After exposure to cold stress, animals enhance the production of beige adipocytes and expedite thermogenesis, leading to improved metabolic health. Although brown adipose tissue in rodents is primarily induced by β3-adrenergic receptor (ADRB3) stimulation, the activation of major β-adrenergic receptors (ADRBs) in pigs has been a topic of debate. To address this, we developed overexpression vectors for ADRB1, ADRB2, and ADRB3 and silenced the expression of these receptors to observe their effects on the adipogenic differentiation stages of porcine preadipocytes. Our investigation revealed that cold stress triggers the transformation of subcutaneous white adipose tissue to beige adipose tissue in pigs by modulating adrenergic receptor levels. Meanwhile, we found that ADRB3 promotes the transformation of white adipocytes into beige adipocytes. Notably, ADRB3 enhances the expression of beige adipose tissue marker genes, consequently influencing cellular respiration and metabolism by regulating lipolysis and mitochondrial expression. Therefore, ADRB3 may serve as a pivotal gene in animal husbandry and contribute to the improvement of cold intolerance in piglets.

Sweet clover (Melilotus albus) is a high-quality leguminous forage grass with salinity tolerance, drought tolerance, and cold hardiness. We selected four varieties of sweet clover with different sensitivities (061898, 061930, No. 55 white flower, and Ningxia white flower) and analyzed the effects of different concentrations of three sodium salts (Na2CO3, NaHCO3, and NaCl) on their physiology and biochemistry responses. Growth and development indexes (such as germination rate, root length, shoot length), chlorophyll content, osmotic regulators (proline, soluble sugar), malondialdehyde (MDA), superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were determined under saline–alkali stress. Seed germination and seedling growth of all four clover species were significantly inhibited under saline–alkali stress. During germination, seed germination rate, root length, and shoot length decreased with increasing saline and alkaline concentration. Under saline–alkali stress, chlorophyll content tended to increase and then decrease, cell damage and death increased, and malondialdehyde, soluble sugar, and proline content tended to increase and then decrease. Moreover, the activities of SOD, POD, and CAT all increased and then decreased. Under Na2CO3 stress, the decrease in chlorophyll content of the resistant variety 061898 was less than in the sensitive Ningxia white flower variety. As the concentrations of Na2CO3, NaHCO3, and NaCl increased, the maximum photochemical efficiency of PSII was significantly affected. The resistant 061898 is capable of maintaining higher photosynthetic efficiency. Furthermore, under treatments with the three kinds of saline–alkali solutions, cell damage and death for Ningxia white flower were greater than in 061898. For 061898, the increases in soluble sugar and proline content were greater and the increase in malondialdehyde content was less, while the antioxidant enzyme activities were higher than those in Ningxia white flower. All four sweet clover varieties had higher stress resistance with neutral than with alkaline salts. When stressed by medium to high saline–alkali concentrations, sweet clover seedlings had increased osmotic substance content, enhanced antioxidant enzyme activity, and regulated physiological metabolism. Additionally, sweet clovers regulated the expression of WRKY33, GH3, CYCD3, OXI1, MKK2, MYC2, JAZ, COI1, PYL, PP2C, TGA, and MPK3 to adapt to the saline–alkali environment and improve saline–alkali tolerance. Our analysis of the sweet clover salinity tolerance mechanism contributes to its further use and is of significant importance for addressing land salinization and promoting sustainable agricultural and pastoral practices in China.

Alfalfa (Medicago sativa L.), a perennial leguminous herb, exhibits robust cold and saline-alkali tolerance. In this study, two alfalfa cultivars, LJ and 218TR, were treated with saline-alkali stress, cold stress, and combined saline-alkali-cold stress , and p henotype, physiology , key metaboli te and stress-responsive genes were analyzed . The results showed m alondialdehyde, soluble sugar, proline content, and the activities of phenylalanine ammonia-lyase (PAL), superoxide dismutase, catalase, and peroxidase initially increased under individual stresses but declined after combined stress . T he maximum photochemical efficiency of photosystem II and chlorophyll content declined under individual and combined stresses. The staining of leaves revealed that combined stress induced significantly higher cell mortality and accumulation of superoxide anio n compared to individual stresses. LJ exhibited superior resistant to saline-alkali, cold, and combined stress compared to 218TR . Metabolite analysis showed salicylic acid (SA) in two alfalfa wa s the most responsive metabolite to combined stress. The isochorismate synthase (ICS) and PAL as critical genes for SA biosynthesis were up-regulated in expression under single or combined stress, and promote d SA accumulation , thereby improving alfalfa resilience to combined saline-alkali-cold stress. This study elucidates the physiological and molecular mechanisms underlying alfalfa’s response to combined saline-alkali and cold stress, providing a theoretical basis for breeding stress-tolerant cultivars.

Cold exposure is a major physiological stimulus that promotes adipose tissue browning and enhances thermogenic capacity. As a cold-tolerant breed, Min pigs may rely on distinct adaptive mechanisms in adipose tissues, but how different adipose depots respond to cold stress remains unclear. This study systematically examines how cold exposure affects adipose tissue morphology, thermogenic gene expression, and the differentiation potential of preadipocytes derived from four adipose depots—cervical, axillary, perirenal, and inguinal—in Min pigs. Adipose tissues from the four depots were collected and analyzed using infrared thermography, histological staining, scanning electron microscopy, real-time quantitative PCR, Western blotting and in vitro differentiation assays. The results demonstrated that cold stress induced depot-specific remodeling patterns. Infrared thermography showed that the subcutaneous adipose depots including inguinal and axillary depots maintained relatively higher surface temperatures following cold exposure. Histological and ultrastructural analyses revealed typical beige-like characteristics in inguinal depots, including the accumulation of small lipid droplets, whereas changes in other depots were minimal. Molecular analyses further confirmed this heterogeneity: inguinal depots exhibited the strongest beiging response, characterized by significant upregulation of key thermogenic markers (PGC1α, UCP3, and DIO2) and lipolytic genes (HSL and ATGL) at the mRNA and/or protein levels. In contrast, visceral adipose depots showed weak or suppressed thermogenic activation. In vitro differentiation assays demonstrated that preadipocytes isolated from inguinal depot had the strongest beiging potential, forming tightly packed small lipid droplets and markedly increasing the expression of beige adipose-related genes upon induction. In conclusion, this study demonstrates that cold stress induces distinct beige remodeling across adipose depots in Min pigs, with the inguinal depot exhibiting the strongest thermogenic response and differentiation potential at morphological, molecular, and cellular levels. These findings identify the inguinal depot as a key site for adaptive thermogenesis and provide novel insights into adipose tissue remodeling and environmental adaptation in large mammals.

Heavy metal contamination is a serious environmental problem worldwide, with substantial negative ecological and economic effects. Serine hydroxymethyltransferase (SHMT) is a key metabolic and photorespiratory enzyme in plant cells, and it is also involved in stress responses. In this study, LcSHMT4 was isolated from sheepgrass (Leymus chinensis (Trin.) Tzvel) after transcriptome sequence analysis. The transcript levels of LcSHMT4 in sheepgrass seedlings increased under Cd and Mn stresses, and subcellular localization analysis in tobacco leaves revealed that its encoded protein localizes at the mitochondria. Transgenic yeast and rice lines overexpressing LcSHMT4 showed increased tolerance to Cd and Mn, compared with that of their controls. In addition, compared with the control, transgenic rice overexpressing LcSHMT4 accumulated more Cd and Mn in brown rice grains. The transcript levels of genes encoding Cd or Mn transporters were increased in the LcSHMT4-overexpressing transgenic rice lines. We speculate that LcSHMT4 may enhance Cd and Mn tolerance by increasing the activities of antioxidant enzymes and the glutathione content and increase heavy metal accumulation by inducing the expression of genes encoding transporters. These results highlight useful genetic resources and provide a theoretical basis for further research on heavy metal tolerance and the phytoremediation of heavy-metal-contaminated soil.
