementation with probiotic L. paracasei strain MSMC39-1 increased the genus Lactobacillus and boosted the other beneficial bacteria in the gut. Thus, the probiotic L. paracasei strain MSMC39-1 exhibited an anti-inflammation effect in the colon and modulated the gut microbiota.

Introduction: The aim of this study was to investigate the effects of glabridin on vascular inflammation and vascular remodeling of the left anterior descending artery (LAD) in diabetic rat heart.Material and methods: Eighty male Wistar rats were divided into five groups: (1) control rats; (2) glabridin control rats that received diet supplemented with glabridin (CGB); (3) diabetic rats (STZ); (4) diabetic rats that received a supplemented with glabridin (STZ + GB) 40 mg/kg BW; and (5) diabetic rats treated with glyburide (STZ + GR) 4 mg/kg BW. Diabetic animals were rendered diabetic by injection of a single dose (60 mg/kg BW) of streptozotocin (STZ). After 8 weeks, the biochemical markers of oxidative stress were assessed. The three-dimensional morphology of LAD was examined by vascular corrosion casting. Enzyme-linked immunosorbent assay was applied to determine the expression of TNF-α and IL-1β proteins. Western blot and immunofluorescence analysis were used to detect the expression of VEGF and TGF-β1 secretions. Results: The lumen diameter of the LAD was notably smaller and stenotic. LAD analysis revealed arterial notch and evolved irregular caliber. Moreover, neovascularization appeared and was extensive. The Trolox equivalent antioxidant capacities and Trolox equivalent antioxidant capacity levels of heart tissue were significantly decreased and levels of malondialdehyde were found to be elevated in STZ rats whereas they were improved in the STZ + GB group. Increased expression levels of VEGF, TGF-β1, TNF-α, and IL-1β proteins in heart tissues were observed in the STZ group. The inflammation cytokine levels were decreased in the STZ + GB group. Conclusions: These results revealed that glabridin was able to reduce LAD and heart tissue damage. Glabridin can be an antioxidant and can reduce the pathology of the LAD in terms of reducing inflammation and restoring condition of the LAD. It would be beneficial in examining the role of glabridin as a therapeutic aim in diabetes treatment research in coronary artery disease.

Small blood vessels in the eyes are more susceptible to injury, which can lead to complications. However, since diabetic retinopathy is often a serious clinical condition, most of this study focuses on the vascular system of the choroid. As part of this study, we looked at how gymnemic acid (from Gymnema sylvestre ) and glabridin (from Glycyrrhiza glabra , or licorice) might help diabetic rats’ choroid thickness and blood vessels. The aim of this study was to explore the effects of glabridin and gymnemic acid on the structural changes of the choroidal layer as well as the expression of VEGF and CD31 in diabetic rats. The animals were separated into five groups: the control group (Control), the diabetic group (DM), the diabetic rats treated with glabridin 40 mg/kg body weight (DM+GB), the diabetic rats treated with gymnemic acid 400 mg/kg body weight (DM+GM), and the diabetic rats treated with glyburide 4 mg/kg body weight (DM+GR). There was an increase in the thickness of both the choroid layer and the wall of the arteries in the diabetic group. A decrease in vascularity and choroidal neovascularization was found in DM rats. After eight weeks of experimentation, the choroidal thickness increased and the walls of choroid arteries, as well as the diameter of the lumen of choroid arteries in the DM+GB, DM+GM, and DM+GR groups, were wider. The expression of VEGF and CD31 was lower compared to the DM group. According to the findings of the current research, glabridin and gymnemic acid can reduce the damage to the choroid, which is a factor that can sometimes result in vision loss.
