Crop productivity is greatly affected by soil salinity; therefore, improvement in salinity tolerance of crops is a major goal in salt-tolerant breeding. The Salt Overly Sensitive (SOS) signal-transduction pathway plays a key role in ion homeostasis and salt tolerance in plants. Here, we report that overexpression of Arabidopsis thaliana SOS1+SOS2+SOS3 genes enhanced salt tolerance in tall fescue. The transgenic plants displayed superior growth and accumulated less Na+ and more K+ in roots after 350 mM NaCl treatment. Moreover, Na+ enflux, K+ influx, and Ca2+ influx were higher in the transgenic plants than in the wild-type plants. The activities of the enzyme superoxide dismutase, peroxidase, catalase, and proline content in the transgenic plants were significantly increased; however, the malondialdehyde content decreased in transgenic plants compared to the controls. These results suggested that co-expression of A. thaliana SOS1+SOS2+SOS3 genes enhanced the salt tolerance in transgenic tall fescue.

Abiotic stressors such as salt stress restrict plant development and output, which lowers agricultural profitability. In this study, alfalfa (Medicago sativa L.) varieties with different levels of salt tolerance were examined using high-throughput RNA sequencing (RNA-Seq) and Tandem Mass Tags (TMT) technologies to study the reactions of the root systems to salt stress, from transcriptomics and proteomics perspectives. The varieties Atlantic (AT) and Zhongmu-1 (ZM-1) were selected and evaluated after 2 h and 6 h of treatment with 150 mM NaCl. The results showed that under salt stress for 2 h, 1810 differentially expressed genes (DEGs) and 160 differentially expressed proteins (DEPs) in AT were screened, while 9341 DEGs and 193 DEPs were screened in ZM-1. Under salt stress for 6 h, 7536 DEGs and 118 DEPs were screened in AT, while 11,754 DEGs and 190 DEPs were screened in ZM-1. Functional annotation and pathway enrichment analyses indicated that the DEGS and DEPs were mainly involved in the glutathione metabolism, biosynthesis of secondary metabolites, glycolysis/gluconeogenesis, carbon fixation in photosynthetic organisms, and photosynthesis pathways. A series of genes related to salt tolerance were also identified, including GSTL3 and GSTU3 of the GST gene family, PER5 and PER10, of the PER gene family, and proteins such as APR and COMT, which are involved in biosynthesis of secondary metabolites. This study provides insights into salt resistance mechanisms in plants, and the related genes and metabolic pathways identified may be helpful for alfalfa breeding in the future.

CRISPR/Cas9 is a valuable tool and has been extensively employed to perform gene editing in plants. However, CRISPR/Cas9 has not been successfully used in spinach, an important leafy vegetable crop. Here, we established a CRISPR/Cas9-based gene-editing system for spinach hairy roots and edited two cellulose synthase-like D (CSLD) genes (SoCSLD2 and SoCSLD3) that were involved in root-hair formation of spinach hairy roots. Four mutation types (i.e., replacement, insertion, deletion, and combined mutations) were observed, among which the replacement accounted for the vast majority (about 64.1%). Mutation rate differed largely among different targets. Seven homozygous/bi-allelic and eight heterozygous/chimeric mutants of SoCSLD2 were obtained from 15 independent transgenic hairy root lines. All of the seven homozygous/bi-allelic mutant lines displayed bulking and short root hairs, which resembled the characteristics of Arabidopsis atcsld2 mutants. The transcriptomic analysis further revealed that multiple gene expressions for cell-wall modulation and membrane trafficking were disturbed, which might result in the inhibition of root hair growth in socsld2 mutants. This indicates that SoCSLD2 was successfully knocked out in spinach root hairs using the CRISPR/Cas9-based gene editing system.

Oat (Avena sativa L.) is an important forage crop widely used in animal husbandry. However, the greenhouse effect, which leads to increasing global temperatures, extreme water scarcity, and more frequent drought events, also creates abiotic stress that inhibits oat growth. Drought stress strongly affects the yield and quality of forage oats, hindering the selection, promotion, and utilization of drought-resistant cultivars. This study investigated alterations in the growth and physiological traits of diverse oat cultivars under drought stress for varying durations. We comprehensively assessed the drought resistance capabilities of each variety. Forty oat cultivars were subjected to drought stress starting from plant growth up to the two-leaf stage a pot-based water withholding method. The stress durations were 0 d, 7 d, and 14 d. Compared with those of the control, the key physiological parameters of the test cultivar decreased with increasing drought stress duration. These factors increased the maximum photochemical quantum yield (Fv/Fm), PS II quantum efficiency (Fv/Fo), soil plant analysis development (SPAD) value, net photosynthetic rate (Pn), transpiration rate (Tr), and stomatal conductance (Gs). Conversely, the malondialdehyde (MDA) and proline (Pro) contents increased. Antioxidant enzyme activity initially increased but subsequently decreased. Changes in osmoregulatory substance content and the modulation of antioxidant enzyme activity are key components of drought resistance mechanisms. Therefore, Fv/Fo, Pro, Tr, Gs, and Pn have emerged as reliable parameters for assessing drought resistance in forage oat seedlings. When assessing seedling drought resistance using biochemical parameters such as photosynthesis, a comprehensive analysis combining multiple indicators and methods is essential. This study provides a theoretical basis for screening drought-resistant oat cultivars and for high-yield cultivation practices.
