Background/Objectives: Desiccation profoundly influences the distribution and abundance of intertidal seaweeds, necessitating robust molecular adaptations. Sargassum muticum is a brown seaweed inhabiting intertidal rocky substrates. During low tides, this species undergoes periodic aerial exposure. Such environmental conditions necessitate robust physiological mechanisms to mitigate desiccation stress. Yet, the molecular basis of this adaptation remains poorly understood. Methods: To investigate desiccation-responsive genes and elucidate the underlying mechanisms of adaptation, we exposed S. muticum to 6 h of controlled desiccation stress in sterilized ceramic trays, simulating natural tidal conditions, and performed comparative transcriptome analysis using RNA-seq on the Illumina NovaSeq 6000 platform. Results: High-quality sequencing identified 66,192 unigenes, with 1990 differentially expressed genes (1399 upregulated and 591 downregulated). These differentially expressed genes (DEGs) were categorized into regulatory genes—including mitogen-activated protein kinase (MAPK), calmodulin, elongation factor, and serine/threonine-protein kinase—and functional genes, such as heat shock protein family members (HSP20, HSP40, and HSP70), tubulin (TUBA and TUBB), and endoplasmic reticulum homeostasis-related genes (protein disulfide-isomerase A6, calreticulin, and calnexin). Gene Ontology (GO) enrichment highlighted upregulated DEGs in metabolic processes like glutathione metabolism, critical for oxidative stress mitigation, while downregulated genes were linked to transport functions, such as ammonium transport, suggesting reduced nutrient uptake during dehydration. KEGG pathway analysis revealed significant enrichment in “protein processing in endoplasmic reticulum” and “MAPK signaling pathway-plant”, implicating endoplasmic reticulum stress response and conserved signaling cascades in desiccation adaptation. Validation via qRT-PCR confirmed consistent expression trends for key genes, reinforcing the reliability of transcriptomic data. Conclusions: These findings suggest that S. muticum undergoes extensive biological adjustments to mitigate desiccation stress, highlighting candidate pathways for future investigations into recovery and tolerance mechanisms.

The black rockfish Sebastes schlegelii is an important fishery species in Japan, South Korea, and China. Overfishing has severely depleted the natural resources of S. schlegelii in recent years, leading to the initiation of programs aimed at enhancing fish stock. However, the genetic structure of northern populations remains elusive, posing challenges in collecting and preserving germplasm resources. In this study, a total of 191 S. schlegelii individuals from seven populations, including one cultured population (Changdao: CDYZ) and six wild populations (Lianyungang: LYG; Qingdao: QD; Weihai: WH; Changdao: CDYS; Beidaihe: BDH) sequenced by 2b-RAD method and their population genetics was analyzed using 27,064 SNPs obtained. The results indicated low genetic diversity in both wild and cultured populations ( PIC < 0.25, Ho : 0.174-0.273, He : 0.173-0.234), with the cultured population exhibiting higher diversity than the wild ones. Moderate genetic differentiation existed between the cultured population and six wild populations (0.05 < Fst < 0.25). However, the genetic differentiation was lower among the other wild populations ( Fst < 0.05). The analyses of population genetic structure indicated that the wild populations clustered together first and finally with the cultured population CDYZ, which clustered separately except for one individual. Therefore, scientific breeding programs and germplasm conservation can be important in stabilizing genetic structure. In addition, gene flow among the seven populations was high ( Nm > 1). This study provides a theoretical basis for conserving and rationalizing germplasm resources for S. schlegelii .

This study examined the survival rates of Anadara broughtonii and Chlamys farreri with varying heights throughout winter and summer aquaculture periods under varying ratios of mixed-culture conditions in marine environments. The aim was to assess the viability and economic advantages of mixed-culture practices among different bivalve species. Results indicate that the winter (from December to February of the following year) mortality rate for A. broughtonii in mixed culture ranges from 12.04% to 16.85%, markedly lower than in single-species cultures. Mixed-culture A. broughtonii measuring 2.5–4.0 cm exhibit a significantly reduced summer (from July to September) mortality rate compared to those in single-species cultures (p < 0.05). No significant mortality difference was observed between the monoculture and polyculture of C. farreri. Growth rates of C. farreri and A. broughtonii showed no significant differences between monoculture and polyculture conditions. The optimal size for combining two different species of bivalves is 2.5 cm, with a 1:1 ratio being the most effective. An equal number of seedlings were cultivated across various seawater aquaculture systems for a period of one year. The survival rates of monoculture seedlings of Anadara broughtonii and Chlamys farreri were 60.34% and 86.55%, respectively. In contrast, polyculture seedlings exhibited higher survival rates after one year of mariculture, with Anadara broughtonii at 73.36% and Chlamys farreri at 89.74%. The polyculture of A. broughtonii and C. farreri in suspension cages demonstrates a favorable input–output ratio of 1:2.02. This approach effectively reduces winter and summer mortality rates of A. broughtonii, representing an efficient new method for marine bivalve aquaculture.
